The ATSDR toxicological profile succinctly characterizes the toxicologic and adverse health effects infonnation for these toxic substances described therein. Other pertinent literature is also presented, but is described in less detail than the key studies. The profile is not intended to be an exhaustive document; however, more comprehensive sources of specialty information are referenced. The focus of the profiles is on health and toxicologic infonnation; therefore, each toxicological profile begins with a public health statement that describes, in nontechnical language, a substance's relevant toxicological properties. Following the public health statement is infonnation concerning levels of significant human exposure and, where known, significant health effects. The adequacy of infonnation to detennine a substance's health effects is described in a health effects summary. Data needs that are of significance to protection of public health are identified by ATSDR. Each profile includes the following: (A) The examination, summary, and interpretation of available toxicologic infonnation and epidemiologic evaluations on a toxic substance to ascertain the levels of significant human exposure for the substance and the associated acute, subacute, and chronic health effects; (B) A determination of whether adequate infonnation on the health effects of each substance is available or in the process of development to detennine levels of exposure that present a significant risk to human health of acute, subacute, and chronic health effects; and (C) Where appropriate, identification of toxicologic testing needed to identifY the types or levels of exposure that may present significant risk of adverse health effects in humans. Human exposure to environmental phenols can be assessed by measuring the urinary concentrations of these compounds or their metabolites Total concentrations, which include both free and conjugated (ie, glucuronide and sulfated) species, are usually reported Because conjugation may reduce the potential bioactivity of the compounds, measuring separately both the concentrations of free and conjugated species can be of interest Data on the stability of these conjugated species in urine is critical if the concentrations of free and conjugated species are to be compared Over a period of 6 months, we investigated the stability of the urinary conjugates of eight environmental phenols (bisphenol A, 2-hydroxy-4-metoxybenzophenone or benzophenone-3, triclosan, 2,5-dichlorophenol, methyl paraben, ethyl paraben, propyl paraben, and butyl paraben) at three storage conditions (room temperature, 4 degrees C, and -70 degrees C) After collection, conjugated species appeared to be stable for at least 7 days when the urine was stored at 4 degrees C, and for at least 180 days at -70 degrees C By contrast, some of the environmental phenol conjugates commenced to degrade within 24 h after collection when the urine was stored at room temperature although the total concentrations remained relatively constant for at least 30 days These results suggest that if the concentrations of free and conjugated species will be used for exposure assessment purposes, urine specimens collected for analysis of environmental phenols should be kept at room temperature for the shortest possible time after collection

Baur, Levels of 8-hydroxy-2'-deoxyguanosine in DNA of white blood cells from workers highly exposed to asbestos in Germany, Mutat. Res. (2000) submitted] we described significant increases (p 0.05) than that for asbestos-exposed patients without tumours. The formation of 8-OHdG adduct levels in WBC DNA of patients with hematopoietic cancer, chondrosarcomas and multiform glioblastomas was not significantly higher than that found in the control group (p>0.05). Our results support the hypothesis that oxidative DNA damage in man caused by asbestos fibers plays a role in the formation of malignant tumours.

1 Chemical risk assessment integrates the identification of hazards and the human exposure levels which can be established from external and/or internal exposure data.2 The availability of biomonitoring and metabolism animal data, the skin penetration ability, and the existence of atmospheric threshold limit values were examined for twelve substances of the European first list of priority existing substances. This investigation was focused on workplace exposures and on urinary biomarkers of exposure. Appropriate biomonitoring data appeared to be available for two substances: styrene and trichloroethylene. Some biomonitoring research has been conducted on acrylonitrile, buta-1,3-diene, cyclohexane, 1,4-dichlorobenzene, hydrogen fluoride, 2-(2-methoxyethoxy)ethanol, however additional studies could be usefully carried out.
