Dendritic cells from patients with myeloma are numerically normal but functionally defective as they fail to up-regulate CD80 (B7-1) expression after huCD40LT stimulation because of inhibition by transforming growth factor-β1 and interleukin-10
Ross D. Brown,Belinda Pope,Allan Murray,Warren Esdale,Daniel M. Sze,John Gibson,P. Joy Ho,Derek N.J. Hart,Doug Joshua +8 more
TL;DR: It has been determined that patients with myeloma have normal numbers of DCs, but CD80 expression may fail to be up-regulated in the presence of tumor-derived TGF-beta1 or IL-10, and autologous high-potency DCs may have to be tested for CD80 up-regulation and biologically modified ex vivo before idiotype priming for immunotherapy.

Multiple myeloma causes clonal T-cell immunosenescence: identification of potential novel targets for promoting tumour immunity and implications for checkpoint blockade.
Hayley Suen,Ross Brown,S Yang,Claire Weatherburn,Claire Weatherburn,Phoebe Joy Ho,Phoebe Joy Ho,Narelle Woodland,Najah T. Nassif,Pasquale M Barbaro,Christian E Bryant,Christian E Bryant,Derek N.J. Hart,John Gibson,John Gibson,Douglas E. Joshua,Douglas E. Joshua +16 more
TL;DR: Low programmed death 1 and cytotoxic T-lymphocyte-associated antigen 4 expression detected on T-cell clones infers that these cells are not exhausted but suggests that there would be a suboptimal response to immune checkpoint blockade in MM.
Long-term survival in multiple myeloma is associated with a distinct immunological profile, which includes proliferative cytotoxic T-cell clones and a favourable Treg/Th17 balance
Christian E Bryant,Hayley Suen,Hayley Suen,Ross Brown,S Yang,James Favaloro,Esther Aklilu,John Gibson,Phoebe Joy Ho,Harry J. Iland,Phillip D. Fromm,Phillip D. Fromm,Narelle Woodland,Najah T. Nassif,Derek N.J. Hart,Derek N.J. Hart,Douglas E. Joshua,Douglas E. Joshua +17 more
TL;DR: It is proposed that immune factors contribute to this longer survival in multiple myeloma and long-term survival in MM is associated with a distinct immunological profile, which is consistent with decreased immune suppression.
Autotransplants for Hodgkin's disease in first relapse or second remission: a report from the autologous blood and marrow transplant registry (ABMTR)
Hillard M. Lazarus,Fausto R. Loberiza,Mei-Jie Zhang,James O. Armitage,Karen K. Ballen,Asad Bashey,Brian J. Bolwell,Linda J. Burns,Cesar O. Freytes,Robert Peter Gale,John Gibson,Roger H. Herzig,Charles F. LeMaistre,David I. Marks,J. Mason,Alan M. Miller,Gustavo Milone,Santiago Pavlovsky,D. E. Reece,J D Rizzo,K. Van Besien,Julie M. Vose,Mary M. Horowitz +22 more
TL;DR: High-dose chemotherapy and autologous hematopoietic stem cell transplantation (autotransplantation) may provide long-term progression-free survival in patients with relapsed Hodgkin's disease and future investigations should focus on strategies designed to decrease relapse after autotrans transplantation, particularly in patients at high risk for relapse.

Is the international staging system superior to the durie salmon staging system? a comparison in multiple myeloma patients undergoing autologous transplant
Parameswaran Hari,Mei-Jie Zhang,Vivek Roy,Waleska S. Pérez,Asad Bashey,Luen Bik To,Gerald J. Elfenbein,Cesar O. Freytes,Robert Peter Gale,John Gibson,Robert A. Kyle,Hillard M. Lazarus,Philip L. McCarthy,Gustavo Milone,Santiago Pavlovsky,Donna E. Reece,Gary J. Schiller,Jorge Vela-Ojeda,Daniel J. Weisdorf,David H. Vesole +19 more
TL;DR: Although both systems were predictive of PFS and OS, the DSS was superior in formal statistical comparison using Brier score, indicating the need for newer schemes incorporating other prognostic markers.
