An aerobic, Gram-stain-negative, straight or curved rods and dimorphic prosthecate bacterium designated as strain LZ-16-1T was isolated from phycosphere microbiota of routinely laboratory-cultured and highly-toxic marine dinoflagellate Alexandrium catenella LZT09. Strain LZ-16-1T produces active bioflocculanting exopolysaccharides (EPS). Cells were dimorphic with non-motile prostheca, or non-stalked and motile by a single polar flagellum. Growth occurred at 10–40 °C, pH 5–9 and 1–8% (w/v) NaCl, with optimum growth at 25 °C, pH 7–8 in the presence of 2–4% (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain LZ-16-1T was affiliated to the genus Maricaulis, and closely related to M. parjimensis MCS 25T (99.5%) and M. virginensis VC-5T (99.0%). However, based on genome sequencing and phylogenomic calculations, the average nucleotide identity (ANI) and digtal DNA-DNA genome hybridization (dDDH) values between strains LZ-16-1T and its closest relative, M. parjimensis MCS 25T were only 85.0 and 20.9%, respectively. The dominant fatty acids of strain LZ-16-1T were summed feature 8, C16:0, C17:0, C18:0, C18:1 ω9c and summed feature 9. Major polar lipids were sulfoquinovosyl diacylglycerol, six glycolipids, one unidentified phospholipid and one unidentified polar lipid. The predominant isoprenoid quinone was Q-10. The DNA G + C content calculated from the genome was 63.6 mol%. Physiological and chemotaxonomic characterizations further confirmed the distinctiveness of strain LZ-16-1T from other Maricaulis members. Thus, strain LZ-16-1T represents a novel species of the genus Maricaulis, for which the name Maricaulis alexandrii sp. nov. (type strain LZ-16-1T = KCTC 72194T = CCTCC AB 2019006T) is proposed.

Maricaulis alexandrii sp. nov., a novel active bioflocculants-bearing and dimorphic prosthecate bacterium isolated from marine phycosphere

Abstract Three bacterial strains, FP830T, FP2034, and FP2262, were isolated from the rhizosphere soil of rice, corn, and highland barley plants in Beijing, Heilongjiang, and Tibet, respectively, in PR China. These strains are gram-negative, aerobic, and rod-shaped. They exhibited optimal growth at 28°C and pH 7.0 in the presence of 2% (w/v) NaCl and produced a diffusible fluorescent pigment under ultraviolet light when cultured on King's B plates. Phylogenetic analysis based on 16S rRNA gene and multilocus sequence data showed that FP830T, FP2034, and FP2262 belonged to the genus Pseudomonas and formed a distinct branch. The genome sizes of these three isolates ranged from 6.4 to 6.6 Mbp with a G+C content of 60.86-61.02 mol%. The orthologous average nucleotide identity and digital DNA-DNA hybridization values of strains FP830T, FP2034, and FP2262, along with their closest relatives, were not higher than 92.7% and 49.8%, respectively. These values were lower than the threshold range for prokaryotic species delineation. The dominant cellular fatty acids (&gt; 10%) were C16:0, summed feature 3 (C16:1 ω7c and/or C16:1 ω6c), and summed feature 8 (C18:1 ω7c). The major polar lipids detected were phosphatidylethanolamine, diphosphatidylglycerol, and aminophospholipids. Additionally, the predominant respiratory quinone was ubiquinone (Q-9). Based on phenotypic, chemotaxonomic, and genotypic data, it was concluded that strains FP830T, FP2034, and FP2262 represented a novel species within the genus Pseudomonas. Therefore, the name Pseudomonas beijingensis sp. nov. was proposed for this novel species. The type strain was FP830T (=ACCC 62448T = JCM 35689T). 

Pseudomonas beijingensis sp. nov., a novel species widely colonizing plant rhizosphere

A Gram-negative aerobic bacterium, strain M30-35 T, was isolated from the rhizosphere of Haloxylon ammodendron in Tengger desert, Gansu province, northwest China. Our previous research indicated that strain M30-35 T can promote the growth of ryegrass (Lolium perenne L.). In this study, strain M30-35 T was subjected to a polyphasic taxonomic study. Phylogenetic analysis of the 16S rRNA gene and two other housekeeping genes (gyrB, rpoD) showed that strain M30-35 T is a member of Pseudomonas anguilliseptica group. The average nucleotide identity (ANI) scores for strains KMM 3042 T and FR1439T were 76.5% and 83.7%, respectively, and DNA-DNA hybridization (DDH) were 21.6% and 26.6%, respectively, and the rates were less than the threshold range for species determination. The dominant cellular fatty acids of strain M30-35 T were C16:0 (22.7%), summed feature 3 (C16:1ω7c and/or C16:1ω6c; 18.5%), summed feature 8 (C18:1ω7c and/or C18:1ω6c; 23.1%). The major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phospholipid and aminophospholipid and the predominant respiratory quinone was ubiquinone (Q9). On the basis of above data, it can be concluded that strain M30-35 T represents a novel species in the genus Pseudomonas, for which the name Pseudomonas rhizovicinus sp. nov. is proposed. The type strain is M30-35 T (= MCCC 1K03247T = KCTC 52664 T).
