Technologies including (3D-) (bio-)printing, diesel engines, laser-induced forward transfer, and spray cleaning require optimization and therefore understanding of micrometer-sized droplets impacting at velocities beyond 10 m s−1. However, as yet, this regime has hardly been addressed. Here we present the first time-resolved experimental investigation of microdroplet impact at velocities up to V0 = 50 m s−1, on hydrophilic and -phobic surfaces at frame rates exceeding 107 frames per second. A novel method to determine the 3D-droplet profile at sub-micron resolution at the same frame rates is presented, using the fringe pattern observed from a bottom view. A numerical model, which is validated by the side- and bottom-view measurements, is employed to study the viscous boundary layer inside the droplet and the development of the rim. The spreading dynamics, the maximal spreading diameter, the boundary layer thickness, the rim formation, and the air bubble entrainment are compared to theory and previous experiments. In general, the impact dynamics are equal to millimeter-sized droplet impact for equal Reynolds-, Weber- and Stokes numbers (Re, We, and St, respectively). Using our numerical model, effective scaling laws for the progression of the boundary layer thickness and the rim diameter are provided. The dimensionless boundary layer thickness develops in time (t) according to , and the diameter of the rim develops as , with drop diameter D0 and inertial time scale τ = D0/V0. These scalings differ from previously assumed, but never validated, values. Finally, no splash is observed, at variance with many predictions but in agreement with models including the influence of the surrounding gas. This confirms that the ambient gas properties are key ingredients for splash threshold predictions.

Epidermal healing in burns: autologous keratinocyte transplantation as a standard procedure: update and perspective.

Severe burns in children are conventionally treated with split-thickness skin autografts or epidermal sheets. However, neither early complete healing nor quality of epithelialization is satisfactory. An alternative approach is to graft isolated keratinocytes. We evaluated paediatric foreskin and auricular skin as donor sources, autologous keratinocyte transplantation, and compared the graft efficiency to the in vitro capacities of isolated keratinocytes to divide and reconstitute epidermal tissue. Keratinocytes were isolated from surgical samples by enzymatic digestion. Living cell recovery, in vitro proliferation and epidermal reconstruction capacities were evaluated. Differentiation status was analysed, using qRT-PCR and immunolabelling. Eleven children were grafted with foreskin-derived (boys) or auricular (girls) keratinocyte suspensions dripped onto deep severe burns. The aesthetic and functional quality of epithelialization was monitored in a standardized way. Foreskin keratinocyte graft in male children provides for the re-epithelialization of partial deep severe burns and accelerates wound healing, thus allowing successful wound closure, and improves the quality of scars. In accordance, in vitro studies have revealed a high yield of living keratinocyte recovery from foreskin and their potential in terms of regeneration and differentiation. We report a successful method for grafting paediatric males presenting large severe burns through direct spreading of autologous foreskin keratinocytes. This alternative method is easy to implement, improves the quality of skin and minimizes associated donor site morbidity. In vitro studies have highlighted the potential of foreskin tissue for graft applications and could help in tissue selection with the prospect of grafting burns for girls.

Any disturbance in the balance between these components can impair the healing process. The function of cells and growth factors can be manipulated and facilitated to aid tissue repair. In the current review, the authors focus on the importance of the skin microenvironment, the pathophysiology of various types of burns, mechanisms and factors involved in skin repair and wound healing and regeneration of the skin using tissue engineering approaches.

Isolation and culture of different epidermal and dermal cell types from human scalp suitable for the development of a therapeutical cell spray

Alloplastic temporomandibular joint replacement has been established as a standard technique for end-stage temporomandibular (TMJ) pathologies. Joint replacement when there are extensive mandibular defects remains a challenging clinical problem. Custom-made extended temporomandibular joint replacement is a feasible option but there is limited information about this emerging technique. In this retrospective case series, the authors report the results, surgical techniques, pitfalls and outcome of more than 2 years follow-up with reference to thirteen extended TMJ-replacements (TMJe) including twelve patients. The most common diagnosis was ameloblastoma of the mandibular ramus. The authors suggest a simplified anatomically based single-stage or two-stage regime, with both regimes achieved excellent anatomic reconstruction, facial appearance and function with low surgical morbidity. Custom-made extended temporomandibular joint protheses appear an advanced and reliable solution for reconstruction of combined complex mandibular defects including the temporomandibular joint. If surgical clearance of the pathology can be achieved, a single-stage regime is favoured.
