Predictive correlates of response to the anti-PD-L1 antibody MPDL3280A in cancer patients
Roy S. Herbst,Jean-Charles Soria,Marcin Kowanetz,Gregg Fine,Omid Hamid,Michael S. Gordon,J. A. Sosman,David F. McDermott,John D. Powderly,Scott N. Gettinger,Holbrook E Kohrt,Leora Horn,Donald P. Lawrence,Sandra Rost,Maya Leabman,Yuanyuan Xiao,Ahmad Mokatrin,Hartmut Koeppen,Priti S. Hegde,Ira Mellman,Daniel S. Chen,F. Stephen Hodi +21 more
TL;DR: Evaluated data suggest that MPDL3280A is most effective in patients in which pre-existing immunity is suppressed by PD-L1, and is re-invigorated on antibody treatment, as well as across multiple cancer types.

TGFβ attenuates tumour response to PD-L1 blockade by contributing to exclusion of T cells.
Sanjeev Mariathasan,Shannon J. Turley,Dorothee Nickles,Alessandra Castiglioni,Kobe C. Yuen,Yulei Wang,Edward E. Kadel,Hartmut Koeppen,Jillian L. Hegde,Richard Bourgon,Thomas Powles +41 more
TL;DR: Tumours from a large cohort of patients with metastatic urothelial cancer who were treated with an anti-PD-L1 agent were examined and major determinants of clinical outcome were identified and suggested that TGFβ shapes the tumour microenvironment to restrain anti-tumour immunity by restricting T-cell infiltration.
Atezolizumab in patients with locally advanced and metastatic urothelial carcinoma who have progressed following treatment with platinum-based chemotherapy: A single-arm, multicentre, phase 2 trial

TL;DR: Treatment with atezolizumab resulted in a significantly improved RECIST v1.1 response rate, compared with a historical control overall response rate of 10%, and Exploratory analyses showed The Cancer Genome Atlas (TCGA) subtypes and mutation load to be independently predictive for response to atezolediazepine.

MPDL3280A (anti-PD-L1) treatment leads to clinical activity in metastatic bladder cancer
Thomas Powles,Joseph Paul Eder,Gregg Fine,Fadi Braiteh,Yohann Loriot,Cristina Cruz,Joaquim Bellmunt,Howard A. Burris,Daniel P. Petrylak,Siew-leng Melinda Teng,Xiaodong Shen,Zachary Boyd,Priti S. Hegde,Daniel S. Chen,Nicholas J. Vogelzang +14 more
TL;DR: It is demonstrated that tumours expressing PD-L1-positive tumour-infiltrating immune cells had particularly high response rates, and patients with UBC, who are often older and have a higher incidence of renal impairment, may be better able to tolerate MPDL3280A versus chemotherapy.
