Psoraleae Fructus (Bu Gu Zhi) is the fruit of Psoralea corylifolia L. (PCL) and has been used for centuries in traditional Chinese medicine formulas to treat osteoporosis (OP). A new drug called "BX" has been developed from PCL, but its mechanism for treating OP is not yet fully understood.To explore the mechanism of action of BX in the treatment of ovariectomy-induced OP based function-oriented multi-omics analysis of gut microbiota (GM) and metabolites.C57BL/6 mice were bilaterally ovariectomized to replicate the OP model. The therapeutic efficacy of BX was evaluated by bone parameters (BMD, BV/TV, Tb.N, Tb.Sp), hematoxylin and eosin (H&E) staining results, and determination of bone formation markers procollagen type Ⅰ amino-terminal peptide (PⅠNP) and bone-specific alkaline phosphatase (BALP). Serum and fecal metabolomics and high-throughput 16S rDNA sequencing were performed to evaluate effects on endogenous metabolites and GM. In addition, an enzyme-based functional correlation algorithm (EBFC) algorithm was used to investigate functional correlations between GM and metabolites.BX improved OP in OVX mice by increasing BMD, BV/TV, serum PⅠNP, BALP, and improving Tb.N and Tb.Sp. A total of 59 differential metabolites were identified, and 9 metabolic pathways, including arachidonic acid metabolism, glycerophospholipid metabolism, purine metabolism, and tryptophan metabolism, were found to be involved in the progression of OP. EBFC analysis results revealed that the enzymes related to purine and tryptophan metabolism, which are from Lachnospiraceae_NK4A136_group, Blautia, Rs-E47_termite_group, UCG-009, and Clostridia_UCG-014, were identified as the intrinsic link between GM and metabolites.The regulation of GM and restoration of metabolic disorders may be the mechanisms of action of BX in alleviating OP. This research provides insights into the function-oriented mechanism discovery of traditional Chinese medicine in the treatment of OP. 

Function-oriented mechanism discovery of coumarins from Psoralea corylifolia L. in the treatment of ovariectomy-induced osteoporosis based on multi-omics analysis.

Irritable bowel syndrome (IBS) is a common clinical functional gastrointestinal disease. It has a complex pathophysiological mechanism, in which the imbalance of gut microbiota might play an important role. Lacidophilin tablets (LH) can regulate gut microbiota, but their effect on IBS is unknown. In this study, the IBS model was established by acetic acid enema combined with the constrained stress method, and rats were fed LH for 2 weeks. LH significantly reduced visceral sensitivity and intestinal propulsion rate and improved IBS-induced anxiety and depressive behavior in IBS rats. LH elevated the expression levels of mucin 2, claudin1, and occludin, and ameliorated IBS-induced structural damage to colonic tissues. The gut microbiota analysis revealed that LH altered the structure and composition of the gut microbiota in IBS rats. In addition, LH reduced the expression levels of inflammatory factor-related genes. These results suggest that LH could significantly improve the visceral sensitivity and intestinal motility disorders of IBS rats, relieve anxiety and depression levels, and alleviate the symptoms of IBS rats by regulating gut microbiota and reducing intestinal inflammation.

ETHNOPHARMACOLOGICAL RELEVANCE
Batri-7 (BT-7) is a compound herbal remedy rooted in traditional Mongolian medicine (TMM), composed of seven medicinal plants. Historically, it has been prescribed to treat various gastrointestinal conditions, including chronic enteritis and ulcerative colitis. However, despite this long-standing use in managing inflammatory disorders of the gut, the preventive effects of BT-7 on colitis-associated colorectal cancer (CAC), as well as its underlying molecular mechanisms, remains incompletely elucidated.


AIM OF THE STUDY
This study aimed to assess the therapeutic efficacy of BT-7 in an AOM/DSS-induced murine model of CAC, while also investigating its modulatory effects on systemic inflammation, intestinal barrier function, gut microbiota dynamics, and inflammation-related molecular signaling pathways.


MATERIALS AND METHODS
A chemically induced CAC model was established in C57BL/6 mice using azoxymethane (AOM) and dextran sodium sulfate (DSS). BT-7 was then administered orally at three dosage levels, and therapeutic outcomes were evaluated through a range of methods:clinical scoring, serum biochemical analysis, histological analysis, immunohistochemistry, and gut microbial profiling via 16S rRNA sequencing. To explore the underlying mechanistic pathways, with a particular focus on inflammasome regulation, transcriptome sequencing, quantitative PCR, and Western blotting were employed.


