Sample preparation is an essential step in analysis, greatly influencing the reliability and accuracy of resulted the time and cost of analysis. Solid-Phase Microextraction (SPME) is a very simple and efficient, solventless sample preparation method, invented by Pawliszyn in 1989. SPME has been widely used in different fields of analytical chemistry since its first applications to environmental and food analysis and is ideally suited for coupling with mass spectrometry (MS). All steps of the conventional liquid-liquid extraction (LLE) such as extraction, concentration, (derivatization) and transfer to the chromatograph are integrated into one step and one device, considerably simplifying the sample preparation procedure. It uses a fused-silica fibre that is coated on the outside with an appropriate stationary phase. The analytes in the sample are directly extracted to the fibre coating. The SPME technique can be routinely used in combination with gas chromatography, high-performance liquid chromatography and capillary electrophoresis and places no restriction on MS. SPME reduces the time necessary for sample preparation, decreases purchase and disposal costs of solvents and can improve detection limits. The SPME technique is ideally suited for MS applications, combining a simple and efficient sample preparation with versatile and sensitive detection. This review summarizes analytical characteristics and variants of the SPME technique and its applications in combination with MS.

This critical review presents information on known and innovative approaches to the manufacture of fibre coatings used in solid-phase microextraction (SPME). The properties, advantages and drawbacks of the different types of commercially available SPME fibre coatings are discussed in detail, as are those of novel types of coatings and the methodologies of their preparation. The applications of fibre coatings in the solid-phase microextraction of a broad spectrum of analytes are analysed, with particular emphasis on the sampling of polar analytes from polar matrices (174 references).

In this article, headspace solid-phase microextraction (HS-SPME) analysis of thiol compounds with both solid and aqueous samples was investigated using N-phenylmaleimide as an in-fiber derivatization reagent. The derivatives were analyzed using GC-MS. The HS-SPME method was executed on an automatic sampler and optimized by studying a variety of factors such as the selection of the fiber, extraction time, temperature, agitation speed, etc. Method validation was carried out by spiking known amounts of thiol compounds into the sample and calculating the concentration by standard addition. The relative standard deviations (RSD) for the majority of the thiols were less than 10%. The developed method resulted in a limit of detection (LOD) in the low microgram per liter for most of the thiols analyzed, which was about one order of magnitude lower than the HS-SPME without derivatization. Finally, the overall method was successfully applied for the determination of thiols in yellow onion, green onion and garlic. Copyright © 2006 John Wiley & Sons, Ltd.
