OBJECTIVE The aim of this study was to establish a suitable rabbit model and select the optimal protocol for laparotomy cryoablation of pancreatic carcinoma. METHODS VX2 tumor tissues were inoculated into rabbit pancreases to build the pancreatic carcinoma model; then, the tumor-bearing rabbits were randomly divided into 4 groups: control, treatment A (the cryoablated-iceball diameter was bigger than the tumor), treatment B (iceball was as big as the tumor), and treatment C (iceball was smaller than the tumor). Related laboratory tests were conducted, and survival time was recorded. RESULTS The VX2 pancreatic carcinoma model was successfully established, and serum neuron-specific enolase levels increased continuously after inoculation. Compared with controls, rabbits in treatments A and C groups had no significant survival benefit (P > 0.05), but treatment B significantly prolonged the survival time (P < 0.01). CONCLUSIONS VX2 pancreatic cancer model was successfully established with neuron-specific enolase as biomarker. Treatment B may be the optimal protocol for pancreatic carcinoma and a new treatment option for patients with unresectable pancreatic cancer.

Laparotomy Cryoablation in Rabbit VX2 Pancreatic Carcinoma.

BACKGROUND The aim of this study was to explore the influence of different input arteries on the parameters of computed tomography (CT) perfusion imaging for patients with different degree of stenosis of internal carotid artery (ICA). MATERIAL AND METHODS Forty patients were enrolled in the present study and divided into mild, moderate, severe stenosis and occlusion groups respectively with each 10 patients in each group. In reconstruction of cerebral CT perfusion (CTP) images, each raw perfusion image was reconstructed 3 times based on different reference input artery, including bilateral middle cerebral artery (MCA) and basilar arteries (BA). Region of interest (ROI) was drawn in the central territories of bilateral anterior cerebral artery, middle cerebral artery and posterior cerebral artery. And regional cerebral blood flow (rCBF) regional cerebral blood volume (rCBV), mean transit time (MTT), time to peak (TTP) and delay time (DT) were obtained from those ROI corresponding perfusion images. RESULTS In patients with mild and moderate ICA stenosis, there was no significant difference of perfusion parameters based on different input arteries (P>0.05). However, in severe ICA stenosis and occlusion CBF, MTT, and DT were significant different in affect side of the MCA group compared to the others (P<0.05). CONCLUSIONS Large intracranial artery can be selected as the input artery for patients with mild to moderate ICA stenosis, while for patients with severe stenosis and occlusion of ICA, the contra lateral middle cerebral artery or basilar artery would be better choice.

Computed Tomography Perfusion Imaging Quality Affected by Different Input Arteries in Patients of Internal Carotid Artery Stenosis.

Objective To investigate the role of dynamic contrast-enhanced computed tomography(DCE-CT) in the evaluation of tumor angiogenesis in patients with lung carcinoma. Methods 34 patients with pathology-proved pulmonary carcinoma underwent DCE-CT scan before operation. The functional imaging parameters: peak height(PH), the time of peak height (Tp), the ratio of PH of the mass to aorta(M/A), perfusion, the relative blood volume (rBV) and permeability (Pm) were calculated and correlated with the microvessel density (MVD) of the carcinoma. The functional imaging parameters MVD and the lymph node involvement between VEGF positive group and VEGF negative group were analyzed. Results The Perfusion, PH, M/A and rBV correlated positively with MVD (r= 0.76, 0.67, 0.70, 0.65 respectively;P0.0001), and no significant correlation was found between Pm and MVD (r= 0.30, P0.05). The functional imaging parameters and MVD of VEGF positive group were higher than those of VEGF negative group (P0.05)and the difference of lymph node involvement between two groups was significant(χ2= 9.409,P0.05). Conclusion DCE-CT functional imaging technique provides general information of lung carcinoma angiogenesis and alternates non-invasive option for evaluation of tumor angiogenesis in patients with lung carcinoma.

Dynamic contrast-enhanced computed tomography functional imaging for the evaluation of tumor angiogenesis in patients with lung carcinoma

Objectives: To explore the significance of the hemodynamic parameters of Computerized Tomography Perfusion Imaging (CTPI) under the deconvolution optimization algorithm for the diagnosis and treatment of patients with acute cerebral infarct (ACI).
Methods: A hundred and ten patients with ACI from December 2018 to September 2019 were selected for research, and CTPI was performed before and after Edaravone injection treatment. Then, the CTPI deconvolution algorithm based on the weighted adaptive (WA) total variation (TV) (WA-TV) optimization was constructed, which was compared with tensor total variation (TTV) and Motion-adaptive sparse parity (MASP). Brain Perfusion 4.0 was applied to obtain the relative time to peak (rTTP), the relative transit time of mean (rMTT), relative cerebral blood volume (rCBV), and relative cerebral blood flow (rCBF) of the core infarction area (CIA) and penumbra ischemic (PI).
Results: In four parameters of rTTP, rMTT, rCBV, and CBF, the peak signal to noise ratio (PSNR) of the WA-TV algorithm was higher than the MSAP and TTV algorithms, while the Mean Square Error (MSE) and Mean Absolute Error (MAE) were lower than MSAP and TTV algorithms (P 0.05).
Conclusion: Compared with TTV and MASP, the WA-TV algorithm performs better in noise reduction and artifact reduction. The CTPI parameters of rCBV, rCBF, rMTT, and rTTP are all important indications for the diagnosis of PI and ACI.

Adoption of computerized tomography perfusion imaging in the diagnosis of acute cerebral infarct under optimized deconvolution algorithm.

