A Safety Assessment of Tumor Necrosis Factor Antagonists During Pregnancy : A Review of the Food and Drug Administration Database
TL;DR: A seemingly high number of congenital anomalies that are part of the VACTERL spectrum have been reported, occurring at a rate higher than historical controls and raises concerns of a possible causative effect of the TNF antagonists.

John D. Carter,Luis R. Espinoza,Robert D. Inman,K. B. Sneed,Louis R. Ricca,Frank B. Vasey,Joanne Valeriano,Jessica A. Stanich,Cynthia Oszust,Hervé C. Gérard,Alan P. Hudson +10 more
TL;DR: These data suggest that a 6-month course of combination antibiotics is an effective treatment for chronic Chlamydia-induced ReA.

Efficacy and safety of olokizumab in patients with rheumatoid arthritis with an inadequate response to TNF inhibitor therapy: outcomes of a randomised Phase IIb study
Mark C. Genovese,Roy Fleischmann,Daniel E. Furst,Namieta M. Janssen,John D. Carter,Bhaskar Dasgupta,Judy Bryson,Benjamin Duncan,Wei Zhu,Costantino Pitzalis,Patrick Durez,Kosmas Kretsos +11 more
TL;DR: OkZ treatment, at several doses, demonstrated similar efficacy to TCZ across multiple endpoints, and Pharmacokinetic/pharmacodynamic modelling demonstrated a shallow dose/exposure response relationship for OKZ, consistent with the safety profile expected of this class of drug.

Tumor necrosis factor-alpha inhibition and VATER association: a causal relationship.
TL;DR: It is proposed that the TNF antagonists, specifically etanercept, be used with caution in pregnant women, and patient registries of women who take TNF-a antagonists during pregnancy need to be followed to see if there is an increase in the birth defects that are part of VATER association.

Rilonacept in the treatment of acute gouty arthritis: a randomized, controlled clinical trial using indomethacin as the active comparator.
Robert Terkeltaub,H. Ralph Schumacher,John D. Carter,Herbert S B Baraf,Robert Evans,Jian Wang,Shirletta King-Davis,Steven P. Weinstein +7 more
TL;DR: Although generally well-tolerated, rilonACEpt in combination with indomethacin and rilonacept alone did not provide additional pain relief over 72 hours relative to indometHacin alone in patients with acute gout flare.
