Periodontal diseases affect millions of people worldwide and can result in tooth loss. Regenerative treatment options for clinical use are thus needed. We aimed at developing new nonwoven-based scaffolds for periodontal tissue engineering. Nonwovens of 16% gelatin/5% hydroxyapatite were produced by electrospinning and in situ glyoxal cross-linking. In a subset of scaffolds, additional porosity was incorporated via extractable polyethylene glycol fibers. Cell colonization and penetration by human mesenchymal stem cells (hMSCs), periodontal ligament fibroblasts (PDLFs), or cocultures of both were visualized by scanning electron microscopy and 4′,6-diamidin-2-phenylindole (DAPI) staining. Metabolic activity was assessed via Alamar Blue® staining. Cell type and differentiation were analyzed by immunocytochemical staining of Oct4, osteopontin, and periostin. The electrospun nonwovens were efficiently populated by both hMSCs and PDLFs, while scaffolds with additional porosity harbored significantly more cells. The metabolic activity was higher for cocultures of hMSCs and PDLFs, or for PDLF-seeded scaffolds. Periostin and osteopontin expression was more pronounced in cocultures of hMSCs and PDLFs, whereas Oct4 staining was limited to hMSCs. These novel in situ-cross-linked electrospun nonwoven scaffolds allow for efficient adhesion and survival of hMSCs and PDLFs. Coordinated expression of differentiation markers was observed, which rendered this platform an interesting candidate for periodontal tissue engineering.

Novel In Situ-Cross-Linked Electrospun Gelatin/Hydroxyapatite Nonwoven Scaffolds Prove Suitable for Periodontal Tissue Engineering

Abstract This study analyzed the prevalence of lectures involving esthetics in the scientific program of Brazilian dental conferences and the gender distribution of speakers. All lectures presented in three dental conferences (Bahia, São Paulo, and Goiás states) held from 2016 to 2020 were evaluated. Three investigators individually divided the lectures according to the specialties recognized by the Brazilian Federal Council of Dentistry (FCD) based on their titles. The lectures were also classified as involving or not esthetics, and the speaker`s gender was recorded. Descriptive statistical analyses were performed, and Chi-square tests assessed possible associations between factors. The words most cited in the titles of the lectures were “esthetic” (13.6%), “dentistry” (9.9%), and “treatment” (8.1%). Oral diseases were barely mentioned in the titles (up to 1.3%). The highest number of lectures was observed for the specialty of Restorative Dentistry (22.3%), followed by Prosthodontics (18.5%). Approximately one-third of lectures involved some aesthetic aspect, but this percentage ranged from 71.9 to 78.6% for the two specialties with more lectures. Regarding the speaker`s gender, the inequity was higher for lectures involving esthetics (81.6% of males) than for topics unrelated to esthetics (66.7%). More male speakers than females were observed for all specialties. The highest gender gap was observed for Pediatric Dentistry with 62.4% male speakers, although only 10.6% of FCD registered specialists were men. In conclusion, the Brazilian dental conferences analyzed seemed to favor offering lectures dealing with esthetic topics and male speakers.

Prevalence of lectures about dental esthetics and female speakers in three Brazilian conferences

Background: The anti-inflammatory, antioxidant, antiviral, antibacterial, antitumor, antiarthritic, and anti-diabetic properties of aloe vera has been well documented in the literature. Considering its antibacterial properties evaluating its benefits as mouth rinse in the oral diseases may facilitate the patients in getting cheaper and safer herbal remedy to clean their oral cavities. Objective:  To evaluate the Antibacterial efficacy of aloevera peel extract against Streptococcus mutans and P. gingivalis. Methodology: It was a preclinical experimental study conducted at Baqai Medical and Dental College Karachi, from February to March 2023. Calculated sample size was n =60. The participants were given an envelope for group randomization. S. mutant samples and P. gingivalis were incubated on appropriate media. The Aloe Vera leaves were washed and gel was removed the peels were shed dried and lastly was grinded to powder form. Ethanol (70%) were used for soaking for 15 days with intermittent shaking. The extract was diluted in distilled water in 1:4 (Extract: Distilled water) concentration. 

Antibacterial efficacy of aloevera peel extract against streptococcus mutans and P. gingivalis

