ADAR1 promotes malignant progenitor reprogramming in chronic myeloid leukemia
Qingfei Jiang,Leslie Crews,Christian L. Barrett,Hye Jung E. Chun,Angela C. Court,Jane Isquith,Jane Isquith,Maria A. Zipeto,Maria A. Zipeto,Daniel Goff,Mark D. Minden,Anil Sadarangani,Jessica M. Rusert,Kim Hien T. Dao,Sheldon R. Morris,Lawrence S.B. Goldstein,Marco A. Marra,Kelly A. Frazer,Catriona Jamieson +18 more
TL;DR: In this paper, whole-transcriptome sequencing of normal, chronic phase, and serially transplantable blast crisis chronic myeloid leukemia (CML) progenitors revealed increased IFNγ pathway gene expression in concert with BCR-ABL amplification, enhanced expression of the IFN-responsive ADAR1 p150 isoform, and a propensity for increased adenosine-to-inosine RNA editing during CML progression.

Hyper-Editing of Cell-Cycle Regulatory and Tumor Suppressor RNA Promotes Malignant Progenitor Propagation
Qingfei Jiang,Jane Isquith,Maria A. Zipeto,Raymond Diep,Jessica Pham,Nathan Delos Santos,Eduardo Reynoso,Julisia Chau,Heather Leu,Elisa Lazzari,Etienne Melese,Wenxue Ma,Rongxin Fang,Rongxin Fang,Mark D. Minden,Sheldon R. Morris,Bing Ren,Bing Ren,Gabriel Pineda,Frida Holm,Catriona Jamieson +20 more
TL;DR: It is shown that ADAR1-induced hyper-editing in normal human hematopoietic progenitors impairs miR-26a maturation, which represses CDKN1A expression indirectly via EZH2, thereby accelerating cell-cycle transit and enhancing blast crisis progenitor propagation.

Malignant A-to-I RNA editing by ADAR1 drives T cell acute lymphoblastic leukemia relapse via attenuating dsRNA sensing.
Maria Rivera,Haoran Zhang,Jessica Pham,Jane Isquith,Qingchen Jenny Zhou,Larisa Balaian,Roman Sasik,Sabina Enlund,Adam Mark,Wen-Shu Ma,Frida Holm,Kathleen M. Fisch,D. Kuo,Catriona Jamieson,Qingfei Jiang +14 more
TL;DR: It is shown that the RNA-editing enzyme ADAR1 is a crucial stemness factor that promotes LIC self-renewal by attenuating aberrant double-stranded RNA (dsRNA) sensing and functions as a self-renewal factor that limits the sensing of endogenous dsRNA.

Inflammation-driven deaminase deregulation fuels human pre-leukemia stem cell evolution.
Qingfei Jiang,Jane Isquith,Luisa Ladel,Adam Mark,Frida Holm,Cayla Mason,Yudou He,Phoebe Mondala,Isabelle Oliver,Jessica Pham,Wenxue Ma,Eduardo Reynoso,Shawn Ali,Isabella Jamieson Morris,Raymond Diep,Chanond A Nasamran,Guorong Xu,Roman Sasik,Sara Brin Rosenthal,Amanda Birmingham,Sanja Coso,Gabriel Pineda,Leslie Crews,Mary Donohoe,J. Craig Venter,Thomas Whisenant,Ruben A. Mesa,Ludmil B. Alexandrov,Kathleen M. Fisch,Catriona Jamieson +29 more
TL;DR: In this paper, the role of base deaminases in pre-leukemia stem cell (pre-LSC) evolution to acute myeloid leukemia stem cells (LSCs) had not been elucidated.

Malignant A-to-I RNA editing by ADAR1 drives T-cell acute lymphoblastic leukemia relapse via attenuating dsRNA sensing
Jessica Pham,Jane Isquith,Maria I. Rivera,Hao Zhang,Qin-Qin Zhou,Roman Sasik,Adam Mark,Wen-Shu Ma,Frida Holm,Kathleen M. Fisch,Catriona Jamieson,Qingfei Jiang +11 more
TL;DR: In this paper , the RNA editing enzyme ADAR1 is shown to be a crucial stemness factor that promotes leukemia self-renewal by attenuating aberrant double-stranded RNA (dsRNA) sensing.
