A double transgenic mouse model expressing human pregnane X receptor and cytochrome P450 3A4.
Xiaochao Ma,Connie Cheung,Connie Cheung,Kristopher W. Krausz,Yatrik M. Shah,Ting Wang,Jeffrey R. Idle,Frank J. Gonzalez +7 more
TL;DR: The TgCYP3A4/hPXR mouse model was generated by bacterial artificial chromosome transgenesis in Pxr-null mice and suggested that it could serve as a useful tool for studies on CYP3A 4 transcription and function in vivo.

A population and familial study of the defective alicyclic hydroxylation of debrisoquine among Egyptians.
TL;DR: Egyptians appear in general to be more extensive oxidizers of debrisoquine than do English subjects and Ramadan fasting was found to lower the absorption of debrisquine.

Comparative Metabolism of Cyclophosphamide and Ifosfamide in the Mouse Using UPLC-ESI-QTOFMS-Based Metabolomics
Fei Li,Andrew D. Patterson,Constance C. Höfer,Kristopher W. Krausz,Frank J. Gonzalez,Jeffrey R. Idle,Jeffrey R. Idle +6 more
TL;DR: Of the total 23 drug metabolites identified by UPLC-ESI-QTOFMS for both IF and CP, five were found to be novel and may suggest that other metabolites, perhaps precursors of thiodiglycolic acid, may be responsible for IF encephalopathy and nephropathy.

A comprehensive investigation of 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) metabolism in the mouse using a multivariate data analysis approach.
Chi Chen,Xiaochao Ma,Michael A. Malfatti,Kristopher W. Krausz,Shioko Kimura,James S. Felton,Jeffrey R. Idle,Frank J. Gonzalez +7 more
TL;DR: In vitro data supported the view that CYP1A2 is the major enzyme responsible for the formation of N2-OH-PhIP, but disruption of the CYP2C gene in mouse failed to inhibit PhIP-induced carcinogenesis and a human-mouse interspecies difference in the catalytic activity of CYP 1A2 was revealed.

Determination of nifedipine and its three principal metabolites in plasma and urine by automated electron-capture capillary gas chromatography.
TL;DR: A sensitive, efficient, linear and reproducible capillary gas chromatographic method with electron-capture detection was developed for the quantitation of nifedipine and its primary metabolite M-I in plasma together with the urinary and principal metabolites M-II and M-III.
