Recently, two alternative targets in insect periphery nerve system have been explored for environmentally-friendly approaches in insect pest management, namely odorant-binding proteins (OBPs) and odorant receptors (ORs). Located in insect antennae, OBPs are thought to be involved in the transport of odorants to ORs for the specific signal transduction of behaviorally active odorants. There is rich information on OBP binding affinity and molecular docking to bioactive compounds as well as ample 3D crystal structures due to feasible production of recombinant proteins. Although these provide excellent opportunities for them to be considered as pest control targets and a tool to design pest control agents, the debates on their binding specificity represent an obstacle. On the other hand, ORs have recently been functionally characterized with increasing evidence for their specificity, sensitivity and functional roles in pest behaviors. However, a major barrier to use ORs for semiochemical discovery is the lack of 3D crystal structures. Thus, OBPs and ORs have not been analyzed comparatively together so far for their feasibility as pest control targets. Here, we summarize the state of OBPs and ORs research in terms of its application in insect pest management. We discuss the suitability of both proteins as pest control targets and their selection toward the discovery of new potent semiochemicals. We argue that both proteins represent promising targets for pest control and can be used to identify new super-ligands likely present in nature and with reduced risk of resistance development than insect pesticides currently used in agriculture. We discuss that with the massive identification of OBPs through RNA-seq and improved binding affinity measurements, these proteins could be reconsidered as suitable targets for semiochemical discovery.

Sexual communication in moths offers a simplified scenario to model and investigate insect sensory perception Both PBPs (pheromone-binding proteins) and PRs (pheromone receptors) are involved in the detection of sex pheromones, but the interplay between them still remains largely unknown In this study, we have measured the binding affinities of the four recombinant PBPs of Chilo suppressalis (CsupPBPs) to pheromone components and analogs and characterized the six PRs using the Xenopus oocytes expression system Interestingly, when the responses of PRs were recorded in the presence of PBPs, we measured in several combinations a dramatic increase in signals as well as in sensitivity of such combined systems Furthermore, the discrimination ability of appropriate combinations of PRs and PBPs was improved compared with the performance of PBPs or PRs alone Besides further supporting a role of PBPs in the pheromone detection and discrimination, our data shows for the first time that appropriate combinations of PRs and PBPs improved the discrimination ability of PBPs or PRs alone The variety of responses measured with different pairing of PBPs and PRs indicates the complexity of the olfaction system, which, even for the relatively simple task of detecting sex pheromones, utilises a highly sophisticated combinatorial approach

Olfactory receptors (ORs) are the key elements of the molecular machinery responsible for the detection of odors in insects. Since their initial discovery in Drosophila melanogaster at the beginning of the twenty-first century, insect ORs have been the focus of intense research, both for fundamental knowledge of sensory systems and for their potential as novel targets for the development of products that could impact harmful behaviors of crop pests and disease vectors. In recent years, studies on insect ORs have entered the genomic era, with an ever-increasing number of OR genes being characterized every year through the sequencing of genomes and transcriptomes. With the upcoming release of genomic sequences from hundreds of insect species, the insect OR family could very well become the largest multigene family known. This extremely rapid identification of ORs in many insects is driving the necessity for the development of high-throughput technologies that will allow the identification of ligands for this unprecedented number of receptors. Moreover, such technologies will also be important for the development of agonists or antagonists that could be used in the fight against pest insects.

Advances in the identification and characterization of olfactory receptors in insects.

Odorant receptors (ORs) are essential for plant-insect interactions. However, despite the global impacts of Lepidoptera (moths and butterflies) as major herbivores and pollinators, little functional data are available about Lepidoptera ORs involved in plant-volatile detection. Here, we initially characterized the plant-volatile-sensing function(s) of 44 ORs from the cotton bollworm Helicoverpa armigera, and subsequently conducted a large-scale comparative analysis that establishes how most orthologous ORs have functionally diverged among closely related species whereas some rare ORs are functionally conserved. Specifically, our systematic analysis of H. armigera ORs cataloged the wide functional scope of the H. armigera OR repertoire, and also showed that HarmOR42 and its Spodoptera littoralis ortholog are functionally conserved. Pursuing this, we characterized the HarmOR42-orthologous ORs from 11 species across the Glossata suborder and confirmed the HarmOR42 orthologs form a unique OR lineage that has undergone strong purifying selection in Glossata species and whose members are tuned with strong specificity to phenylacetaldehyde, a floral scent component common to most angiosperms. In vivo studies via HarmOR42 knockout support that HarmOR42-related ORs are essential for host-detection by sensing phenylacetaldehyde. Our work also supports that these ORs coevolved with the tube-like proboscis, and has maintained functional stability throughout the long-term coexistence of Lepidoptera with angiosperms. Thus, beyond providing a rich empirical resource for delineating the precise functions of H. armigera ORs, our results enable a comparative analysis of insect ORs that have apparently facilitated and currently sustain the intimate adaptations and ecological interactions among nectar feeding insects and flowering plants.

The widespread planting of genetically engineered cotton producing the Cry1Ac toxin has led to significantly reduced pesticide applications since 1997. However, consequently, the number of green mirid bugs (GMB), Apolygus lucorum (Meyer-Dur) has increased. So far the GMB, instead of the cotton bollworm Helicoverpa armigera (Hubner), has become the major pest in the transgenic Bt cotton field and has influenced cotton yield. Disproportionately, only a few studies on GMB at molecular level have been reported. Libraries from both third instar nymphs and adults were sequenced using Illumina technology, producing more than 106 million short reads and assembled into 63 029 unigenes of mean length 597 nt and N50 813 nt, ranging from 300 nt to 9771 nt. BLASTx analysis against Nr, Swissprot, GO and COG was performed to annotate these unigenes. As a result, 26 478 unigenes (42.01%) matched to known proteins and 107 immune-related, 320 digestive-related and 53 metamorphosis-related genes were detected in these annotated unigenes. Additionally, we profiled gene expression using mapping based differentially expressed genes (DEGs) strategy between the two developmental stages: nymph and adult. The results demonstrated that thousands of genes were significantly differentially expressed at different developmental stages. The transcriptome and gene expression data provided comprehensive and global gene resources of GMB. This transcriptome would improve our understanding of the molecular mechanisms of various underlying biological characteristics, including development, digestion and immunity in GMB. Therefore, these findings could help elucidate the intrinsic factors of the GMB resurgence, offering novel pest management targets for future transgenic cotton breeding.

Antennal olfaction, which is extremely important for insect survival, mediates key behaviors such as host preference, mate choice, and oviposition site selection. In insects, odor detection is mediated by multiple proteins in the antenna, especially the odorant receptors (ORs) and ionotropic receptors (IRs), which ensure the specificity of the olfactory sensory neuron responses. In this study, we identified the olfactory gene repertoire of the rice stem borer, Chilo suppressalis, an economically important agricultural pest, which inflicts great damage to the rice yield in south and east part of Asia, especially in Southern China. By Illumina sequencing of male and female antennal transcriptomes, we identified 47 odorant receptors, 20 ionotropic receptors, 26 odorant binding proteins, 21 chemosensory proteins and 2 sensory neuron membrane proteins. Our findings make it possible for future research of the olfactory system of C. suppressalis at the molecular level.
