Iron is the most abundant mineral in the human body and plays essential roles in sustaining life, such as the transport of oxygen to systemic organs. The Fenton reaction is the reaction between iron and hydrogen peroxide, generating hydroxyl radical, which is highly reactive and highly toxic to living cells. “Ferroptosis”, a programmed cell death in which the Fenton reaction is closely involved, has recently received much attention. Furthermore, various applications of the Fenton reaction have been reported in the medical and nutritional fields, such as cancer treatment or sterilization. Here, this review summarizes the recent growing interest in the usefulness of iron and its biological relevance through basic and practical information of the Fenton reaction and recent reports.

Synergistic photothermal therapy (PTT) with gene therapy (GT) has drawn emerging interest in the improvement of cancer therapeutic efficiency, while the co-delivery of photothermal agents (PTAs) and therapeutic genes by an integrated nanoplatform, with controllability and biodegradability, is still challenging and urgently desired. Herein, a multi-functional metal–organic framework (MOF) based PTT–GT platform (siRNA@PT-ZIF-8) was developed, which was constructed with siRNA, a near-infrared (NIR) responsive organic dye IR780 derivative (IR780-1), and 2-methylimidazole (2-MIM) by a facile one-pot self-assembly method. This “all-in-one” system of siRNA@PT-ZIF-8 enabled not only photothermal/photoacoustic/fluorescence multimodal imaging but also tumor microenvironment responsiveness for specific and on-demand release of therapeutic cargos, overcoming the inherent limitations of free gene or organic PTA molecules (e.g., short blood circulation half-life and weak stability) in conventional PTT and GT. This nanoplatform provides an efficient and safe strategy for cancer theranostics, and the one-step assembly strategy favors personalized formulation design for diverse demands in cancer management.

Cancer is a complex and multifaceted disease that continues to be a global health challenge. It exerts a tremendous burden on individuals, families, healthcare systems, and society as a whole. To mitigate the impact of cancer, concerted efforts and collaboration on a global scale are essential. This includes strengthening preventive measures, promoting early detection, and advancing effective treatment strategies. In the field of cancer treatment, researchers and clinicians are constantly seeking new approaches and technologies to improve therapeutic outcomes and minimize adverse effects. One promising avenue of investigation is the utilization of the Fenton reaction, a chemical process that involves the generation of highly reactive hydroxyl radicals (·OH) through the interaction of hydrogen peroxide (H2O2) with ferrous ions (Fe2+). The generated ·OH radicals possess strong oxidative properties, which can lead to the selective destruction of cancer cells. In recent years, researchers have successfully introduced the Fenton reaction into the cancer microenvironment through the application of nanotechnology, such as polymer nanoparticles and light-responsive nanoparticles. This article reviews the progress of the application of the Fenton reaction, catalyzed by polymer nanoparticles and light-responsive nanoparticles, in the cancer microenvironment, as well as the potential applications and future development directions of the Fenton reaction in the field of tumor treatment.

Single-site bimetallic nanosheet for imaging guided mutually-reinforced photothermal-chemodynamic therapy

Cisplatin (CDDP)-based chemotherapy is substantially limited in the clinic due to its high postoperative recurrence rate. Synergy therapy has been proven as a potent approach to minimize recurrence and achieve enhanced treatment effects. Herein, chemotherapy drug CDDP is assembled with the photothermal-Fenton agent of bovine serum albumin (BSA) stabilized gallic acid-functionalized iron nanoparticles (GA-Fe NPs) to achieve chemo/chemodynamic synergistic cascade oncotherapy. The Pt-GA-Fe NPs can be utilized to generate H2O2 via the activation of nicotinamide adenine dinucleotide phosphate (NADPH) oxidases (NOXs) in the tumor microenvironment (TME), which would then greatly boost H2O2-depending chemodynamic therapy (CDT). The generated cytotoxic reactive oxygen species (hydroxyl radicals, ·OH) and the depletion of glutathione (GSH) would further promote CDDP-induced DNA damage. Moreover, benefiting from the absorption in the near-infrared (NIR) region, Pt-GA-Fe NPs exhibit excellent photothermal conversion efficiency (η = 45.5%) and allow photoacoustic imaging (PAI) guided photothermal therapy (PTT). In vitro and in vivo experiments show that synergy therapy can effectively kill cancer cells and successfully cure cancer without systemic toxicity. The work highlights a new type of therapeutic agent based on CDDP with the ability of H2O2 self-generation, thermal responsiveness, and enhanced CDT effects for applications in cancer therapy.
