A direct projection from the retina to the hypothalamus was demonstrated in the rat. Following injection of tritiated leucine or proline into the posterior chamber of the eye, labelled protein was shown autoradiographically in the suprachiasmatic nuclei of the medial hypothalamus, both ipsilateral and contralateral to the injected eye. The labelling of the nucleus was heaviest in its ventral portion but extended throughout the nucleus. No evidence for a projection to the supraoptic nucleus or any other hypothalamic nucleus was observed. All of the known terminal nuclei of the primary and accessory optic tracts were heavily labelled. The projection to the suprachiasmatic nucleus could not be clearly confirmed in material prepared using the Fink-Heimer method for the demonstration of degenerating axon terminals. Electron microscopic study of the suprachiasmatic nucleus following orbital enucleation showed degenerating endings making synaptic contacts with small dendrites of the suprachiasmatic nucleus cells. These first appeared at three days after operation and were nearly gone by seven days. Thus, the retinohypothalamic tract in the rat appears to arise from the ganglion cells of the retina and to terminate on the smaller dendritic branches of the neurons of the suprachiasmatic nucleus.

Publisher Summary The accessory optic system (AOS) is a visual system organized in vestibular coordinates. Recordings from neurons in the three terminal nuclei of the AOS have revealed that this visual system processes information about the speed and direction of movement of large parts of the visual world. Best modulation of terminal nuclei neurons requires use of large textured stimuli and occurs at low speeds with peak sensitivity at 0.1–1 degree/sec. For medial terminal nucleus and lateral terminal nucleus, the preferred directions in space are vertical, but with a posterior component. For dorsal terminal nucleus, preference is for horizontal movement from posterior to anterior. Taken collectively, the preferred directions for terminal nuclei neurons define three directions in visual space: anterior, up with a posterior component, and down with a posterior component. The speed and direction selectivity of neurons of the accessory optic terminal nuclei are identical to those of the on-direction selective class of rabbit retinal ganglion cells and to those of visually driven climbing fibers in the cerebellar flocculus. It is most likely that the AOS serves to signal self-motion; the sensitivity of AOS neurons to slow speeds makes the AOS an ideal complement to the vestibular system.

The accessory optic system and its relation to the vestibulocerebellum.

Retinal projections and visual thalamo-cortical connections were studied in the subterranean mole rat, belonging to the superspecies Spalax ehrenbergi, by anterograde and retrograde tracing techniques. Quantitative image analysis was used to estimate the relative density and distribution of retinal input to different primary visual nuclei. The visual system of Spalax presents a mosaic of both regressive and progressive morphological features. Following intraocular injections of horseradish peroxidase conjugates, the retina was found to project bilaterally to all visual structures described as receiving retinal afferents in non-fossorial rodents. Structures involved in form analysis and visually guided behaviors are reduced in size by more than 90%, receive a sparse retinal innervation, and are cytoarchitecturally poorly differentiated. The dorsal lateral geniculate nucleus, as defined by cyto- and myelo-architecture, cytochrome oxidase, and acetylcholinesterase distribution as well as by afferent and efferent connections, consists of a narrow sheet 3-5 neurons thick, in the dorsal thalamus. Connections with visual cortex are topographically organized but multiple cortical injections result in widespread and overlapping distributions of geniculate neurons, thus indicating that the cortical map of visual space is imprecise. The superficial layers of the superior colliculus are collapsed to a single layer, and the diffuse ipsilateral distribution of retinal afferents also suggests a lack of precise retinotopic relations. In the pretectum, both the olivary pretectal nucleus and the nucleus of the optic tract could be identified as receiving ipsilateral and contralateral retinal projections. The ventral lateral geniculate nucleus is also bilaterally innervated, but distinct subdivisions of this nucleus or the intergeniculate leaflet could not be distinguished. The retina sends a sparse projection to the dorsal and lateral terminal nuclei of the accessory optic system. The medial terminal nucleus is not present. In contrast to the above, structures of the "non-image forming" visual pathway involved in photoperiodic perception are well developed in Spalax. The suprachiasmatic nucleus receives a bilateral projection from the retina and the absolute size, cytoarchitecture, density, and distribution of retinal afferents in Spalax are comparable with those of other rodents. A relatively hypertrophied retinal projection is observed in the bed nucleus of the stria terminalis. Other regions which receive sparse visual input include the lateral and anterior hypothalamic areas, the retrochiasmatic region, the sub-paraventricular zone, the paraventricular hypothalamic nucleus, the anteroventral and anterodorsal nuclei, the lateral habenula, the mediodorsal nucleus, and the basal telencephalon.(ABSTRACT TRUNCATED AT 400 WORDS)

The efferent projections of the nBOR complex, have been studied with both anterograde autoradiographic and retrograde horseradish peroxidase (HRP) techniques. The nBOR complex includes three distinct subdivisions: the nucleus of the basal optic root (nBOR), the nBOR pars dorsalis (nBORd) and the nBOR pars lateralis (nBORl).



Unilateral injections of 3H-leucine or 3H-proline/3H-leucine mixtures into the nBOR complex have demonstrated prominent bilateral projections upon (1) the vestibulocerebellum, (2) the inferior olivary complex, (3) the oculomotor nuclear complex, (4) the nucleus interstitialis, contralateral projections upon (5) the contralateral nBOR complex and ipsilateral projections upon (6) a pretectal nucleus, the nucleus lentiformis mesencephali, pars magnocellularis.



Unilateral injections of HRP confined to folia IXc, d and paraflocculus of the cerebellum, the contralateral nBOR complex or the nucleus lentiformis mesencephali resulted in retrograde labeling of predominantly medium and large size cells within the entire nBOR complex. Unilateral injections of HRP within the inferior olive resulted in retrograde labeling of small, spindle-shaped cells within nBOR and nBORd. Unilateral injections of the oculomotor complex which included the trochlear nucleus resulted in retrograde labeling of small cells within the ipsilateral nBORd and predominantly medium and large cells in the contralateral nBOR.



The displaced ganglion cells of the retina give rise to a prominent and distinct projection upon the nBOR complex (Karten et al., '77). The nBOR complex in turn projects upon the oculomotor nuclear complex, the nucleus interstitialis and the vestibulocerebellum, regions which have been implicated in oculomotor function. These findings strongly suggest that the displaced ganglion cells and the accessory optic system have a major influence upon oculomotor reflexes including eye and head movements.

The accessory optic system (AOS) is formed by a series of terminal nuclei receiving direct visual information from the retina via one or more accessory optic tracts. In addition to the retinal input, derived from ganglion cells that characteristically have large receptive fields, are direction-selective, and have a preference for slow moving stimuli, there are now well-characterized afferent connections with a key pretectal nucleus (nucleus of the optic tract) and the ventral lateral geniculate nucleus. The efferent connections of the AOS are robust, targeting brainstem and other structures in support of visual-oculomotor events such as optokinetic nystagmus and visual-vestibular interaction. This chapter reviews the newer experimental findings while including older data concerning the structural and functional organization of the AOS. We then consider the ontogeny and phylogeny of the AOS and include a discussion of similarities and differences in the anatomical organization of the AOS in nonmammalian and mammalian species. This is followed by sections dealing with retinal and cerebral cortical afferents to the AOS nuclei, interneuronal connections of AOS neurons, and the efferents of the AOS nuclei. We conclude with a section on Functional Considerations dealing with the issues of the response properties of AOS neurons, lesion and metabolic studies, and the AOS and spatial cognition.
