Volatile organic compounds (VOCs) of walnut oil (WO) samples obtained from 5 ripening stages were analyzed by headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) and HS-solid phase microextraction-GC-mass spectrometry (HS-SPME-GC-MS). A total of 75 VOCs were identified in WO, of which 24 VOCs were found to be the key aroma-active compounds for WO by using odor activity values (OAVs) analysis. Based on chemometrics methods, flavor of WO samples can be characterized into three categories, i.e., early, mid-, and late stages. WO from early ripening stage had stronger green and sweet odor due to 1,8-cineole (OAV 280) and ethanol (OAV 134.5). While nonanal (OAV 181.82), (E)-2-octenol (OAV 160), and hexanal (OAV 103.78) were sources of intense fatty and oily odor in mid-ripening stage. For WO of later ripening stage, the flavor was affected by nonanal (OAV 192.28), 1-heptanol (OAV 150), heptanal (OAV 71.11) and some organic acids.

Impact of ripening periods on the key volatile compounds of Cheddar cheese evaluated by sensory evaluation, instrumental analysis and chemometrics method

Abstract Background Juglans sigillata L. (walnut) has a high economic value for nuts and wood and has been widely grown and eaten around the world. Light plays an important role in regulating the development of the walnut embryo and promoting nucleolus enlargement, which is one of the factors affecting the yield and quality of walnut. However, little is known about the effect of light on the growth and quality of walnuts. Studies have shown that far red prolonged hypocotyl 3 (FHY3) and far red damaged response (FAR1) play important roles in plant growth, light response, and resistance. Therefore, FHY3/FAR1 genes were identified in walnuts on a genome-wide basis during their growth and development to reveal the potential regulation mechanisms involved in walnut kernel growth and development. Results In the present study, a total of 61 FHY3/FAR1 gene family members in walnuts have been identified, ranging in length from 117 aa to 895 aa. These gene family members have FHY3 or FAR1 conserved domains, which are unevenly distributed on the 15 chromosomes (Chr) of the walnut (except for the Chr16). All 61 FHY3/FAR1 genes were divided into five subclasses (I, II, III, IV, and V) by phylogenetic tree analysis. The results indicated that FHY3/FAR1 genes in the same subclasses with similar structures might be involved in regulating the growth and development of walnut. The gene expression profiles were analyzed in different walnut kernel varieties (Q, T, and F). The result showed that some FHY3/FAR1 genes might be involved in the regulation of walnut kernel ripening and seed coat color formation. Seven genes ( OF07056-RA , OF09665-RA , OF24282-RA , OF26012-RA , OF28029-RA , OF28030-RA , and OF08124-RA ) were predicted to be associated with flavonoid biosynthetic gene regulation cis-acting elements in promoter sequences. RT-PCR was used to verify the expression levels of candidate genes during the development and color change of walnut kernels. In addition, light responsiveness and MeJA responsiveness are important promoter regulatory elements in the FHY3/FAR1 gene family, which are potentially involved in the light response, growth, and development of walnut plants. Conclusion The results of this study provide a valuable reference for supplementing the genomic sequencing results of walnut, and pave the way for further research on the FHY3/FAR1 gene function of walnut. 

Genome-wide identification and molecular expression profile analysis of FHY3/FAR1 gene family in walnut (Juglans sigillata L.) development

The color of endopleura is a vital factor in determining the economic value and aesthetics appeal of nut. Walnuts (Juglans) are a key source of edible nuts, high in proteins, amino acids, lipids, carbohydrates. Walnut had a variety endopleura color as yellow, red, and purple. However, the regulation of walnut endopleura color remains little known.To understand the process of coloration in endopleura, we performed the integrative analysis of transcriptomes and metabolomes at two developmental stages of walnut endopleura. We obtained total of 4,950 differentially expressed genes (DEGs) and 794 metabolites from walnut endopleura, which are involved in flavonoid and phenolic biosynthesis pathways. The enrichment analysis revealed that the cinnamic acid, coniferyl alcohol, naringenin, and naringenin-7-O-glucoside were important metabolites in the development process of walnut endopleura. Transcriptome and metabolome analyses revealed that the DEGs and differentially regulated metabolites (DRMs) were significantly enriched in flavonoid biosynthesis and phenolic metabolic pathways. Through co-expression analysis, CHS (chalcone synthase), CHI (chalcone isomerase), CCR (cinnamoyl CoA reductase), CAD (cinnamyl alcohol dehydrogenase), COMT (catechol-Omethyl transferase), and 4CL (4-coumaroyl: CoA-ligase) may be the key genes that potentially regulate walnut endopleura color in flavonoid biosynthesis and phenolic metabolic pathways.This study illuminates the metabolic pathways and candidate genes that underlie the endopleura coloration in walnuts, lay the foundation for further study and provides insights into controlling nut's colour. 

