Genomic hallmarks of cellular dormancy in cancer and therapeutic implications
Maria Secrier,Anna Julia Wiecek,Stephen Cutty,Daniel Kornai,Mario Parreno-Centeno,Lucie Gourmet,Guidantonio Malagoli Tagliazucchi,Daniel Hadar Jacobson,Ping Zhang,Lingyun Xiong,Gareth Bond,Alexis R. Barr +11 more
TL;DR: In this paper, the authors developed a methodology to uniquely identify dormancy from transcriptomic signals and characterised its prevalence and genomic constraints in solid primary tumours, showing that dormancy preferentially emerges in the context of more stable, less mutated genomes which maintain TP53 integrity and lack the hallmarks of DNA damage repair deficiency, while presenting increased APOBEC mutagenesis.
An efficient and adaptable workflow for editing disease-relevant single nucleotide variants using CRISPR/Cas9
Inga Usher,Lorena Ligammari,Sara Ahrabi,Emily Hepburn,Calum Connolly,Gareth Bond,Adrienne M. Flanagan,Adrienne M. Flanagan,Lucia Cottone +8 more
TL;DR: In this article, a scalable and adaptable workflow for single nucleotide editing that incorporates contemporary techniques including Illumina MiSeq sequencing, TaqMan qPCR and digital droplet PCR was described.
A Genetic Model for Central Chondrosarcoma Evolution Correlates with Patient Outcome
William Cross,Iben Lyskjaer,Tom Lesluyes,Steven Hargreaves,Anna-Christina Strobl,Christopher Davies,Christopher Davies,Sara Waise,Sara Waise,Shadi Hames,Dahmane Oukrif,Hongtao Ye,Fernanda Amary,Roberto Tirabosco,Toby Baker,David Barnes,Christopher D. Steele,Ludmil B. Alexandrov,Gareth Bond,Paul Cool,Paul Cool,Nischalan Pillay,Peter Van Loo,Adrienne M. Flanagan,Adrienne M. Flanagan +24 more
TL;DR: In this paper, the presence and absence of mutations in IDH1, and IDH2 are defining events, and TERT mutations have been recently been associated with poor outcome.