RESULTS
BT-7 treatment was well-tolerated, with no evidence of liver or kidney toxicity. Mice receiving BT-7 exhibited improved body weight maintenance, lower disease activity scores, and preserved colon length. Histologically, there was substantial restoration of mucosal architecture and reduced epithelial hyperplasia, alongside significant suppression of tumor formation. Systemically, BT-7 reduced levels of key pro-inflammatory cytokines (IL-1β, IL-6, TNF-α, and KC/GRO) and enhanced intestinal barrier integrity, as evidenced by the elevation of Occludin and ZO-1. Additionally, BT-7 decreased the abundance of pro-inflammatory taxa, such as Bacteroidota, and enriched beneficial genera like Lactobacillus and Dubosiella. The increased relative abundance of Lactobacillus_johnsonii and decreased Lactobacillus _murinus highlighted the probiotic effects of BT-7. Functional pathway prediction revealed that BT-7's microbiota was enriched in pathways related to carbohydrate and amino acid metabolism, as well as digestive system and immune-related functions. Transcriptomic analysis revealed downregulation of inflammatory signaling, particularly within the NOD-like receptor pathway, with reduced NLRP3 expression at both mRNA and protein levels confirming this effect.


CONCLUSIONS
This work demonstrates that BT-7 exerts protective effects against CAC by suppressing inflammation, improving epithelial integrity, and restoring microbial homeostasis. Mechanistically, the NLRP3 inflammasome is identified as BT-7's primary target in this context, which aligns with its traditional use in treating gastrointestinal disorders. Collectively, these findings highlight BT-7's therapeutic potential as a chemopreventive agent, supporting its utility in aiding inflammation-associated colorectal cancer treatment.

Linderae Radix (Lindera aggregata (Sims) Kosterm) is a traditional Chinese medicine known for its capability to regulate qi and relieve pain, particularly in the context of gastrointestinal disorders.While our previous research has demonstrated the efficacy of the Linderae Radix water extract (LRWE) in the treatment of diarrhea-predominant irritable bowel syndrome (IBS-D), the precise mechanisms remain elusive. This study aims to provide a comprehensive understanding of the therapeutic effects of LRWE on IBS-D through multi-omics techniques.16 S rRNA gene sequencing combined with LC-MS metabolomics was employed to investigate the effect of LRWE on the gut microbiota and metabolites of IBS-D rats. Spearman correlation analysis was performed on the gut microbiota and metabolites.LRWE administration significantly ameliorated IBS-D rats' symptoms, including diarrhea, visceral hypersensitivity, and low-grade intestinal inflammation. Gut microbiota analysis revealed that LRWE influenced the diversity of the gut microbiota in IBS-D rats by significantly reducing the relative abundance of Patescibacteria and Candidatus Saccharimonas, while increasing the relative abundance of Jeotgalicoccus. Serum metabolomic analysis identified 16 differential metabolites, associated with LRWE's positive effects on IBS-D symptoms, focusing on glyoxylate and dicarboxylic acid metabolism, and cysteine and methionine metabolism. Spearman analysis demonstrated a strong correlation between cecal microbiota composition and serum metabolite levels.This study elucidates that LRWE plays a crucial role in the comprehensive therapeutic approach to IBS-D by restoring the relative abundance of gut microbiota and addressing the disturbed metabolism of endogenous biomarkers. The identified bacteria and metabolites present potential therapeutic targets for IBS-D.

Psoralea corylifolia L. (P. corylifolia) has attracted increasing attention because of its potential hepatotoxicity. In this study, we used network analysis (toxic component and hepatotoxic target prediction, proteinprotein interaction, GO enrichment analysis, KEGG pathway analysis, and molecular docking) to predict the components and mechanism of P. corylifolia-induced hepatotoxicity and then selected 4-hydroxylonchocarpin and corylifol A for experimental verification. HepG2 cells were treated with low, medium, and high concentrations of 4-hydroxylonchocarpin or corylifol A. The activities of ALT, AST, and LDH in cell culture media and the MDA level, SOD activity, and GSH level in cell extracts were measured. Moreover, apoptosis, ROS levels, and mitochondrial membrane potential were evaluated. The results showed that the activities of ALT, AST, and LDH in the culture medium increased, and hepatocyte apoptosis increased. The level of MDA increased, and the activity of SOD and level of GSH decreased, and the ROS level increased with 4-hydroxylonchocarpin and corylifol A intervention. Furthermore, the mitochondrial membrane potential decreased in the 4-hydroxylonchocarpin and corylifol A groups. This study suggests that 4-hydroxylonchocarpin and corylifol A cause hepatocyte injury and apoptosis by inducing oxidative stress and mitochondrial dysfunction, suggesting that these compounds may be the potential hepatotoxic components of P. corylifolia.
