The concentrations studied ranged from 0.1 to 2.0 mg l-1 for copper, nickel, lead and zinc, and from 0.1 to 10.0 mg l-1 for pentachlorophenol. Exposure time was set to 24 h. Copper and pentachlorophenol turned out to be especially toxic for photosynthetic efficiency (PE) in C. reinhardtii. Copper and pentachlorophenol turned out to be especially toxic for PE in C. reinhardtii. Zinc has been concluded to be moderately toxic while nickel and lead had stimulatory effects on the PE. Because of high variance, motility was not considered a reliable physiological parameter when assessing toxicity of the substances using C. reinhardtii.

This report summarizes research results for the programs that use the facilities at the NIST Center for Neutron Research (NCNR) for the period from October 1996 through September 1997. The programs range from the use of thermal and cold neutron beams for studies of the structure and dynamics of materials to nuclear physics and neutron standards to sample irradiations for activation analysis. The NCNR is a national user facility, which served more than 1300 participants during the year, including scientists and engineers from 57 companies, 92 universities, and 38 other Government agencies in the U.S.A.

Euglena gracilis was exposed for 24 hr to concentrationsof copper, nickel, lead and zinc ranging from 0.1 to 2.0 mg L-1 and to pentachlorophenol ranging from 0.1 to 10.0 mg L-1. Photosynthetic efficiency (PE) was inhibited by increasing concentrations of the heavy metals. Nickel was foundto be the least toxic of the metals to E. gracilis, whilethe other three metals exhibited similar levels of toxicity. Treatment with pentachlorophenol caused unusual responses of the photosynthetic apparatus probably due to the especially high toxicity of this substance. Both cell shape and motility turned out to be insensitive physiological parameters for the toxicity testing of the substances studied.

The IAEA established in 1994 a co-ordinated research programme (CRP) on "Reference Materials for Microanalytical Nuclear Techniques" as part of its efforts to promote and strengthen the use of nuclear analytical technologies in member states with the specific aim of improving the quality of analysis of nuclear, environmental, and biological materials. The objectives of this initiative were: to identify suitable biological reference materials which could serve the needs for quality control in microanalytical techniques; to evaluate existing CRMs for use in microanalytical investigations; to evaluate appropriate sample pretreatment procedures for materials being used for analysis with microanalytical techniques; to identify analytical techniques which can be used for characterisation of homogeneity determination, and to apply such techniques to the characterization of candidate reference materials for use with microanalytical techniques. The CRP lasted for 4 years and seven laboratories and the Agency's Laboratories in Seibersdorf participated. A number of materials including the candidate reference materials IAEA 338 (lichen) and IAEA 413 (single cell algae, elevated level) were evaluated for the distribution of elements such as Cl, K, Ca, Cr, Mn, Fe, Zn, As, Br, Rb, Cd, Hg, and Pb. The results obtained during this CRP suggest that: each element exhibits its characteristic distribution in a matrix described by the "Ingamels' sampling constant" or the "relative homogeneity factor" of Kurfuerst; both concepts are valid over a large range of sample mass used for analysis (from 0.1 μg to around 100 mg); and materials being characterised quantitatively for element homogeneity could be used for the experimental determination of total uncertainty of other analytical techniques. As far as we are aware this is the first time the concept of quantitative characterisation of homogeneity has been applied to potential reference materials and the first demonstration of the feasibility and usefulness of the concept with particular emphasis on enhancing quality control opportunities for microanalytical techniques.

Assessment of element-specific homogeneity in reference materials using microanalytical techniques.

Three batches of single cell green algal biomass have been produced as model substances for green biomass reference materials with the aim of arriving at three different concentration levels for selected heavy metals in the same matrix. The green algal biomass is produced in a water based culture medium in open and closed bioreactors. An outdoor open bioreactor technology was used to produce a batch of “natural level” algae as well as a batch with elevated levels of As, Cd, Cr, Hg, Ni, and Pb. A photoreactor constructed without metal parts and employing artificial light sources was used in a clean air environment with high purity chemicals to reduce heavy metal concentrations in the third batch of algal biomass. For the above elements, the depleted level material, with exception of Hg, showed about one order of magnitude lower concentration than the natural level, while concentrations in the enriched material were up to four orders of magnitude higher. Analysts can use these materials to sustain verification of calibration and analytical quality over the entire operational range of the analytical work.
