Fused Deposition Modelling: Current Status, Methodology, Applications and Future Prospects

Polyhydroxyalkanoates (PHA) are bio-based microbial biopolyesters; their stiffness, elasticity, crystallinity and degradability are tunable by the monomeric composition, selection of microbial production strain, substrates, process parameters during production, and post-synthetic processing; they display biological alternatives for diverse technomers of petrochemical origin. This, together with the fact that their monomeric and oligomeric in vivo degradation products do not exert any toxic or elsewhere negative effect to living cells or tissue of humans or animals, makes them highly stimulating for various applications in the medical field. This article provides an overview of PHA application in the therapeutic, surgical and tissue engineering area, and reviews strategies to produce PHA at purity levels high enough to be used in vivo. Tested applications of differently composed PHA and advanced follow-up products as carrier materials for controlled in vivo release of anti-cancer drugs or antibiotics, as scaffolds for tissue engineering, as guidance conduits for nerve repair or as enhanced sutures, implants or meshes are discussed from both a biotechnological and a material-scientific perspective. The article also describes the use of traditional processing techniques for production of PHA-based medical devices, such as melt-spinning, melt extrusion, or solvent evaporation, and emerging processing techniques like 3D-printing, computer-aided wet-spinning, laser perforation, and electrospinning.

Biodegradable and Biocompatible Polyhydroxy-alkanoates (PHA): Auspicious Microbial Macromolecules for Pharmaceutical and Therapeutic Applications.

The increasing demand for environmental protection has led to the rapid development of greener and biodegradable polymers, whose creation provided new challenges and opportunities for the advancement of nanomaterial science. Biodegradable polymer materials and even nanofillers (e.g., natural fibers) are important because of their application in greener industries. Polymers that can be degraded naturally play an important role in solving public hazards of polymer materials and maintaining ecological balance. The inherent shortcomings of some biodegradable polymers such as weak mechanical properties, narrow processing windows, and low electrical and thermal properties can be overcome by composites reinforced with various nanofillers. These biodegradable polymer composites have wide-ranging applications in different areas based on their large surface area and greater aspect ratio. Moreover, the polymer composites that exploit the synergistic effect between the nanofiller and the biodegradable polymer matrix can lead to enhanced properties while still meeting the environmental requirement. In this paper, a broad review on recent advances in the research and development of nanofiller reinforced biodegradable polymer composites that are used in various applications, including electronics, packing materials, and biomedical uses, is presented. We further present information about different kinds of nanofillers, biodegradable polymer matrixes, and their composites with specific concern to our daily applications.

Nanofiller Reinforced Biodegradable PLA/PHA Composites: Current Status and Future Trends

Polyhydroxyalkanoates (PHA) are microbial biopolyesters utilized as “green plastics”. Their production under controlled conditions resorts to bioreactors operated in different modes. Because PHA biosynthesis constitutes a multiphase process, both feeding strategy and bioreactor operation mode need smart adaptation. Traditional PHA production setups based on batch, repeated batch, fed-batch or cyclic fed-batch processes are often limited in productivity, or display insufficient controllability of polyester composition. For highly diluted substrate streams like is the case of (agro) industrial waste streams, fed-batch enhanced by cell recycling has recently been reported as a viable tool to increase volumetric productivity. As an emerging trend, continuous fermentation processes in single-, two- and multi-stage setups are reported, which bring the kinetics of both microbial growth and PHA accumulation into agreement with process engineering and allow tailoring PHA’s molecular structure. Moreover, we currently witness an increasing number of CO2-based PHA production processes using cyanobacteria; these light-driven processes resort to photobioreactors similar to those used for microalgae cultivation and can be operated both discontinuously and continuously. This development is parallel to the emerging use of methane and syngas as abundantly available gaseous substrates, which also calls for bioreactor systems with optimized gas transfer. The review sheds light on the challenges of diverse PHA production processes in different bioreactor types and operational regimes using miscellaneous microbial production strains such as extremophilic Archaea, chemoheterotrophic eubacteria and phototrophic cyanobacteria. Particular emphasis is dedicated to the limitations and promises of different bioreactor–strain combinations and to efforts devoted to upscaling these processes to industrially relevant scales.

A Review on Established and Emerging Fermentation Schemes for Microbial Production of Polyhydroxyalkanoate (PHA) Biopolyesters

Being designated to protect other tissues, skin is the first and largest human body organ to be injured and for this reason, it is accredited with a high capacity for self-repairing. However, in the case of profound lesions or large surface loss, the natural wound healing process may be ineffective or insufficient, leading to detrimental and painful conditions that require repair adjuvants and tissue substitutes. In addition to the conventional wound care options, biodegradable polymers, both synthetic and biologic origin, are gaining increased importance for their high biocompatibility, biodegradation, and bioactive properties, such as antimicrobial, immunomodulatory, cell proliferative, and angiogenic. To create a microenvironment suitable for the healing process, a key property is the ability of a polymer to be spun into submicrometric fibers (e.g., via electrospinning), since they mimic the fibrous extracellular matrix and can support neo- tissue growth. A number of biodegradable polymers used in the biomedical sector comply with the definition of bio-based polymers (known also as biopolymers), which are recently being used in other industrial sectors for reducing the material and energy impact on the environment, as they are derived from renewable biological resources. In this review, after a description of the fundamental concepts of wound healing, with emphasis on advanced wound dressings, the recent developments of bio-based natural and synthetic electrospun structures for efficient wound healing applications are highlighted and discussed. This review aims to improve awareness on the use of bio-based polymers in medical devices.
