A novel electrochemical immunosensor for sensitive detection of cancer biomarker α-fetoprotein (AFP) is described that uses a graphene sheet sensor platform and functionalized carbon nanospheres (CNSs) labeled with horseradish peroxidase-secondary antibodies (HRP-Ab2). Greatly enhanced sensitivity for the cancer biomarker is based on a dual signal amplification strategy: first, the synthesized CNSs yielded a homogeneous and narrow size distribution, which allowed several binding events of HRP-Ab2 on each nanosphere. Enhanced sensitivity was achieved by introducing the multibioconjugates of HRP-Ab2-CNSs onto the electrode surface through “sandwich” immunoreactions. Second, functionalized graphene sheets used for the biosensor platform increased the surface area to capture a large amount of primary antibodies (Ab1), thus amplifying the detection response. Sensitive Immunosensor for Cancer Biomarker Based on Dual Signal Amplification Strategy of Graphene Sheets and Multienzyme Functionalized Carbon Nanospheres

Attachment of biomolecules on gold, silicon or glass surfaces has direct implications for the development of novel biosensors in the context of nanoscale detection of pathogens and other metabolites related to issues of human health. In this critical review, we have highlighted the current developments in various techniques of immobilization of biomolecules, specifically biological macromolecules on surfaces through the modification of a functional self-assembled monolayer. The utility of such immobilized biomolecules in the area of biosensing in nanoscale has been surveyed. Merits and demerits of some of the methods with reference to sensitivity of detection and practical use have been discussed (221 references).

Immobilization of bio-macromolecules on self-assembled monolayers: methods and sensor applications

The cardiovascular disease (CVD) is considered as a major threat to global health. Therefore, there is a growing demand for a range of portable, rapid and low cost biosensing devices for the detection of CVD. Biosensors can play an important role in the early diagnosis of CVD without having to rely on hospital visits where expensive and time-consuming laboratory tests are recommended. Over the last decade, many biosensors have been developed to detect a wide range of cardiac marker to reduce the costs for healthcare. One of the major challenges is to find a way of predicting the risk that an individual can suffer from CVD. There has been considerable interest in finding diagnostic and prognostic biomarkers that can be detected in blood and predict CVD risk. Of these, C-reactive protein (CRP) is the best known biomarker followed by cardiac troponin I or T (cTnI/T), myoglobin, lipoprotein-associated phospholipase A(2), interlukin-6 (IL-6), interlukin-1 (IL-1), low-density lipoprotein (LDL), myeloperoxidase (MPO) and tumor necrosis factor alpha (TNF-α) has been used to predict cardiovascular events. This review provides an overview of the available biosensor platforms for the detection of various CVD markers and considerations of future prospects for the technology are addressed.

Biosensors for cardiac biomarkers detection: A review

This review examines the current state of the art lab-on-a-chip and microfluidic based biosensor technologies used in the detection of cardiac biomarkers. The determination and quantification of blood based, cardiac biomarkers are crucial in the triage and management of a range of cardiac related conditions, where time delay has a major impact on short and longer-term outcomes of a patient. The design and manufacturing of biomarker detection systems are multi-disciplinary in nature and require researchers to have knowledge of both life sciences and engineering for the full potential of this field to be realised. This review will therefore provide a comprehensive overview of chip based immunosensing technology as applied to cardiac biomarker detection, while discussing the potential suitability and limitations of each configuration for incorporation within a clinical diagnostics device suitable for point-of-care applications.

Lab-on-a-chip based immunosensor principles and technologies for the detection of cardiac biomarkers: a review

Biosensors consist of a biocatalyst that can detect a biological element and a transducer which can convert the combination event of the biocatalyst and the biological element into a detectable parameter. The biocatalyst can be biomolecules like, enzyme, DNA, RNA, metabolites, cells, oligonucleotides etc., and the transducers can be electrochemical, optical, piezoelectric, acoustics, calorimetric etc. In disease diagnostics biosensors utilizing immobilized cells, enzymes, nucleic acids have come into the field in recent years. Nanobiosensors exploiting the ultrasmall size and unique properties have also been used for engineering disease diagnostic biosensors. The use of biosensors can rapidly assess the health status, onset of the disease and its progression and can help to plan treatment for many diseases with the aid of multidisciplinary combination of chemistry, medical science and nanotechnology. The devices are cost effective, highly sensitive, rapid, user friendly and can be produced in bulk for human use. This review focuses on the different biosensors for the diagnosis of three major diseases like diabetes, cardiovascular disease and cancer.

A simple microfluidic immunoassay card was developed based on polystyrene (PS) substrate for the detection of horse IgG, an inexpensive model analyte using fluorescence microscope. The primary antibody was captured onto the PS based on covalent bonding via a self-assembled monolayer (SAM) of thiol to pattern the surface chemistry on a gold-coated PS. The immunosensor chip layers were fabricated from sheets by CO2 laser ablation. The functionalized PS surfaces after each step were characterized by contact angle measurement, X-ray photoelectron spectroscopy (XPS), and atomic force microscopy (AFM). After the antibody–antigen interaction as a sandwich immunoassay with a fluorescein isothiocyanate (FITC)-conjugated secondary antibody, the intensity of fluorescence was measured on-chip to determine the concentration of the target analyte. The present immunosensor chip showed a linear response range for horse IgG between 1 μg/ml and 80 μg/ml (r = 0.971, n = 3). The detection limit was found to be 0.71 μg/ml. The developed microfluidic system can be extended for various applications including medical diagnostics, microarray detection and observing protein–protein interactions.