A periodontite consiste em um distúrbio infeccioso e imunoinflamatório multifatorial que leva à destruição tecidual, incluindo o osso alveolar, constituindo um sério problema de saúde pública. A formação do biofilme constitui um dos principais fatores associados ao início e progressão da doença. O tratamento é realizado através de procedimentos mecânicos e químicos que visam minimizar a formação de biofilmes periodontopatogênicos. No entanto, estas estratégias têm contribuído para o aumento da resistência bacteriana e o aparecimento de efeitos colaterais. O atual cenário tem despertado o interesse pelo desenvolvimento de tratamentos novos e eficazes, principalmente envolvendo produtos naturais. Assim, a Psidium guajava é considerada um excelente potencial terapêutico, devido à grande versatilidade da sua ação que inclui atividades anti-inflamatória, antimicrobiana e antioxidante. Diante disso, o presente artigo tem como objetivo desenvolver uma prospecção científica e tecnológica a partir da Psidium guajava com foco nas atividades anti-inflamatórias e antimicrobianas para o tratamento da periodontite em sites de depósitos de patentes e bases de dados para publicações de artigos científicos nacionais e internacionais. Para isso, foi realizado um levantamento nas bases de dados PubMed, Web of ScienceTM e Scientific Electronic Library Online (SciELO) para artigos e European Patent Office (EPO), Instituto Nacional de Propriedade Industrial do Brasil (INPI), Banco de Patentes Latinoamericanas (LATIPAT), United States Patent and Trademark Office (USPTO) e World Intellectual Property Organization (WIPO) para as patentes. A pesquisa foi realizada durante o período de outubro a dezembro de 2023 e foram utilizados os descritores “Psidium guajava” e “periodontitis”, bem como a combinação entre eles e com os termos “anti inflammatory” e “antimicrobials” usando o operador booleano “and” e de acordo com os Descritores em Ciências da Saúde (DeCs). A partir dos artigos foi possível observar que a Psidium guajava possui atividades antimicrobianas, anti-inflamatórias e antioxidantes bem descritas na literatura. Além disso, já existem alguns estudos in vitro analisando o uso dos extratos dessa planta no tratamento da periodontite. No entanto, mais pesquisas devem ser realizadas a fim de elucidar a concentração ideal para o tratamento, os aspectos relacionados à toxicidade e a ação sobre os patógenos periodontais, pois são escassas. Nas buscas de patentes da “Psidium guajava” a classificação que registrou maior número de depósitos na prospecção tecnológica foram as classificações internacionais de patentes (CIP) B01D (25%) e A01N (25%), seguida das C25D (12,5%), A61K (12,5%), A23C (12,5%), A23L (12,5%). Além disso, ao combinar os descritores “Psidium guajava” and “Periodontitis”, foram encontrados na EPO (15), LATIPAT (1), WIPO (1) e na USPTO (3) registros, enquanto no INPI não foi encontrado nenhum resultado. Diante disso, o uso da Psidium guajava no tratamento da periodontite constitui um cenário promissor para a realização de novas pesquisas, visto que as atividades biológicas dos extratos dela já são bem descritas na literatura e possuem resultados eficazes no tratamento de outras doenças.

Prospecção científica e tecnológica da Psidium guajava relacionada com a atividade anti-inflamatória e antimicrobiana no tratamento da periodontite

Abstract The aim of this review was to update knowledge about the prevalence of periodontitis in Latin America and the Caribbean. A critical review of was performed of all cross-sectional or cohort studies selected, pertaining to the region, and thirty-five studies conducted in 12 countries were selected. The countries with nationally representative studies were Brazil, Chile, Colombia, and Uruguay. The prevalence of periodontal disease or need for periodontal treatment varied between the different studies and countries depending on the age group, the methodology used, and the case definition. The prevalence of severe periodontitis aged between 5.8% and 49.7% in adults. In adolescents, the prevalence of moderate to severe periodontitis was 15.3%. Furthermore, a high prevalence of gingival bleeding in adolescents was reported. When analyzing the studies that used the Community Periodontal Index (CPI), Centers for Diseases Control and American Academy Periodontology (CDC/AAP) case definition, it was observed that as the age of the individuals analyzed increased, the prevalence of periodontal disease also increased. Whereas this rereview revealed that although the number of regional and nationally representative studies that analyzed the prevalence of periodontitis has risen in recent years, their methodological heterogeneity prevents global conclusions to be drawn concerning the region. Therefore, this ratifies the need to generate alliances between countries with the purpose of joining individual efforts to achieve collective goals which, among other objectives, will translate into conducting multicenter studies. These studies would allow description and monitoring of the epidemiological behavior of periodontitis in Latin America and the Caribbean.

Prevalence of periodontal diseases: Latin America and the Caribbean Consensus 2024

Abstract INTRODUCTION Epidemiological surveys revealed that Brazil has a high burden of oral diseases. However, no prior study has reported estimates of untreated dental caries, periodontitis, and edentulism over a three-decade period. The objective of this study is to report the trends of prevalence, incidence, and years-lived with disability (YLDs) due to untreated dental caries in primary and permanent teeth, periodontitis, and edentulism in Brazil between 1990 and 2019. METHODS Estimates of prevalence, incidence, and YLDs due to dental caries in primary and permanent teeth, periodontitis, and edentulism were produced for Brazil, by sex and age, between 1990 and 2019, using Dismod-MR 2.1, as part of the Global Burden of Disease Study 2019 (GBD 2019). Trends of oral disorders were analyzed using generalized linear regression models applying the Prais-Winsten method. RESULTS Almost 100 million Brazilians presented at least one oral disorder in 2019, which was equivalent to a prevalence of 45.3%. All oral diseases combined ranked eighth among all causes of disability, causing more than 970,000 YLDs. Untreated dental caries in primary teeth were estimated to affect 13.5 million children, and untreated dental caries in permanent teeth affected more than 52 million people. Periodontitis affected 29.5 million people, and edentulism affected almost 22 million. The generalized linear regression models revealed a trend of stability of oral disorders between 1990 and 2019. CONCLUSIONS The burden of oral diseases in Brazil is extremely high. Oral disorders, edentulism in particular, caused disability at levels that are comparable to other important chronic diseases.
