The tuning of photo physical properties of highly emissive carbon quantum dots which was synthesized from citric acid in DMF medium through solvothermal method is investigated inside micelles formed by different types of surfactants like cationic surfactant cetylpyridinium chloride (CPC), cetrimonium bromide (CTAB), anionic surfactant sodium dodecyl sulfate (SDS) and neutral surfactant polyethylene glycol tert-octyl phenyl ether (TX100), polyoxoethylene lauryl ether (BRIJ-35). Interestingly, only CPC interacts with greater extent and formed co-assembly with negatively-charged carbon quantum dots (CQDs) whereas other surfactants like CTAB, SDS, TX100 and BRIJ-35 do not alter the fluorescence intensity extensively. This results in the formation of a non fluorescent vesicle. No phase separation was observed over the large concentration barrier of the CPC surfactant. However, no such vesicle formation takes place for another cationic surfactant CTAB excluding the role of only electrostatic interaction. The synthesized vesicle is characterized by various techniques including HRTEM, DLS. For the aqueous solution of CQDs having concentration of 0.5 mg/ml, addition of CPC leads to formation of different sized vesicles of size 40 nm to 100 nm. The driving force behind this formation of non-fluorescent vesicles is oxidative photo induced electron transfer which is revealed by the highest occupied molecular orbital (HOMO) –lowest unoccupied molecular orbital (LUMO) interaction between the donor (CQDs) and the acceptor (CPC). We trust such type vesicle can be a suitable alternative for drug carriers, biomarkers and other various applications also.

The early diagnosis of cancer is vital for effective treatment and improved prognosis. Tumor biomarkers, which can be used for the early diagnosis, treatment, and prognostic evaluation of cancer, have emerged as a topic of intense research interest in recent years. Nucleic acid, as a type of tumor biomarker, contains vital genetic information, which is of great significance for the occurrence and development of cancer. Currently, living cell nucleic acid probes, which enable the in situ imaging and dynamic monitoring of nucleic acids, have become a rapidly developing field. This review focuses on living cell nucleic acid probes that can be used for the early diagnosis of tumors. We describe the fundamental design of the probe in terms of three units and focus on the roles of different nanomaterials in probe delivery.

High-order Framework Nucleic Acid for Targeted-delivery of Antisense Peptide Nucleic Acid to Overcome Drug Resistance

Designing dynamic assemblies in living cells is crucial for creating organelle‐like structures, yet precisely controlling their morphological transitions in response to specific signals is a significant challenge. In this study, a DNA framework is combined with hybridization chain reaction (HCR) to achieve specific assembly of hyperbranched aggregates in cancer cells. HCR, distinguished for its signal amplification and linear extension capabilities, enables the morphological transition of precursors to be specifically triggered by trace amounts of endogenous microRNA‐21 (miR‐21). The spatial constraints of the framework and the diversity of hairpin orientations significantly accelerate the assembly kinetics of hyperbranched networks, and the resulting micrometer‐scale aggregates possess enhanced intracellular retention capabilities. Introducing Ce6 molecules as a proof of concept, the regulatory function of aggregates can be activated under light irradiation and remains effective over a long period. The probe we constructed  demonstrates good stability and biocompatibility, offers easy functionalization, and works inside cells long‐term, making it an ideal candidate material for the construction of organelle‐like structures.

Multi-target recognition and DNAzyme signal amplification integrated functional nucleic acid framework for lncRNA in situ live cell imaging

Framework nucleic acids (FNAs) represent a new type of DNA-based nanomaterials and possess great potentials in biosensing, bioimaging, and molecular delivery. Hierarchical DNA nanostructures that consist of multiple FNA monomers increase the capacity for drug delivery and multifunctional modification. However, there are relatively few studies devoted to the behavior and regulation of hierarchical FNAs in living cells, impeding their further applications. Herein, we constructed a dendritic nanostructure with five tetrahedral DNA nanocages and characterized the real-time internalization, inter-organelle trafficking, and exocytosis in living mammalian cells. In comparison to FNA monomers, FNA dendrimers exhibit increased endocytosis and prolonged cellular retention. Single-particle tracking on hundreds of FNA dendrimers exhibits no interference on the mobility or kinetics of subcellular organelles, implying that FNAs as well as their higher-order derivatives are ideal intracellular imaging probes and nanocarriers. Our study validates the suitability and superiority of hierarchical DNA nanostructures as high-valency scaffolds for biomedical applications.