Integrated metabolomic and transcriptomic dynamic profiles of endopleura coloration during fruit maturation in three walnut cultivars

Polyphenols, abundant in the pellicle of walnuts, confer health benefits to the human body. The intricate relationship between light and phenolic compounds underscores the impact of shading, which can effectively diminish phenolic content and enhance the palatability of food. In this study, we delved into the connection between polyphenol accumulation, related enzyme activities, and gene expression in the pellicle of Juglans sigillata Dode under varying degrees of shading. The results show that different shade intensities can change polyphenols' content and affect the activity of key polyphenols anabolism enzymes and their gene expression. With the increase of the shading intensity, the total phenolics content (TPC) and total flavonoids content (TFC) were lower than the nomal light. In the mature stage, shading 60 % was lowest. 7 individual phenolics were identified in the pellicle, and the content of each varied significantly with the decrease of light intensity. Except for p-coumaric acid and ferulic acid, which were higher than the control at mature stage, all phenolics were lower than the control, but the overall trend was the same as that of the control. The activity of key enzymes in the anabolism of polyphenols was significantly affected after shading. In complete darkness phenylalanine ammonia lyase (PAL) was highest at the kernel-filling stage, shading 20 % of chalcone synthase (CHS) and shading 60 % of flavonol synthase (FLS) responded most strongly at the hard-core stage, while flavanone 3-hydroxylase (F3H) was most active at shading 60 % of kernel-filling stage, which was 2.03, 1.38, 4.67, and 2.08 times that of the control, respectively. The expression levels of key genes showed that the expression level of JsPAL was significantly increased at hard-core stage and mature stage, and the expression of JsPAL was 3.85 times higher than that of the control at the mature stage with 60 % shading. Correlation analysis showed that PAL enzyme activity was highly correlated with TPC, TFC and various individual phenolics with the increase of shading intensity. The content of p-coumaric acid was positively correlated with the activities of 4-coumarate coenzyme A ligase (4CL), F3H and FLS. The increase of shading intensity enhanced the correlation between total phenolics content, total flavonoids content, individual phenolics substances and JsPAL, JsC4H, JsCHS, JsCHI and JsFLS. Overall, PAL was a key enzyme affecting polyphenol accumulation under different shading levels, while at the genetic level, JsPAL, JsC4H, JsCHS, JsCHI and JsFLS were the key genes affecting them. This study will potentially provide more information for light management and polyphenol regulation in walnut cultivation.

Transcriptome analysis of walnut quality formation and color change mechanism of pellicle during walnut development.

HYW (Hong Yi Wu line) heavy oil emulsion in Xinjiang Oilfield (Karamay, China) is a kind of heavy oil with high viscosity and high emulsification. Its viscosity reaches 120,000 mPa·s at 40 °C. The emulsion has no demulsification. Even if the demulsification temperature reaches 90 degrees, the concentration of demulsifier reaches 260 mg/L. In this paper, a new process of thermochemical demulsification of heavy oil after blending is studied. First, SE low-viscosity oil with viscosity of 640 mPa·s and water cut of 90% was selected as blended oil. Study the viscosity of SE line and HYW line at different temperatures after fully blended. The results show that the heavy oil blended model conforms to Bingham model. When the temperature is 40 °C and the content of SE line is 30%, the viscosity is less than 10,000 mPa·s. With the increase of temperature, the viscosity continues to decline. When the temperature exceeds 80 °C, the viscosity is less than 1000 mPa·s. The final design SE line content is 30%, the demulsification temperature is 70 °C, and the demulsifier concentration is 160 mg/L as the best demulsification parameter. The field results show that the demulsification rate of heavy oil in this process reaches more than 90%. This experiment lays a foundation for demulsification of high emulsified crude oil developed by heavy oil in Xinjiang oilfield.
