Enzymes are extensively used in biotechnological processes in several areas of industry. They are sustainable and safe, and their specificity is another characteristic that improves the performance in the process. Among enzymes, lipase is relevant due to the ability to play different roles in the industry and the possibility of collecting them from microbial sources that are found in industrial residues. This can reduce the costs of enzyme production. In relation to that, lipase immobilization is an interesting process that allows the enzymes to be reused and improves enzyme robustness. Among them, the cross-linked enzyme aggregates (CLEAs) methodology is attractive due to its simplicity, low cost (given the absence of support), and greater interaction with the substrate. Thus, in this review, we discussed the potential of lipase. We reviewed the traditional and new sources of obtaining lipases, along with the ways of improving production, activity, and application in the industry.

Lipases: Sources of Acquisition, Ways of Production, and Recent Applications

Isolation and identification of a newly isolated lipase-producing bacteria (Acinetobacter baumannii RMUTT3S8-2) from oily wastewater treatment pond in a poultry processing factory and its optimum lipase production

Acinetobacter indicus strain UBT1 has shown efficient lipase (243 U ml−1) and biosurfactant (61.1% E24% emulsification and surface tension reduction to 37.7 mN m−1) production capabilities using agro-industrial waste as sole carbon source. We report here the draft genome sequence of A. indicus strain UBT1 having genome size of 2.97 Mb with 45.90% GC content. Total 2721 coding genes were predicted using National Center for Biotechnology Information-Prokaryotic Genome Annotation Pipeline (NCBI-PGAP). The whole genome shotgun project sequence data are accessible through NCBI Gene Bank under accession no. JABFOI000000000. PGAP annotation revealed the presence of the triacylglycerol lipase, phospholipase etc., that circuitously confers the oil consumption competency to the strain UBT1. Rapid Annotation using the Subsystem Technology (RAST) server used for mapping the genes to the subsystem resulted in 278 subsystem with 30% subsystem coverage. The draft genome data can be used to exploit the A. indicus strain UBT1 for its advance biotechnological application and also for further comparative genomic studies.

Draft Genome Analysis of Acinetobacter indicus Strain UBT1, an Efficient Lipase and Biosurfactant Producer.

The rapid increase in population and industrialization has brought an adverse effect on the environment because of the excessive use of its resources. Our environment is now over burdened with several pollutants like heavy metals, volatile organic compounds (VOCs), synthetic dyes, pesticides, etc. In order to reduce their harmful consequences on the ecosystem, degradation and remediation of these pollutants is essential. Numerous physical, chemical, or biological mechanisms have been discovered to eliminate their toxic impact on the environment. Bioremediation is a mechanism in which contaminants are transformed into nonhazardous compounds with the help of bioremediators, i.e., fungi, bacteria, algae, etc. Mycoremediation is a clean-green technology which is a noninvasive, easy, safer, and cheaper solution. Biosorption, bioaccumulation, and biodegradation are the three principal mechanisms of the mycoremediation of pollutants in question. Fungal groups are hyperaccumulators of toxicants in the fruiting bodies or mycelia and cover the surface of the pollutant with white thread forming a huge mass of mycelium. They take its nutrients from the contaminated substances like keratin, cellulose, fats, chitin, and breakdown into simple molecules. Fungi show diversified metabolism because of the presence of numerous enzymes like peroxidase, laccase, monooxygenases, transferases, etc., which degrade the toxic substances. In this chapter, we have discussed potential of fungal-based remediation strategy of several environmental pollutants including VOCs, heavy meals, synthetic dyes, and municipal solid wastes. It further describes fungal diversity exploited for remediation, mechanisms of action, recent advancements, and factors affecting their bioremediation potential in detail. 

Fungal diversity in the bioremediation of toxic effluents

Lipase, as a handy hydrolytic enzyme, is gaining importance in environmental management like oil and fat-containing wastewater treatment. In this study, we have focused on cost-effective production of lipase by Pseudomonas aeruginosa UKHL1 and its applicability in oily wastewater treatment. Plackett-Burman randomized factorial design with 20 experiments recognized ammonium oxalate, peptone, yeast extract, coconut oil, NaCl, and CaCl2 as significant factors among the screened 13 factors by one factor at a time approach (OFAT). Further fine-tuning of three most significant media components (ammonium oxalate (0.01% w/v), peptone (0.246% w/v) and coconut oil (0.1% v/v)) was done by Central Composite Design experiment of Response Surface Method with 20 runs. Lipase production by P. aeruginosa UKHL1 was improved 9.4 and 3.91-fold after 24 h and 36 h, respectively under optimized conditions. Strain in this study also showed good tolerance to iso-octane and n-hexadecane solvents having higher log_P value. P. aeruginosa UKHL1 strain degraded 37% oil after 72 h in an experimental setup under laboratory condition.

The world’s worst offenders in terms of pollution are related to the xenobiotic organopollutants, often toxic and recalcitrant in nature. They have complex aromatic structures which are persistent and recalcitrant, for example, xenobiotics such as phenols, plastics, hydrocarbons, paints, synthetic dyes, pesticides, insecticides, paper and pulp mill effluents, and pharmaceuticals. Applications of physicochemical methods are quite expensive to operate; moreover, they introduce secondary pollutants during the “remediation” process. The white rot fungi technology is eco-friendly and cost-effective and thus has emerged as a viable method for bioremediation in a wide range of synthetic dyes. Moreover, they do not require preconditioning to a particular pollutant, tolerate high concentrations of pollutants and nutrient limitation induces the production of extracellular ligninolytic enzymes with broad substrate specificity which includes several kinds of laccases, peroxidases, and oxidases producing H2O2. They utilize soluble as well as insoluble hazardous compounds as a nutrient source and convert them to simple fragmented forms. White rot fungi also remove the pollutants from the effluents by absorption, adsorption, and accumulation. Recently, many researchers have successfully exploited white rot fungi, for example, Pleurotus ostreatus, Phanerochaete chrysosporium, Tramates hirsute, T. versicolor, Lentinus edodes, and Trichoderma longibrachiatum for bioremediation of xenobiotics. The aim of this chapter is to address the present status and development of bioremediation strategies for synthetic textile dyes using white rot fungi and their enzymes.
