Thrombin receptor activation. Confirmation of the intramolecular tethered liganding hypothesis and discovery of an alternative intermolecular liganding mode.
TL;DR: While intermolecular liganding between two throm bin receptor molecules can occur, the intramolecular tethered liganding mechanism is the predominant mode of thrombin receptor activation, and the release from inhibition hypothesis is refuted.

Thrombin Receptor Activation CONFIRMATION OF THE INTRAMOLECULAR TETHERED LIGANDING HYPOTHESIS AND DISCOVERY OF AN ALTERNATIVE INTERMOLECULAR LIGANDING MODE* (Received for publication, February 18, 1994, and in revised form, March 29, 1994)

- 01 Jan 1994
TL;DR: In this paper, it was shown that the SFLLRN sequence is a tethered peptide ligand; receptor cleavage unmasks this agonist which then binds intramolecularly to effect receptor activation.

Mechanisms of Thrombin Receptor Agonist Specificity CHIMERIC RECEPTORS AND COMPLEMENTARY MUTATIONS IDENTIFY AN AGONIST RECOGNITION SITE
Tania Nanevicz,Maki Ishii,Ling Wang,Mian Chen,Ji Chen,Christoph W. Turck,Fred E. Cohen,Shaun R. Coughlin +7 more
TL;DR: Two independent approaches, chimeric receptors and arginine scanning for complementary mutations, identified the Glu region and to a lesser degree Phe as important determinants of agonist specificity.

Thrombin receptor structure and function.
Kenji Ishii,Ji Chen,Maki Ishii,Thienkhai Vu,Robert E. Gerszten,Tania Nanevicz,Ling Wang,Shaun R. Coughlin +7 more
TL;DR: Thrombin is a multifunctional serine protease generated at sites of vascular injury or wounding that is the most potent activator of platelets in vitro and mediates not only hemostatic but inflammatory and proliferative or reparative responses.

- 07 Jun 1995
TL;DR: In this article, a tethered ligand was used for probing receptor binding and the sequence of the ligand that produces signal as a result of the interaction between the agonist and receptor can be then be isolated using sib selection.
