Network pharmacology is an emerging area of systematic drug research that attempts to understand drug actions and interactions with multiple targets. Network pharmacology has changed the paradigm from 'one-target one-drug' to highly potent 'multi-target drug'. Despite that, this synergistic approach is currently facing many challenges particularly mining effective information such as drug targets, mechanism of action, and drug and organism interaction from massive, heterogeneous data. To overcome bottlenecks in multi-target drug discovery, computational algorithms are highly welcomed by scientific community. Machine learning (ML) and especially its subfield deep learning (DL) have seen impressive advances. Techniques developed within these fields are now able to analyze and learn from huge amounts of data in disparate formats. In terms of network pharmacology, ML can improve discovery and decision making from big data. Opportunities to apply ML occur in all stages of network pharmacology research. Examples include screening of biologically active small molecules, target identification, metabolic pathways identification, protein-protein interaction network analysis, hub gene analysis and finding binding affinity between compounds and target proteins. This review summarizes the premier algorithmic concepts of ML in network pharmacology and forecasts future opportunities, potential applications as well as several remaining challenges of implementing ML in network pharmacology. To our knowledge, this study provides the first comprehensive assessment of ML approaches in network pharmacology, and we hope that it encourages additional efforts toward the development and acceptance of network pharmacology in the pharmaceutical industry.

Machine learning for synergistic network pharmacology: a comprehensive overview

Recent developments in the Internet of Things (IoT) and various communication technologies have reshaped numerous application areas. Nowadays, IoT is assimilated into various medical devices and equipment, leading to the progression of the Internet of Medical Things (IoMT). Therefore, various IoMT-based healthcare applications are deployed and used in the day-to-day scenario. Traditionally, machine learning (ML) models use centralized data compilation and learning that is impractical in pragmatic healthcare frameworks due to rising privacy and data security issues. Federated Learning (FL) has been observed as a developing distributed collective paradigm, the most appropriate for modern healthcare framework, that manages various stakeholders (e.g., patients, hospitals, laboratories, etc.) to carry out training of the models without the actual exchange of sensitive medical data. Consequently, in this work, the authors present an exhaustive survey on the security of FL-based IoMT applications in smart healthcare frameworks. First, the authors introduced IoMT devices, their types, applications, datasets, and the IoMT security framework in detail. Subsequently, the concept of FL, its application domains, and various tools used to develop FL applications are discussed. The significant contribution of FL in deploying secure IoMT systems is presented by focusing on FL-based IoMT applications, patents, real-world FL-based healthcare projects, and datasets. A comparison of FL-based security techniques with other schemes in the smart healthcare ecosystem is also presented. Finally, the authors discussed the challenges faced and potential future research recommendations to deploy secure FL-based IoMT applications in smart healthcare frameworks.

With the advent of the Internet of Things (IoT), artificial intelligence (AI), machine learning (ML), and deep learning (DL) algorithms, the landscape of data-driven medical applications has emerged as a promising avenue for designing robust and scalable diagnostic and prognostic models from medical data. This has gained a lot of attention from both academia and industry, leading to significant improvements in healthcare quality. However, the adoption of AI-driven medical applications still faces tough challenges, including meeting security, privacy, and Quality-of-Service (QoS) standards. Recent developments in federated learning (FL) have made it possible to train complex machine-learned models in a distributed manner and have become an active research domain, particularly processing the medical data at the edge of the network in a decentralized way to preserve privacy and address security concerns. To this end, in this article, we explore the present and future of FL technology in medical applications where data sharing is a significant challenge. We delve into the current research trends and their outcomes, unraveling the complexities of designing reliable and scalable FL models. This article outlines the fundamental statistical issues in FL, tackles device-related problems, addresses security challenges, and navigates the complexity of privacy concerns, all while highlighting its transformative potential in the medical field. Our study primarily focuses on medical applications of FL, particularly in the context of global cancer diagnosis. We highlight the potential of FL to enable computer-aided diagnosis tools that address this challenge with greater effectiveness than traditional data-driven methods. Recent literature has shown that FL models are robust and generalize well to new data, which is essential for medical applications. We hope that this comprehensive review will serve as a checkpoint for the field, summarizing the current state of the art and identifying open problems and future research directions. 

Federated Learning for Medical Applications: A Taxonomy, Current Trends, Challenges, and Future Research Directions

Since its inception in 2016, federated learning has evolved into a highly promising decentral-ized machine learning approach, facilitating collaborative model training across numerous devices while ensuring data privacy. This survey paper offers an exhaustive and systematic review of federated learning, emphasizing its categories, challenges, aggregation techniques, and associated development tools. To start, we outline our research strategy used for this survey and evaluate other existing reviews related to federated learning. We initiate the discussion, about federated learning concepts, with a detailed examination of the primary challenges inherent in federated learning, including communication overhead, device and data heterogeneity, and data privacy issues. Subsequently, we scrutinize and classify various aggregation techniques designed to mitigate these challenges, such as federated averaging, secure aggregation, and strategies leveraging clustering and optimization methodologies. Furthermore, we delve into the exploration of cutting-edge development tools and frameworks that expedite efficient implementations of federated learning. Through our review, we aspire to provide a holistic understanding of the federated learning landscape, thereby setting the stage for future investigations, advancements, and practical implementations in this prosperous field. 

A systematic review of federated learning: Challenges, aggregation methods, and development tools

Machine learning is expected to fuel significant improvements in medical care. To ensure that fundamental principles such as beneficence, respect for human autonomy, prevention of harm, justice, privacy, and transparency are respected, medical machine learning systems must be developed responsibly. Many high-level declarations of ethical principles have been put forth for this purpose, but there is a severe lack of technical guidelines explicating the practical consequences for medical machine learning. Similarly, there is currently considerable uncertainty regarding the exact regulatory requirements placed upon medical machine learning systems. This survey provides an overview of the technical and procedural challenges involved in creating medical machine learning systems responsibly and in conformity with existing regulations, as well as possible solutions to address these challenges. First, a brief review of existing regulations affecting medical machine learning is provided, showing that properties such as safety, robustness, reliability, privacy, security, transparency, explainability, and nondiscrimination are all demanded already by existing law and regulations - albeit, in many cases, to an uncertain degree. Next, the key technical obstacles to achieving these desirable properties are discussed, as well as important techniques to overcome these obstacles in the medical context. We notice that distribution shift, spurious correlations, model underspecification, uncertainty quantification, and data scarcity represent severe challenges in the medical context. Promising solution approaches include the use of large and representative datasets and federated learning as a means to that end, the careful exploitation of domain knowledge, the use of inherently transparent models, comprehensive out-of-distribution model testing and verification, as well as algorithmic impact assessments.

Responsible and Regulatory Conform Machine Learning for Medicine: A Survey of Challenges and Solutions

Federated learning (FL) is a computational paradigm that enables organizations to collaborate on machine learning (ML) projects without sharing sensitive data, such as, patient records, financial data, or classified secrets. Open Federated Learning (OpenFL this https URL) is an open-source framework for training ML algorithms using the data-private collaborative learning paradigm of FL. OpenFL works with training pipelines built with both TensorFlow and PyTorch, and can be easily extended to other ML and deep learning frameworks. Here, we summarize the motivation and development characteristics of OpenFL, with the intention of facilitating its application to existing ML model training in a production environment. Finally, we describe the first use of the OpenFL framework to train consensus ML models in a consortium of international healthcare organizations, as well as how it facilitates the first computational competition on FL.
