The PTEN-regulating microRNA miR-26a is amplified in high-grade glioma and facilitates gliomagenesis in vivo
Jason T. Huse,Cameron Brennan,Dolores Hambardzumyan,Boyoung Wee,John Pena,Sara H. Rouhanifard,Cherin Sohn-Lee,Carlos le Sage,Reuven Agami,Thomas Tuschl,Eric C. Holland +10 more
TL;DR: It is demonstrated that miR-26a-mediated PTEN repression in a murine glioma model both enhances de novo tumor formation and precludes loss of heterozygosity and the PTEN locus.
MicroRNA-24 Regulates Vascularity After Myocardial Infarction
Jan Fiedler,Virginija Jazbutyte,Bettina C. Kirchmaier,Bettina C. Kirchmaier,Shashi Kumar Gupta,Johan M. Lorenzen,Dorothee Hartmann,Paolo Galuppo,Susanne Kneitz,John Pena,Cherin Sohn-Lee,Xavier Loyer,Juergen Soutschek,Thomas Brand,Thomas Brand,Thomas Tuschl,Joerg Heineke,Ulrich Martin,Stefan Schulte-Merker,Georg Ertl,Stefan Engelhardt,Johann Bauersachs,Thomas Thum +22 more
TL;DR: It is shown that the small noncoding RNA microRNA-24 (miR-24) is enriched in cardiac endothelial cells and considerably upregulated after cardiac ischemia, and is suitable for therapeutic intervention in the setting of ischemic heart disease.
miRNA in situ hybridization in formaldehyde and EDC-fixed tissues.
John Pena,Cherin Sohn-Lee,Sara H. Rouhanifard,Janos Ludwig,Janos Ludwig,Markus Hafner,Aleksandra Mihailovic,Cindy Lim,Daniel Holoch,Philipp Berninger,Mihaela Zavolan,Thomas Tuschl +11 more
TL;DR: It is shown that in situ hybridization using conventional formaldehyde fixation results in substantial microRNA loss from mouse tissue sections, which can be prevented by fixation with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide that irreversibly immobilizes the microRNA at its 5′ phosphate.
Barcoded cDNA library preparation for small RNA profiling by next-generation sequencing
TL;DR: A step-by-step protocol for generating barcoded small RNA cDNA libraries compatible with Illumina HiSeq sequencing is presented, thereby facilitating miRNA and other small RNA profiling of large sample collections.

Inhibition of neuronal phenotype by PTEN in PC12 cells
Sergei Musatov,Jill Roberts,Andrew I. Brooks,John Pena,Simone A. Betchen,Donald W. Pfaff,Michael G. Kaplitt,Michael G. Kaplitt +7 more
TL;DR: It is reported that the tumor suppressor PTEN has a profound effect on differentiation by affecting several pathways involved in nerve growth factor (NGF) signaling, and a previously uncharacterized mechanism of PTEN action in neuron-like cells is suggested.