BACKGROUND Computed tomography perfusion imaging (CTPI) and perfusion-weighted imaging (PWI) are non-invasive technologies that can quantify tumor vascularity and blood flow. This study explored the blood flow information, tumor cell viability, and hydrothoraces in a rabbit pleural VX2-implanted model through use of CTPI, PWI, and DWI. MATERIAL AND METHODS A pleural VX2-implanted model was established in 58 New Zealand white rabbits. CTPI, PWI, and DWI were applied with a 16-slice spiral CT and an Archival 1.5 T dual-gradient MRI. RESULTS Compared with muscle tissue, PV, PEI, and BV of parietal and visceral pleural tumor implantation rabbits showed significant differences. The t values of PV, PEI, and BV between parietal and visceral pleura were 2.08, 2.29, and 2.88, respectively. Compared with muscle tissue, WIR, WOR, and MAXR of parietal and visceral pleural tumor implantation rabbits showed significant differences. In parietal pleural tumor implantation rabbits, the section surface of lesion tissues was 5.2±2.7 cm². Hydrothorax appeared 6.0±2.0 days after tumor implantation. The mean value of ADC was 1.5±0.6. In visceral pleural tumor implantation rabbits, the section surface of lesion tissues was 1.6±0.8 cm². Hydrothorax appeared 7.0±3.0 days after tumor implantation. The mean value of ADC was 1.4±0.5. The t values of the above 3 indices for the parietal and visceral pleura were 1.85, 1.83, and 1.76, respectively (P<0.05). CONCLUSIONS The combined application of CTPI, PWI, and DWI accurately and visually reflects the blood perfusion of tumor tissues and quantitatively analyzes blood flow information and the mechanism underlying hydrothorax generation in tumor tissues.

Tumor Growth Assessment by Computed Tomography Perfusion Imaging (CTPI), Perfusion-Weighted Imaging (PWI), and Diffusion-Weighted Imaging (DWI) in a Rabbit Pleural Squamous Cell Carcinoma VX2-Implanted Model.

Objective: To detect the activity of tumor cells and tumor blood flow before and after the radiotherapy of implanted pulmonary VX-2 carcinoma in rabbit models by using magnetic resonance diffusion-weighted imaging (MR-DWI) and magnetic resonance perfusion weighted imaging (MR-PWI), and to evaluate the effectiveness and safety of the radiotherapy based on the changes in the MR-DWI and MR-PWI parameters at different treatment stages. Methods: A total of 56 rabbit models with implanted pulmonary VX-2 carcinoma were established, and then equally divided into treatment group and control group. MR-DWI and MR-PWI were separately performed using a Philips Acheiva 1.5T MRI machine (Philips, Netherland). MRI image processing was performed using special perfusion software and the WORKSPACE advanced workstation for MRI. MRDWI was applied for the observation of tumor signals and the measurement of apparent diffusion coefficient (ADC) values; whereas MR-PWI was used for the measurement of wash in rate (WIR), wash out rate (WOR), and maximum enhancement rate (MER). The radiation treatment was performed using Siemens PRIMUS linear accelerator. In the treatment group, the radiotherapy was performed 21 days later on a once weekly dosage of 1,000 cGy to yield a total dosage of 5,000 cGy. Results: The ADC parameters in the region of interest on DWI were as follows: on the treatment day for the implanted pulmonary VX-2 carcinoma, the t values at the center and the edge of the lesions were 1.352 and 1.461 in the treatment group and control group (P>0.05). During weeks 0-1 after treatment, the t values at the center and the edge of the lesions were 1.336 and 1.137 (P>0.05). During weeks 1-2, the t values were 1.731 and 1.736 (P 0.05); in addition, the t values of the WIR, WOR, and MER at the edge of the lesions were 1.35, 1.07, and 1.51 (P>0.05). During weeks 0-1 after treatment, the t values of the WIR, WOR, and MER at the center of the lesions were 1.821, 1.856, and 1.931 (P<0.05); in addition, the t values of the WIR, WOR, and MER at the edge of the lesions were 1.799, 2.016, and 2.137 (P<0.05). During weeks 1-1 after treatment, the t values of the WIR, WOR, and MER at the center of the lesions were 2.574, 2.156, and 2.059 (P<0.05) and the t values of the WIR, WOR, and MER at the edge of the lesions were 1.869, 2.058, and 2.057 (P<0.05). During weeks 2-3 after treatment, the t values of the WIR, WOR, and MER at the center of the lesions were 2.461, 2.098, and 2.739 (P<0.05) and the t values of the WIR, WOR, and MER at the edge of the lesions were 2.951, 2.625, and 2.154 (P<0.05). During weeks 3-4 after treatment, the t values of the WIR, WOR, and MER at the center of the lesions were 2.584, 2.107, and 2.869 (P<0.05) and the t values of the WIR, WOR, and MER at the edge of the lesions were 2.057, 2.637, and 2.951 (P<0.05). During weeks 4-5 after treatment, the t values of the WIR, WOR, and MER at the center of the lesions were 2.894, 2.827, and 3.285 (P<0.05) and the t values of the WIR, WOR, and MER at the edge of the lesions were 3.45, 3.246, and 3.614 (P<0.05). After the radiotherapy (500 cGy), the tumors shrank on the T1WI, WIR, WOR, and MER; meanwhile, the PWI parameter gradually decreased and reached its minimum value. Conclusions: MR-DWI and MR-PWI can accurately and directly reflect the inactivation of tumor cells and the tumor hemodynamics in rabbit models with implanted pulmonary VX-2 carcinoma, and thus provide theoretical evidences for judging the clinical effectiveness of radiotherapy for the squamous cell carcinoma of the lung.
