The role of complete response in multiple myeloma.
TL;DR: Although CR achievement is necessary in patients with poor-risk disease, it might not be as critical for long survival in more indolent MM, and it is possible to further improve the depth of response and the outcome by continuing treatment after CR achievement.
Achievement of VGPR to induction therapy is an important prognostic factor for longer PFS in the IFM 2005-01 trial
Philippe Moreau,Michel Attal,Brigitte Pegourie,Lucie Planche,Cyrille Hulin,Thierry Facon,Anne-Marie Stoppa,Jean-Gabriel Fuzibet,Bernard Grosbois,Chantal Doyen,Nicolas Ketterer,Catherine Sebban,Brigitte Kolb,Carine Chaleteix,Mamoun Dib,Laurent Voillat,Jean Fontan,Laurent Garderet,Jérôme Jaubert,Claire Mathiot,Dixie Lee Esseltine,Hervé Avet-Loiseau,Jean-Luc Harousseau +22 more
TL;DR: Progression-free survival was significantly improved with bortezomib-dexamethasone induction therapy in patients with poor-risk cytogenetics and ISS stages 2 and 3 compared with vincristine-adriamycin-d examethAsone, and achievement of at least VGPR after induction was of major importance.
A practical guide for mutational signature analysis in hematological malignancies
Francesco Maura,Andrea Degasperi,Ferran Nadeu,Daniel Leongamornlert,Helen Davies,Helen Davies,Helen Davies,Luiza Moore,Romina Royo,Bachisio Ziccheddu,Xose S. Puente,Hervé Avet-Loiseau,Peter J. Campbell,Serena Nik-Zainal,Serena Nik-Zainal,Serena Nik-Zainal,Elias Campo,Nikhil C. Munshi,Niccolo Bolli +18 more
TL;DR: Using whole-genome sequencing of multiple myeloma, chronic lymphocytic leukemia and acute myeloid leukemia, the performance of public signature analysis tools is compared and a framework for the generation of accurate and reproducible signature data is suggested.
Biological and prognostic impact of APOBEC-induced mutations in the spectrum of plasma cell dyscrasias and multiple myeloma cell lines.
Francesco Maura,Mia Petljak,Marta Lionetti,Ingrid Cifola,Winnie S. Liang,Eva Pinatel,Ludmil B. Alexandrov,Anthony Fullam,Inigo Martincorena,Kevin J. Dawson,Nicos Angelopoulos,Mehmet Kemal Samur,Raphael Szalat,Jorge Zamora,Patrick S. Munshi,Niccolo Bolli,Niccolo Bolli +26 more
TL;DR: Biological and prognostic impact of APOBEC-induced mutations in the spectrum of plasma cell dyscrasias and multiple myeloma cell lines is studied.
A DNA target-enrichment approach to detect mutations, copy number changes and immunoglobulin translocations in multiple myeloma
Niccolo Bolli,Niccolo Bolli,Y. Li,V. Sathiaseelan,Keiran Raine,David T. Jones,Peter Ganly,F. Cocito,Graham R. Bignell,Michael A Chapman,Adam S. Sperling,KC Anderson,Hervé Avet-Loiseau,Stephane Minvielle,Purdey J. Campbell,Nikhil C. Munshi +15 more
TL;DR: A target-enrichment strategy followed by next-generation sequencing (NGS) is developed to streamline simultaneous analysis of gene mutations, copy number changes and immunoglobulin heavy chain translocations in MM in a high-throughput manner and validated it in a panel of cell lines.
