Effect of strawberry vein banding virus and strawberry mottle virus co-infection on the growth and development of strawberry

The vegetative propagation of strawberries favors transmission of systemic pathogens, such as viruses, which are one of the main yield-limiting factors for this crop. More than 20 viruses have been described as infecting this species; one of the most frequent is the  Strawberry mottle  virus (SMoV), which is responsible for significant economic losses. SMoV is usually detected by reverse transcription polymerase chain reaction (RT-PCR), given that serum is not available for serological tests. In this study, a non-radioactive molecular probe was developed for SMoV detection. The cDNA was synthesized by RT-PCR using specific primers designed from the 3'UTR region of the viral genome. The cloned cDNA segment was labeled and used as a probe. Six RNA extraction protocols were evaluated, and the modified cetyltrimethylammonium bromide (CTAB) method showed the highest sensitivity level. Leaves at different phenological stages and petioles were evaluated; the highest reaction was observed in old leaves and in petioles.

Development of a non-radioactive molecular hybridization probe for detecting Strawberry mottle virus in strawberry

Strawberry mottle virus (SMoV) is one of the viral pathogens affecting strawberries (Fragaria spp.) production severely worldwide. Currently, reverse transcription-polymerase chain reaction (RT-PCR) is frequently used to detect SMoV, which is sometimes not successful because of unsatisfied nucleic acid quality and the contamination of secondary metabolites resulting in poor PCR amplification. In the study, we developed a digoxigenin-labelled cDNA probe for SMoV detection by dot-blot hybridization. Total nucleic acid was extracted with modified CTAB method from strawberry plants infected by SMoV, the SMoV specific fragment was amplified with RT-PCR, which was cloned and sequenced. The cloned fragment was then labeled with digoxigenin by PCR, which is used as probe in dot-blot hybridization to test the samples infected by SMoV. The result showed that dot-blot hybridization had the same result of RT-PCR in testing SMoV. Our research provided an important alternative to RT-PCR technique for detecting SMoV. To our knowledge, this is the first report for detection of SMoV by cDNA probe. In addition, it is a potential method to test other strawberry viruses.

Detection of Strawberry mottle virus by molecular hybridization

Many microorganisms in soil are able to transform insoluble form of phosphorus to an accessible soluble one, contributing to plant nutrition as plant growth-promoting microorganisms. The objective of this work was to isolate, screen and evaluate the phosphate-solubilizing activity of microorganisms in soybean rhizosphere soil, and select potential microbial inoculants. These microorganisms were selected based on the phosphate-solubilizing efficiency in a modified Pikovskaya’s liquid medium culture. The isolates were identified based on the morphological characteristic and nucleotide sequence data from the 16S ribosomal DNA (rDNA) for bacteria. Strain BAâÂ¢, identified as Bacillus megaterium, is the most effective one for solubilization of phosphate in medium. Taken together, these results demonstrate that strain BAâÂ¢ has the ability to convert nonavailable forms of phosphorus into plant-available forms, therefore, it has a great potential to develop as a bio-fertilizer to enhance soil fertility and promote plant growth.
