Antibiotic susceptibility and phylogenetic analyses for the origins and serotypes of Listeria monocytogenes strains isolated from ice cream and cream cakes

The present study involved characterization of variability of random amplified polymorphic DNA (RAPD) markers and protein patterns in some Pisum sativum cultivars. Twenty RAPD primers were used to estimate the genetic diversity in ten pea cultivars. A total of 89 bands were scored, of which 53 bands ( 59.55 %) were polymorphic and two of polymorphic band were unique bands. The average of amplified and polymorphic bands was 4.45 and 2.65 , respectively. The highest polymorphic bands found at locus C 10 , R 11 and V 03 ( 5 bands), whereas the lowest number of polymor phic bands found at locus OPA - 09 , OPA 13 , OPA - 07 , P 19 , N 7 and N 20 ( 1 band) . Distance matrix based on Jaccard coefficient ranged from 0.333 to 0.774 . The highest value was found between O - P 622 and O - P 234 , while the lowest genetic distance was observed betwe en O - P 923 and O - P 888 . A dendrogram revealed two major clusters: cluster I and cluster II. The results of dendrogram indicated that the groups of cluster II is most genetically distant from groups of cluster I. The SDS - PAGE patterns of seed storage proteins , showed that among 23 electrophoretic protein bands, only five bands were recorded to be polymorphic. The distance matrix values ranged from 0.00 to 0.75 . At distance 23 , cluster analysis sorting the cultivars into two major groups. The dendrogram constru cted from both methods divided pea cultivars into two clusters. In this study, analysis using RAPD markers provide more information about genetic diversity among pea cultivars and the results of SDS - PAGE show low variation among pea cultivars.

Fifteen pea (Pisum sativum L.) genotypes were evaluated for their agro-morphological performance and molecular diversity during 2017-2018 and 2018-2019 seasons. Combined analysis of variance revealed that there are highly significant differences among pea genotypes tested for all agro-morphological traits studied. The phenotypic coefficients of variation (PCV) values were relatively greater than Genotypic coefficients of variation (GCV) for all traits; however, GCV values were near to PCV values for the traits, like plant height, pods/plant, 100 seeds weight and pod width. High heritability coupled with high genetic advance was obtained for, plant height, reflecting the presence of additive gene action for the expression of this trait. High to moderate heritability coupled with moderate genetic gain were exhibited for number of pods per plant, 100 seeds weight, pod length, seeds per pod, pod weight and number of branches per plant. The Euclidean Distance among all genotypes based on agro-morphological traits was relatively wide. ISSR and SRAP markers were used to study the molecular diversity among pea genotypes. The molecular marker parameters revealed that SRAP markers were more efficient than ISSR markers with regards to polymorphism detection and in distinction among pea genotypes. Also, both ISSR and SRAP markers were able to amplify unique bands specific to a particular genotype.  A positive and significant correlation (r=0.389: p ≤ 0.001) between ISSR and SRAP matrix was observed according to Mantel's test. Also, significant correlation (r = 0.411: p ≤ 0.001) between the matrices of combined molecular markers data and agro-morphological data.

Genetic Relationships and Diversity among Pea (Pisum sativum L.) Genotypes Assessed using Agro-Morphological and Molecular Markers

The present study was conducted to identify polymorphism among eight notified varieties of pea (Azad Pea 1, Azad Pea 2, Azad Pea 3, Azad Pea 4, Azad Pea 5, Arkel, Lincoln & Bonneville) for identification of varieties and diversity studies. Twelve random decamer primers were used to amplify genomic DNA of the said 8 varieties of pea and the banding pattern of all genotypes against each primer were compared to check for polymorphism followed by cluster analysis using UPGMA method. Fourteen unique bands were recognized out of all the polymorphic bands. One unique band was obtained for Azad Pea 1 from OPD-18, 4 unique bands for Azad Pea 2 from OPA-10, OPU-16, OPE-16, A-07, one band for Azad Pea 4 from OPD-07, 2 unique bands for Arkel from OPU-16 and OPN-06, 4 unique bands for Bonneville using OPA-02, OPD-07, OPY-15, OPN-06 and 2 unique bands for Lincoln from OPC-02. Cluster analysis of pea genotypes based on molecular data showed that a significant genetic variation was present among the various pea genotypes used. Bonneville was most divergent of all the genotypes used .

