Co-Delivery of Ciprofloxacin and Colistin in Liposomal Formulations with Enhanced In Vitro Antimicrobial Activities against Multidrug Resistant Pseudomonas aeruginosa
Shaoning Wang,Shaoning Wang,Shihui Yu,Shihui Yu,Yu-Wei Lin,Peizhi Zou,Guihong Chai,Heidi H. Yu,Hasini Wickremasinghe,Nivedita Shetty,Junhong Ling,Junhong Ling,Jian Li,Qi Tony Zhou +13 more
TL;DR: Liposomal formulations of two synergistic antibiotics was promising against multidrug resistant P. aeruginosa infections as well as the cytotoxicity study using A549 cells showed the liposomal formulation is as non-toxic as the drug solutions.
Metabolic Perturbations Caused by the Over-Expression of mcr-1 in Escherichia coli.
Yiyun Liu,Yan Zhu,Hasini Wickremasinghe,Phillip J. Bergen,Jing Lu,Xiao Qing Zhu,Qiao Li Zhou,Mohammad A. K. Azad,Sue C Nang,Mei-Ling Han,Tao Lei,Jian Li,Jian-Hua Liu +12 more
TL;DR: It is indicated that over-expression of mcr-1 results in global metabolic perturbations that mainly involve disruption to the bacterial membrane, pentose phosphate pathway as well as pantothenate and CoA biosynthesis.
Comparative metabolomics revealed key pathways associated with the synergistic killing of multidrug-resistant Klebsiella pneumoniae by a bacteriophage-polymyxin combination
Mei-Ling Han,Sue C Nang,Yu-Wei Lin,Yan Zhu,Heidi H. Yu,Hasini Wickremasinghe,Christopher K. Barlow,Darren J. Creek,Simon Crawford,Gauri G. Rao,Chongshan Dai,Jeremy J. Barr,Kim Sang Chan,Robert Turner Schooley,Tony Velkov,Jian Li +15 more
TL;DR: In this paper , the combination of polymyxin B with a novel phage pK8 against an mcr-1-carrying polymyinxin-resistant clinical isolate Kp II-503 was investigated.

A novel chemical biology and computational approach to expedite the discovery of new-generation polymyxins against life-threatening Acinetobacter baumannii
Xukai Jiang,Xukai Jiang,Nitin A. Patil,Mohammad Abul Kalam Azad,Hasini Wickremasinghe,Heidi Yu,Jinxin Zhao,Xinru Zhang,Mengyao Li,Bin Gong,Lin Wan,Wendong Ma,Philip E. Thompson,Kai Yang,Bing Yuan,Falk Schreiber,Falk Schreiber,Lushan Wang,Tony Velkov,Kade D. Roberts,Jian Li +20 more
TL;DR: This study systematically investigated the structure–activity relationship (SAR) of polymyxins using a quantitative lipidomics-informed outer membrane (OM) model of Acinetobacter baumannii and a series of chemically synthesizedpolymyxin analogs, and deciphered how each residue of the poly myxin molecule modulated its conformational folding and specific interactions with the bacterial OM.
Polymyxin dose tunes the evolutionary dynamics of resistance in multidrug-resistant Acinetobacter baumannii.
Jinxin Zhao,Yan Zhu,Yu-Wei Lin,Heidi H. Yu,Hasini Wickremasinghe,Jiru Han,Tony Velkov,Michael J. McDonald,Jian Li +8 more
TL;DR: In this paper , the authors investigated varying concentrations of the last-resort polymyxins on the evolution of resistance in Acinetobacter baumannii and found that the strength of the selective pressure applied by polymyixin tunes the dynamics of genetic variants within the population, leading to different evolutionary outcomes for the degree, cost and reversibility of resistance.
