Even in well-controlled laboratory environments, apparently identical repetitions of an experimental trial can give rise to highly variable perceptual outcomes and behavioral responses. This variability is generally discarded as a reflection of intrinsic noise in neuronal systems. However, part of this variability may be accounted for by trial-by-trial fluctuations of the phase of ongoing oscillations at the moment of stimulus presentation. For example, the phase of an EEG oscillation reflecting the rapid waxing and waning of sustained attention can predict the perception of a subsequent visual stimulus at threshold. Similar ongoing periodicities account for a portion of the trial-by-trial variability of visual reaction times. We review the available experimental evidence linking ongoing EEG phase to perceptual and attentional variability, and the corresponding methodology. We propose future tests of this relation, and discuss the theoretical implications for understanding the neuronal dynamics of sensory perception.

Stochastic Methods in Neuroscience

Synaptic noise due to intense network activity can have a significant impact on the electrophysiological properties of individual neurons. This is the case for the cerebral cortex, where ongoing activity leads to strong barrages of synaptic inputs, which act as the main source of synaptic noise affecting on neuronal dynamics. Here, we characterize the subthreshold behavior of neuronal models in which synaptic noise is represented by either additive or multiplicative noise, described by Ornstein-Uhlenbeck processes. We derive and solve the Fokker-Planck equation for this system, which describes the time evolution of the probability density function for the membrane potential. We obtain an analytic expression for the membrane potential distribution at steady state and compare this expression with the subthreshold activity obtained in Hodgkin-Huxley-type models with stochastic synaptic inputs. The differences between multiplicative and additive noise models suggest that multiplicative noise is adequate to describe the high-conductance states similar to in vivo conditions. Because the steady-state membrane potential distribution is easily obtained experimentally, this approach provides a possible method to estimate the mean and variance of synaptic conductances in real neurons.

Characterization of subthreshold voltage fluctuations in neuronal membranes

Synaptically generated subthreshold membrane potential (Vm) fluctuations can be characterized within the framework of stochastic calculus. It is possible to obtain analytic expressions for the steady-state Vm distribution, even in the case of conductance-based synaptic currents. However, as we show here, the analytic expressions obtained may substantially deviate from numerical solutions if the stochastic membrane equations are solved exclusively based on expectation values of differentials of the stochastic variables, hence neglecting the spectral properties of the underlying stochastic processes. We suggest a simple solution that corrects these deviations, leading to extended analytic expressions of the Vm distribution valid for a parameter regime that covers several orders of magnitude around physiologically realistic values. These extended expressions should enable finer characterization of the stochasticity of synaptic currents by analyzing experimentally recorded Vm distributions and may be applicable to other classes of stochastic processes as well.

An Extended Analytic Expression for the Membrane Potential Distribution of Conductance-Based Synaptic Noise

Neurological diseases can be studied by performing bio-hybrid experiments using a real-time biomimetic Spiking Neural Network (SNN) platform. The Hodgkin-Huxley model offers a set of equations including biophysical parameters which can serve as a base to represent different classes of neurons and affected cells. Also, connecting the artificial neurons to the biological cells would allow us to understand the effect of the SNN stimulation using different parameters on nerve cells. Thus, designing a real-time SNN could useful for the study of simulations of some part of the brain. Here, we present a different approach to optimize the Hodgkin-Huxley equations adapted for Field Programmable Gate Array (FPGA) implementation. The equations of the conductance have been unified to allow the use of same functions with different parameters for all ionic channels. The low resources and high-speed implementation also include features, such as synaptic noise using the Ornstein-Uhlenbeck process and different synapse receptors including AMPA, GABAa, GABAb, and NMDA receptors. The platform allows real-time modification of the neuron parameters and can output different cortical neuron families like Fast Spiking (FS), Regular Spiking (RS), Intrinsically Bursting (IB), and Low Threshold Spiking (LTS) neurons using a Digital to Analog Converter (DAC). Gaussian distribution of the synaptic noise highlights similarities with the biological noise. Also, cross-correlation between the implementation and the model shows strong correlations, and bifurcation analysis reproduces similar behavior compared to the original Hodgkin-Huxley model. The implementation of one core of calculation uses 3% of resources of the FPGA and computes in real-time 500 neurons with 25,000 synapses and synaptic noise which can be scaled up to 15,000 using all resources. This is the first step toward neuromorphic system which can be used for the simulation of bio-hybridization and for the study of neurological disorders or the advanced research on neuroprosthesis to regain lost function.
