The present invention generally relates to microfluidics and, in particular, to systems and methods of producing droplets (55), including double and other multiple emulsion droplets (55). In some embodiments, the present invention is generally directed to systems and methods for producing multiple emulsion droplets (55). In some cases, the multiple emulsion droplets (55) may have relatively thin shells. These may be produced, for example, in a one-step process where multiple fluids (15,25,45) are brought together at a junction (18,48), or by passing the multiple emulsion droplets (55) through constrictions (76) in microfluidic channels. Other aspects of the invention are generally directed to systems and methods for using such double and other multiple emulsion droplets (55), kits involving such double and other multiple emulsion droplets (55), or the like.

Systems and methods for producing droplet emulsions with relatively thin shells

Methods and systems for sorting droplets are provided. In some cases, occupied droplets may be sorted from unoccupied droplets. In some cases, singularly occupied droplets may be sorted from unoccupied droplets and multiply occupied droplets. Methods and systems for sorting cell beads are provided. In some cases, cell beads may be sorted from particles unoccupied with cell derivatives. In some cases, singularly occupied cell beads may be sorted from unoccupied particles and multiply occupied cell beads. Methods and systems for selectively polymerizing droplets based on occupancy and size of the droplets are provided.

Devices, systems, and their methods of use, for generating droplets are provided. One or more geometric parameters of a microfluidic channel can be selected to generate droplets of a desired and predictable droplet size.

Methods and devices for forming droplets are provided. In certain embodiment's, the methods and devices form droplets having different diameters. Exemplary embodiment's of the present disclosure relate to systems and methods for forming droplets, including a multi-step microchannel emulsification device. One embodiment provides an emulsification device comprising: a channel having an inlet portion; a first step in fluid communication with the inlet portion; a second step in fluid communication with the first step; and a third step in fluid communication with the second step.

Apparatus and methods for multi-step channel emulsification

Droplet generation devices and systems that parallelize droplet generation devices are provided. The droplet generation devices can include a symmetric block-and-break system and a tapered droplet generation zone. The symmetric block-and-break system can include a pair of break channels and a pair of bypass channels symmetrically arranged with respect to the dispersed-phase input channel and the output channel. The droplet generation devices can generate monodisperse droplets with a predefined volume over a range of flow rates, pressures, and fluid properties. The droplet generation devices are therefore capable of parallelization to achieve large-capacity droplet generation, e.g. greater than 1 L/hr, with small overall coefficients of variation.

System, including methods and apparatus, for forming droplets of an emulsion. The system may include a channel junction at which a stream of sample fluid is divided into droplets by a dividing flow of carrier fluid. The system also may include one or more features configured to position sample fluid for reduced contact between the sample fluid and one or more surface regions of the channel junction, which may improve the consistency of droplet formation. In exemplary embodiments, sample fluid may be positioned by a step member produced by an increase in channel depth, and/or by directing flow of carrier fluid to form a barrier layer between sample fluid and a wall region, such as a ceiling region or floor region, of a channel network.
