Nauclea diderrichii, is a common woody plant in Africa where it is traditionally used for the relief and treatment of multiple pain-related conditions. The aim was to evaluate the inflammatory and anti-arthritic properties of the aqueous extract of N. diderrichii stem bark. Carrageenan and formalin were used to induce inflammation while zymosan A was used to induce monoarthritis in rats. The extract was given orally at doses of 100, 200 and 400 mg/kg. The extract significantly reduced carrageenan-induced edema up to 6 h, with 61.94% (6th hour). The extract induces a significant inhibition of the formalin-induced edema from the 2nd to the 8th hour with 61.50% (2nd hour). In continuous treatment, the activity of the extract remains significant from the 2nd to the 10th day of treatment with 63.85% (9th day). The extract (400 mg/kg) induces a significant decrease of the organ weight while the body weight increases significantly. At 400 mg/kg, the extract significantly increases the levels of red blood cells, hemoglobin and proteins, as well as the catalase and SOD activities, then a significant decrease of the levels of white blood cells, lymphocytes, hematocrit, MDA and NO. Furthermore, the extract significantly inhibited zymosan A-induced edema from the 1st to the 5th day with 60.99% (5th day). The joints of the treated animals show a close to normal architecture compared to that of the untreated animals. These results demonstrate the anti-inflammatory and anti-arthritic properties of N. diderrichii and confirm its traditional use to relieve pain and inflammation.

Anti-inflammatory and anti-arthritic properties of aqueous extract of Nauclea diderrichii (Rubiaceae) stem bark in rats

The current investigation deals with phytochemical screening and in vitro antiplasmodial activity of crude ethanol leaf extract and three fractions of crude ethanol leaf-extract of Nauclea diderrichii. Phytochemical test to screen bioactive compounds was carried out via standard protocols which uncovered the presence of alkaloids, saponins, steroids, phenols, tannins, flavonoids, glycosides and carbohydrates, extraction was done using absolute ethanol to afford the crude extract (Nd-ET) while maceration was done using solvents of different polarity gradient (petroleum ether, chloroform and ethyl acetate) to afford the remaining fractions (Nd-F1, Nd-F2 and Nd-F3). The antiplasmodial activity of the crude-extract and those of crude-extract-fractions against plasmodium falciparum unveil promising percentage elimination at all concentrations, with ethanol crude extract (Nd-ET) and ethyl acetate fraction (Nd-F3) having the highest, with 75.50% and 72.65% at 625µg/ml, and 87.83% and 86.33% at 5000µg/ml, respectively. These results clearly indicated that the active compounds present in the crude leaf extract/fractions of Nauclea diderrichii are highly potent eliminators of plasmodium falciparum and validate their popular usage in folk medicine in Gusau Local Government, Zamfara State, Nigeria, for the treatment of malaria.

In Vitro Phytochemistry and Antiplasmodial Activity of Leaf Extract and Fractions of Nauclea diderrichii

Background Diarrheal diseases are a major cause of morbidity and mortality throughout the world and particularly in developing countries. Nauclea diderrichii is a plant used in traditional medicine in the treatment of anemia, fever, gastric ulcer, malaria, abdominal pain, skin infections, and diarrhea. The present work is aimed at evaluating the antisecretory and spasmolytic activities of aqueous and ethanolic stem bark extracts of Nauclea diderrichii in Wistar rats. Methods The effect of aqueous and ethanolic extracts of Nauclea diderrichii was tested at doses of 100, 200, and 300 mg/kg on castor oil-induced secretory diarrhea, misoprostol-induced fluid accumulation, and the effect of pretreatment with yohimbine and glibenclamide. They were also tested on normal motility and castor oil- and carbachol-induced hypermotility. Results The results showed that the aqueous and ethanolic extracts of Nauclea diderrichii significantly (p < 0.001) inhibited castor oil-induced secretory diarrhea at all the doses. Both extracts significantly (p < 0.001) inhibit fluid accumulation induced by misoprostol. The pretreatment with glibenclamide reduced the antidiarrheal activity of aqueous extract of Nauclea diderrichii. The pretreatment with yohimbine did not alter the effect of the aqueous extract of Nauclea diderrichii. On intestine transit as on castor oil- and carbachol-induced motility, the aqueous and ethanolic extracts at doses of 100 and 200 mg/kg reduced significantly (p < 0.05, p < 0.01, and p < 0.001) the travelled distance by charcoal and peristaltic index. Conclusions The study demonstrated that the aqueous and ethanolic extracts of Nauclea diderrichii possess antisecretory and antispasmolytic properties hence its use in traditional medicine against diarrhea.

The present study investigates the phytochemicals and thin layer chromatographic profile of Nauclea diderrichii (Rubiaceae) leaf extracts. Phytochemical in the hexane, ethyl acetate and methanol extracts were determined using standard chemical tests. Thin layer chromatographic techniques were carried out using various solvent systems of varying polarity on these extracts. Preliminary phytochemical screening revealed the presence of alkaloids, glycosides, saponins, phenolic compounds, tannins, phytosterols, carbohydrates, flavonoids and terpenoids. Further screening using thin layer chromatographic technique on the N. diderrichii leaf extracts also revealed different phytochemical compounds with different Rf values. The results obtained in present study indicated that Nauclea diderrichii leaf is a rich source of phytochemicals. This could justifies the use of plant species in traditional medicine for treatment of various diseases.
