Summary Transgenesis is identified as being of special interest in the study of growth factors where their multicellular origins and complex interactions make them particularly difficult to characterize using classical experimental, approaches developed to investigate hormones originating in specialized cells in discrete glands. Through allowing molecular ‘tinkering’ in intact animals, transgenesis enables specific growth factors to be ‘ablated or replaced’ from specific tissues and organs and target cell response and impact of modulatory factors such as binding proteins to be explored in the intact animal. To the endocrinologist, the potential applications of such technology are legend. This chapter provides a brief overview of the technique and provides linkages to the rapidly developing body of literature in establishing transgenesis in growth factor research.

El problema que plantea el estudio de la enfermedad de Alzheimer, aparte de ser una enfermedad multigenica, es que no se tiene un modelo de estudio ya sea in vivo o in vitro que nos permita entender, de manera factible, como es que se inicia la enfermedad. Los modelos hasta ahora propuestos son ratas o ratones transgenicos, sin embargo estos a lo largo de la vida no presentaran todas las caracteristica de la enfermedad, lo mismo sucede con los cultivos celulares. Es necesario que se acepte y se defina la enfermedad en estos modelos y que cumplan con las caracteristicas que hasta ahora solo ofrecen los estudios post-mortem.

Transgenic mice carry cloned DNA fragments that have been introduced into the mouse genome, generally by microinjection into pronuclei. They can be used to determine the cis-acting DNA elements responsible for the tissue-specific expression of genes. For this purpose, transgenic mice were obtained carrying either the mouse albumin gene or the human insulin gene. In the first instance, it was shown that the regulatory sequences necessary for expression of the albumin gene transfected into differentiated hepatoma cells are not sufficient to induce its expression in transgenic mice. In contrast with the albumin gene, the short sequence upstream to the insulin gene necessary for its expression in transfected insulinoma cells is sufficient to allow expression in transgenic mice. Similar experiments were performed to study another crucial step in differentiation of lymphoid B cells, namely rearrangement of the immunoglobulin (Ig) genes. The chicken Ig lambda light chain gene in the germ line (unrearranged) form was introduced into the mouse genome, and the rearrangement observed in transgenic mouse lines is briefly described.