Identification of Pea (Pisum Sativum L.) Varieties Using RAPD Markers

One of the best ways in FEC is Hamming code, which is a single bit error correction and double bit error detection method by using redundancy. In this paper, Hamming code is improved by appending the redundant bits at the end of data stream instead of positions in conventional Hamming code, to easily identify the parity bits in the data stream received at the receiver end. This method is appropriate for large data transmission with low redundancy bits per data bit ratio. In this paper proposed method achieve low delay circuit and optimize the load on receiver. Simulation is done using Xilinx ISE 14.7 Simulator.

Broccoli (Brassica oleracea var italica) is an edible green plant in the cabbage (Brassicaceae) family whose large, flowering head is eaten as a vegetable The present study on “Fingerprinting of broccoli (Brassica olereacea var italica ) varieties using RAPD markers” was conducted on  a total of 4 varieties of broccoli ie Palam Samradhi (PS) Early Green (EG), Palam Kanchan (Pk) and Palam Vachitar (PV) for varietal identification, using 16 random primers (RAPDs)  A total of 450 bands were produced and based on primer sequence and variation in specific, nineteen unique bands were recognised out of all the polymorphic bands Six unique bands were obtained for Palam Samridhi (PS) which could be used to differentiate it from other three varieties using primers OPY-15, OPA-02, OPC-14, OPQ-20 and OPU-03 Six unique bands were obtained for variety Early Green (EG) and could be differentiated from other three varieties using primers OPY-15, OPA-02, OPC-09, OPO-02 and OPU-17 Four unique bands were obtained for Palam Kanchan (PK) which could further be exploited to differentiate it from other three varieties using primers OPD-07, OPN-10, OP0-20 and OPC-18 Similarly for Palam Vachitra (PV) three unique bands were obtained which could be used to differentiate it from other three varieties using primers OPN-06, OPU-20 and OPN-10 Thus from the present investigation it can be concluded that, majority of the RAPD primers were able to discriminate among various varieties of broccoli, and therefore these could be used for identification of theses varieties using these primers These results clearly signify that RAPDs are good and easy enough to characterize the germplasm on molecular basis

Fingerprinting of Broccoli (Brassica olereacea var. italica.) Varieties Using RAPD Markers

BACKGROUND
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection started in Wuhan, China, and spread to the rest of the world to become a pandemic affecting over 385 million people throughout the world to date. Coronavirus disease 2019 (COVID-19) is primarily started as a respiratory tract infection. Recent studies indicate that it should be regarded as a systemic disease involving multiple systems including the hematopoietic system. Complete blood count and its parameters are important investigative tools in its prognosis. However, very few studies highlight the importance of peripheral blood cell morphology in this disease.


AIM
To study the hematological parameters (complete blood count and peripheral blood film) of COVID-19-positive patients and to compare the hematological parameters of those admitted in intensive care units (ICUs) with those admitted in non-ICUs of the hospitals.


MATERIALS AND METHODS
This retrospective study was carried out at a COVID-19 dedicated tertiary care center over a period of 3 months from July 2020 to September 2020. In our study, all 79 patients had complete blood counts performed at the time of admission. Complete blood count was repeated during the hospital stay for all severe cases. The data which provided information on the age and gender of each patient were obtained from the Laboratory Information System (LIS) of the hospital.


RESULTS
The mean age of our study group was 46.05 years. Out of 79 cases, lymphopenia was seen in 16.5% with five patients presenting with severe lymphopenia (<0.5 × 109 /L). All the patients that required ICU care presented with moderate to severe lymphopenia. The patients in the ICU setting showed significant neutrophilia (mean 14.16 × 109 /L) on follow-up complete blood count. Thrombocytopenia was observed in 35.3% of cases. It was observed that the mean neutrophil- lymphocyte ratio was higher in ICU admitted patients as compared to the non-ICU admitted patients. Among the ICU patients, 80% showed a neutrophil-lymphocyte ratio above the baseline cutoff (3.1). A wide array of morphological changes were observed in the peripheral blood smear including toxic-like granules in neutrophils, fetus-like C-shaped nucleus, lymphoplasmacytoid cells, bizarre cells, and apoptotic cells.


CONCLUSION
The study highlights that at the time of admission older age, decreased lymphocyte count, and raised neutrophil-lymphocyte ratio were closely associated with ICU admissions. Also, the morphological changes in peripheral blood film reveal atypical changes predominantly in the white blood cell (WBC) lineage.
