17-Beta-estradiol induces transformation and tumorigenesis in human breast epithelial cells
Jose Russo,Sandra V. Fernandez,Patricia A. Russo,Rachael Fernbaugh,Fathima Sheriff,Hasan M. Lareef,James Garber,Irma H. Russo +7 more
TL;DR: The induction of complete transformation of MCF‐10F cells in vitro confirms the carcinogenicity of E2, supporting the concept that this hormone could act as an initiator of breast cancer in women.

The estrogen antagonist ICI-182-780 does not inhibit the transformation phenotypes induced by 17-beta-estradiol and 4-OH estradiol in human breast epithelial cells.
TL;DR: It is demonstrated that E(2) and 4-OH-E(2), at all the doses tested, increase cell proliferation and induce transformation in MCF-10F cells, phenomena that are not abrogated by ICI-182-780.

VP1-2022: Pre-specified event driven analysis of Overall Survival (OS) in the OlympiA phase III trial of adjuvant olaparib (OL) in germline BRCA1/2 mutation (gBRCAm) associated breast cancer
A. Tutt,James Garber,R. D. Gelber,Kelly-Anne Phillips,Andrea Eisen,Oskar T. Johannsson,PS Rastogi,Karen Y. Cui,Sa Im,Rinat Yerushalmi,Adam Brufsky,M Taboada,G. Rossi,Greg Yothers,C. Singer,Luis Fein,Niklas Loman,D. Cameron,Carolyn Campbell,CE Geyer +19 more
TL;DR: OlympiA as discussed by the authors is the first randomized clinical trial to test a PARP inhibitor as adjuvant therapy and showed that OL significantly improved the primary endpoint of invasive disease-free survival (IDFS) and secondary endpoint of distant DFS (DDFS) compared with placebo (PL).
