Light Antennas in phototactic algae.

The control of flagellar activity in the biflagellate green alga, Chlamydomonas reinhardtii was investigated by the in vitro reactivation of the isolated flagellar apparatus (the 2 flagella attached to their respective basal bodies plus accessory structures). The waveform and beat frequency of the isolated apparatus in the presence of 1 mM adenosine triphophate (ATP) were comparable to those recorded for living cells. Equimolar concentrations of adenosine diphosphate (ADP) could be substituted for ATP with little change in beat frequency and no apparent change in waveform, suggesting that the latter is converted to ATP by axonemal adenylate kinase. No reactivation occurred in adenosine monophosphate (AMP). But frequencies in cytidine, guanosine and uridine triphosphates (CTP, GTP and UTP) were approximately 10% that obtained in ATP. Reactivation was optimal over a broad pH range between pH 6.4 and pH 8.9 in both APT and ADP. Isolated flagellar apparatus could be induced to change from forward to reverse motion in vitro by manipulation of exogenous calcium ions. The 2 types of motion were directly comparable to recorded responses of living cells. Forward swimming occurred at levels of calcium below 10(-6)M, the isolated apparatus changing to backward motion above this level. Motility was inhibited at concentrations above 10(-3)M. The threshold for reversal of motion by calcium was lowered to 10(-7)M when the flagellar membranes were solubilized with detergent, indicating that the flagellar membranes are involved in the regulaion of the level of calcium within the axoneme. The reversal of motion by calcium was itself freely reversible. The relationship of these observations to the known tactic responses of Chlamydomonas is discussed.

Phototaxis in the broadest sense means positive or negative displacement along a light gradient or vector. Prokaryotes most often use a biased random walk strategy, employing type I sensory rhodopsin photoreceptors and two-component signalling to regulate flagellar reversal. This strategy only allows phototaxis along steep light gradients, as found in microbial mats or sediments. Some filamentous cyanobacteria evolved the ability to steer towards a light vector. Even these cyanobacteria, however, can only navigate in two dimensions, gliding on a surface. In contrast, eukaryotes evolved the capacity to follow a light vector in three dimensions in open water. This strategy requires a polarized organism with a stable form, helical swimming with cilia and a shading or focusing body adjacent to a light sensor to allow for discrimination of light direction. Such arrangement and the ability of three-dimensional phototactic navigation evolved at least eight times independently in eukaryotes. The origin of three-dimensional phototaxis often followed a transition from a benthic to a pelagic lifestyle and the acquisition of chloroplasts either via primary or secondary endosymbiosis. Based on our understanding of the mechanism of phototaxis in single-celled eukaryotes and animal larvae, it is possible to define a series of elementary evolutionary steps, each of potential selective advantage, which can lead to pelagic phototactic navigation. We can conclude that it is relatively easy to evolve phototaxis once cell polarity, ciliary swimming and a stable cell shape are present.

Evolution of phototaxis

The ad hoc Committee on Behavioral Tcrminology convened at the Conference on Sensory Transduction in Microorganisms in Santa Barbara, California, in January 1976, has considered recommendations for uniform naming of stimulus-induced movement responses of microorganisms (bacteria, motile algae, protozoa, sperm cells, etc.). We wish to emphasize that the terminology recommended by us is not meant to be used throughout every scientific publication, and also that it is not necessarily restricted to microorganisms. We are mostly concerned with usage in titles, because of the importance of title-based literature search procedures. Once an unambiguous term has been introduced, an abbreviation or othcr simplification will often be the preferred means of identifying the phenomenon under study, except in legends of figures. The following terminology, based on the original work by Pfeffer (1904), Kuehn (1919) and Fraenkel and Gunn (1 960), is recommended, and can be modified or extended when novel types of behavior are discovered.

Terminology of behavioral responses of motile microorganisms

Many phytoplankton and Zooplankton species display a daily pattern of vertical movement, generally termed diurnal vertical migration (DVM). The photoresponses of these organisms become functionally significant during such migrations, since light is agreed to be the dominant environmental factor controlling DVM. Light underwater, however, is a complex environmental stimulus, various aspects of which can participate in initiating, controlling, and/or orienting DVM.

— Young sporangiophores of the fungus, Pilobolus kleinii, respond to unilateral illumination by bending or by growing toward light of wavelengths between 312 and 530 mμ, with peaks of sensitivity near 360 and 450 mμ. Young sporangiophores exhibit a negative phototropic response to wavelengths shorter than 300 mμ, with a strong negative response at 280 mμ. Since the action spectrum did not correspond to the absorption spectrum of the pigmented zone as measured in vivo, and since colorless sporangiophores formed on media containing diphenylamine were capable of phototropic response, it is unlikely that the conspicuous orange-yellow pigment in young sporangiophores is the photoreceptor for phototropism. The results of probing with small beams of light and the behavior of sporangiophores submerged in mineral oil, together with measurements of the refractive index of the tip and base indicate that the photosensitive region is located in the tip of the young sporangiophore.

Two techniques have been used to study the quantitative relationship between stimulus intensity and oriented phototactic response (topotaxis) in Chlamydomonas. The net response of a cell population was monitored photometrically and was recorded continuously against time. The responses of individual cells were observed through a microscope and their swimming tracks were recorded on film.



The net response of the population is positive at low stimulus intensity and negative at high intensity. The direction of response can be reversed within two seconds by raising or lowering the intensity. The intensity-response curve for phototaxis is similar to the dose-response curve for phototropism. The net response has no distinct threshold; it increases linearly with log intensity; then it decreases and finally becomes negative.



The individual-cell studies reveal that the intensity-dependent increase in net topotactic response is due primarily to an increase in the number of cells responding and in the directness of their swimming path. As stimulus intensity is raised, the swimming path becomes increasingly well-aligned with the stimulus beam, whether net response is positive throughout the intensity range tested, negative throughout that range, or changing from positive to negative.



Changes in swimming rate do not contribute significantly to the intensity-dependent changes in net response. Swimming rate shows virtually no change throughout the intensity range of positive topotaxis and shows only a small increase in the negative range. However, a transient decrease in swimming rate (stop response) is often observed at the onset of stimulation. The implications of these results for the orientation mechanism are discussed.
