Biodiversity is a challenging field of research. Approaches are manifold and mostly cover few aspects of the total wealth of phenomena only. The mapping of the vascular plants and the inventory of Basidiomycota in Bavaria are projects to be mentioned in this context as one part of the commitment of the author. In the following article the author describes further activities during his lifework in regard A. In fungi the investigations include breeding systems, isolation barriers, polyploidy, pigment patterns and DNA phylogeny. The pigment patterning in Boletales correlates well with the phylogeny as revealed by DNA analysis of selected gene sections.

Progress in Botany

The endophytic fungus Penicillium restrictum was isolated from the stems of a milk thistle (Silybum marianum) plant. In culture, the fungus produced distinct red guttates, which have been virtually uninvestigated, particularly from the standpoint of chemistry. Hence, this study examined the chemical mycology of P. restrictum and, in doing so, uncovered a series of both known and new polyhydroxyanthraquinones (1–9). These compounds were quorum sensing inhibitors in a clinical isolate of methicillin-resistant Staphylococcus aureus (MRSA), with IC50 values ranging from 8 to 120 μM, suggesting antivirulence potential for the compounds. Polyhydroxyanthraquinones as Quorum Sensing Inhibitors from the Guttates of Penicillium restrictum and Their Analysis by Desorption Electrospray Ionization Mass Spectrometry

Development and applications of destruxins: a review.

Pseudogymnoascus destructans is the causative agent of an emerging infectious disease that threatens populations of several North American bat species. The fungal disease was first observed in 2006 and has since caused the death of nearly six million bats. The disease, commonly known as white-nose syndrome, is characterized by a cutaneous infection with P. destructans causing erosions and ulcers in the skin of nose, ears and/or wings of bats. Previous studies based on sequences from eight loci have found that isolates of P. destructans from bats in the US all belong to one multilocus genotype. Using the same multilocus sequence typing method, we found that isolates from eastern and central Canada also had the same genotype as those from the US, consistent with the clonal expansion of P. destructans into Canada. However, our PCR fingerprinting revealed that among the 112 North American isolates we analyzed, three, all from Canada, showed minor genetic variation. Furthermore, we found significant variations among isolates in mycelial growth rate; the production of mycelial exudates; and pigment production and diffusion into agar media. These phenotypic differences were influenced by culture medium and incubation temperature, indicating significant variation in environmental condition - dependent phenotypic expression among isolates of the clonal P. destructans genotype in North America.

Clonal expansion of the Pseudogymnoascus destructans genotype in North America is accompanied by significant variation in phenotypic expression.

Nutritional conditions causing droplet exudation by Metarhizium anisopliae var. anisopliae were studied. Exudation in droplets occurred only on media with more than one carbon source and was highly dependent on the ratio of a well metabolized sugar such as trehalose and a nonpreferred sugar, in particular arabinose. Exuded droplets contained destruxin A, B and E in concentrations similar to those on submerged culture on Czapek Dox medium with equivalent C : N ratios but was clearly less than previously reported on standard Czapek Dox or Sabouraud dextrose broth. Destruxins also were found in agar samples from directly below mycelium and from up to 2 cm from the colony edge. Exudates retrieved from different media were proven to have Pr1 protease-related enzyme activity. Additional HPLC analysis indicated that droplets from diverse media did not differ in their sugar and acid content. A hypothesis is presented regarding the trigger for guttation in Metarhizium during growth under these conditions.

Formation of exudate droplets by Metarhizium anisopliae and the presence of destruxins

Destruxins (dtx) A, B, and E, showing a variety of biological activities, are the main toxic secondary metabolites of the entomopathogenous ascomycete Metarhizium anisopliae Bipesco 5, a widely used biocontrol production strain. Dynamics of dtx biosynthesis were monitored during liquid fermentation in a chemically defined medium. During shake flask cultivation with excess carbon, nitrogen and phosphate, approximately 50, 20, and 100 mg l–1 dtx A, B, and E were produced after 12 d. Destruxins were produced during exponential growth phase and in the stationary phase. Carbon exhaustion in the culture broth was demonstrated to affect destruxin production to a minor degree: Absolute dtx amounts in the liquid increased also after glucose exhaustion; dtx amounts referred to biomass increased further evidently in shake flasks or slightly in bioreactor experiments after carbon limitation occurred. Contrarily, nitrogen exhaustion resulted in an evident decline in dtx amounts referred to biomass. Absolute amounts in the culture broth, however, still increased slightly the following four days in bioreactor experiments. From this we conclude that dtx production is highly influenced by nitrogen availability.
