Cellular dynamics following CAR T cell therapy are associated with response and toxicity in relapsed/refractory myeloma.
Luise Fischer,N. Grieb,Patrick Born,Ronald Weiss,Sabine Seiffert,Andreas Boldt,Stephan Fricke,Paul Franz,Simone Heyn,Anne Sophie Kubasch,Ronny Baber,Heike Weidner,S. Wang,Enrica Bach,Sandra Hoffmann,J. Ussmann,Janine Kirchberg,Saskia Hell,Sebastian Schwind,K. Metzeler,M. Herling,Madlen Jentzsch,Georg-Nikolaus Franke,Ulrich Sack,Kristin Reiche,Ulrike Köhl,Uwe Platzbecker,Vladan Vucinic,Maximilian Merz +28 more
TL;DR: The results show that in vivo expansion of CD8+ CAR T cells is correlated to response, however persistence is not essential for durable remission in RRMM patients, and provide evidence, that an increased fraction of CD8+ T cells at day of leukapheresis in combination with successful lymphodepletion positively influence the outcome.

Single-cell multiomic dissection of response and resistance to chimeric antigen receptor T cells against BCMA in relapsed multiple myeloma.
Michael Rade,N. Grieb,Ronald Weiss,Jaren Sia,Luise Fischer,Patrick Born,Andreas Boldt,Stephan Fricke,Paul Franz,Jonathan Scolnick,Lakshmi Venkatraman,Stacy Xu,Christina Kloetzer,Simone Heyn,Anne Sophie Kubasch,Ronny Baber,S. Platzbecker,Kristin Reiche,Vladan Vucinic,M. Merz +32 more
TL;DR: This work subjected mononuclear cells isolated from peripheral blood and bone marrow before and after the application of approved B cell maturation antigen-directed CAR T cells to single-cell multiomic analyses to identify markers associated with resistance and early relapse.

Prognostic Impact of the 2022 European Leukemia Net Risk Classification in Patients with Acute Myeloid Leukemia Undergoing Allogeneic Stem Cell Transplantation
Madlen Jentzsch,L. Bischof,J. Ussmann,Donata Backhaus,Dominic Brauer,Klaus H. Metzeler,Marco Herling,Maximilian Merz,Vladan Vucinic,Georg-Nikolaus Franke,Uwe Platzbecker,Sebastian Schwind +11 more
TL;DR: Jentzsch et al. as mentioned in this paper presented an analysis of 522 acute myeloid leukemia (AML) patients who received an allogeneic stem cell transplantation (HSCT) between 2000 and 2021 in first complete remission (CR, 54), second CR (11%), CR with incomplete count recovery (CRi, 13%), or relapsed/refractory (22%) at their center.

Clinical Implications of the FLT3-ITD Allelic Ratio in Acute Myeloid Leukemia in the Context of an Allogeneic Stem Cell Transplantation
Madlen Jentzsch,L. Bischof,Dominic Brauer,Donata Backhaus,J. Ussmann,Georg-Nikolaus Franke,Vladan Vucinic,Uwe Platzbecker,Sebastian Schwind +8 more
TL;DR: In this article , the presence of FLT3-ITD is among the most common molecular aberrations in acute myeloid leukemia (AML), but little is known about how the FLT 3-ITd allelic ratio impacts patient outcomes after HSCT.
Prognostic impact of measurable residual clonal hematopoiesis in acute myeloid leukemia patients after allogeneic hematopoietic stem cell transplantation.
L. Bischof,J. Ussmann,Juliane Grimm,Marius Bill,Dominic Brauer,Donata Backhaus,Lisa Herrmann,Maximilian Merz,Marco Herling,K. Metzeler,Georg-Nikolaus Franke,Vladan Vucinic,U. Platzbecker,Sebastian Schwind,Madlen Jentzsch +14 more
TL;DR: Measurable residual clonal hematopoiesis post-allogeneic hematopoietic stem cell transplantation in acute myeloid leukemia patients is associated with inferior overall survival and relapse-free survival, highlighting its prognostic significance in this patient population.
