Actomyosin-Mediated Cellular Tension Drives Increased Tissue Stiffness and β-Catenin Activation to Induce Epidermal Hyperplasia and Tumor Growth
Michael S. Samuel,Jose Lopez,Ewan J. McGhee,Daniel R. Croft,David Strachan,Paul Timpson,June Munro,Ewald Schroder,Jing Zhou,Valerie G. Brunton,Nick Barker,Hans Clevers,Owen J. Sansom,Kurt I. Anderson,Valerie M. Weaver,Michael F. Olson +15 more
TL;DR: Tumor number, growth, and progression were increased by ROCK activation, while ROCK blockade was inhibitory, implicating actomyosin-mediated cellular tension and consequent collagen deposition as significant tumor promoters.

Role of the alternative INK4A proteins in human keratinocyte senescence: evidence for the specific inactivation of p16INK4A upon immortalization.
TL;DR: The INK4A locus on human chromosome 9p21 encodes two genes that have been implicated in replicative senescence and tumor suppression, p16ink4A and p14ARF, and there was evidence for the inactivation of p16INK4A, independent of p14 ARF, in 6 of 10 lines that lacked large homozygous deletions.

Polarized Cell Motility Induces Hydrogen Peroxide to Inhibit Cofilin via Cysteine Oxidation
Jenifer M. Cameron,Mads Gabrielsen,Ya Hua Chim,June Munro,Ewan J. McGhee,David Sumpton,Philip Eaton,Kurt I. Anderson,Huabing Yin,Michael F. Olson +9 more
TL;DR: It is shown that motile cells generate H2O2 at membranes and cell protrusions and that H2 O2 inhibits cofilin activity through oxidation of cysteines 139 and 147 (C147).

Migration through physical constraints is enabled by MAPK-induced cell softening via actin cytoskeleton re-organization.
Dominika A. Rudzka,Giulia Spennati,David J. McGarry,Ya-Hua Chim,Matthew Neilson,Aleksandra Ptak,June Munro,Gabriela Kalna,Ann Hedley,Daniela Moralli,Catherine M. Green,Susan M. Mason,Karen Blyth,Margaret Mullin,Huabing Yin,Michael F. Olson +15 more
TL;DR: Selecting for tumour cells that efficiently pass through narrow diameter microporous membranes reveals a prominent role for MAPK signalling in regulating cell elasticity.
