Laboratory methods for work with plant and soil nematodes

Oomycetes have colonised all continents and oceans in a great variety of habitats and are arguably one of the most successful eukaryotic lineages. This is contrasted by the limited knowledge available for this group in various fields in comparison to other ubiquitous eukaryotes, such as unikont fungi, animals or plants. In this review an overview is given on the evolution and diversification of the oomycetes, with focus on the plant parasitic lineages and aspects of wild pathosystems.

Plant-parasitic nematodes as a potential threat to protected agriculture: Current status and management options

Root-knot nematodes (Meloidogyne spp.) are important pests of many cultivated plants. Recently, the most efficient chemical control products (e.g. methyl bromide) have now been restricted due to their toxic characteristics. Research on agents that work against root-knot nematodes and do not have a detrimental impact on the environment is becoming increasingly important. Advances in the last decades produced quite a number of biocontrol products that are already marketed. Some of the well-accepted commercial products contain bacteria Bacillus firmus and Pasteuria penetrans, and fungus Purpureocillium lilacinus. In this review we summarize the antagonistic activity of bacteria and fungi, with their advantages and limitations in biocontrol of root-knot nematodes.

Biological Control of Root-Knot Nematodes (Meloidogyne spp.): Microbes against the Pests

Food security is a global challenge that aligns with several Sustainable Development Goals (SDGs), including SDG 1 - “No Poverty”, SDG 2 - “Zero Hunger,” SDG 3 - “Good Health and Well-being,” SDG 13 - “Climate Action,” and SDG 15 - “Life on Land.”. To effectively address this issue, a convergence of agriculture and technology is crucial, incorporating precision agriculture, sustainable agriculture, bio-economy and advanced technologies such as machineries, Artificial intelligence-meachine learning and geospatial technology. Recent trends in food security worldwide have witnessed the adoption of technological advancements. However, it is important to consider biodiversity when implementing and adopting technological advancements. The integration of technology in agricultural practices aim to reduce chemical usage in farms, while increased production remains the main objective of smart farming and Precision Agriculture. By minimizing the use or implementing localized approaches for chemicals on farms, we can preserve soil-faunal diversity, which is at risk along with targets due to excessive applications of pesticides, herbicides, and insecticides. Geospatial technology plays a significant role by leveraging remotely sensed images acquired through satellites, aerial platforms, and GPS-tagged drones to enhance agricultural practices, improve crop growth and increase crop yields. Through geospatial technology, we can map and monitor soil and crop conditions, and effectively conserve, protect, and manage biodiversity on a global scale. The applications of remote sensing technology consider ecological and environmental parameters, soil factors, crop conditions, and plant-soil diversity to optimize yields and agricultural productivity. Additionally, the evaluation of various agricultural practices such as sustainable agriculture, bio-economy, using technology can contribute to increased yields, profitability, and biodiversity conservation as central components of these practices. This study emphasizes that achieving the SDG objectives is attainable through agriculture, which has direct and indirect connections to all SDGs. Geospatial technology plays a vital role in supporting the objectives at spatial and temporal scales by facilitating agricultural practices, ensuring food security, preserving biodiversity, monitoring soil and crop conditions, and promoting sustainable development. 

Highlighting the role of agriculture and geospatial technology in food security and sustainable development goals

Meloidogyne ethiopica is a tropical root-knot nematode species which has recently been found in Europe. We examined its ability to survive in open fields located in regions with sub-Mediterranean and continental European climates. The outdoor microplot experiment consisted of two locations and lasted three growing and two winter seasons. It was demonstrated that M. ethiopica was able to survive at both locations and also that it retained its infection ability although temperatures below zero were recorded. The correct species was confirmed after each winter season by isozyme electrophoresis. Furthermore, the influence of temperature on the reproduction cycle of M. ethiopica was investigated. Meloidogyne ethiopica required 67, 48 and 36 days to complete the reproduction cycle at mean daily temperatures of 18.3, 22.7 and 26.3°C, respectively. At 13.9°C, M. ethiopica was not able to reproduce. The data obtained from these experiments were used to develop a correlation between temperature and the time needed for M. ethiopica to complete a reproduction cycle using a mathematical equation. Furthermore, eight vegetable crops that are important for agricultural production in Slovenia were tested for their suitability as hosts for M. ethiopica.

Effect of Slovenian climatic conditions on the development and survival of the root-knot nematode Meloidogyne ethiopica

The dagger nematode, Xiphinema rivesi Dalmasso, a member of the X. americanum group, was detected in 2002 for the first time in Slovenia and for the fourth time in Europe (4). X. rivesi is a vector of at least four North American nepoviruses including Cherry rasp leaf virus (CRLV), Tobacco ringspot virus (TRSV), Tomato ringspot virus (ToRSV), and Peach rosette mosaic virus (PRMV) (1,2). All of these viruses are included on the EPPO and EU lists of quarantine organisms, but none of the Xiphinema species found in Europe have been reported to transmit these nepoviruses. Three virus isolates, including TRSV (from Lobelia spp.; virus collection of the Plant Protection Service, Wageningen, The Netherlands), ToRSV (grapevine isolate PV-0381; DSMZ, Braunschweig, Germany), and Arabis mosaic virus (ArMV) (from Vinca spp.; virus collection of the Plant Protection Service), were used in transmission tests with a population of X. rivesi found in Slovenia. X. rivesi is not known to transmit ArMV and this virus was included as a check. The nematodes were extracted from peach orchard soil collected near the village of Dornberk, and transmission tests fulfilled the set of criteria proposed by Trudgill et al. (3). Cucumis sativus cv. Eva, grown in a growth chamber at 25°C, was used as acquisition hosts and transmission bait plants. The acquisition hosts were mechanically inoculated and showing systemic symptoms before the introduction of nematodes. Noninoculated acquisition plants were included as controls. After a 10-day acquisition feeding period, the nematodes were transferred to healthy bait plants and allowed a 14-day inoculation feeding period. X. rivesi transmitted TRSV and ToRSV but not ArMV. TRSV and ToRSV bait plants developed systemic symptoms 4 to 6 weeks after the nematodes were transferred. Transmission of TRSV and ToRSV was confirmed by testing leaf and root sap of bait plants in a double antibody sandwich (DAS)-ELISA. High virus concentrations were detected in the roots and leaves of TRSV and ToRSV symptomatic plants. DAS-ELISA on bait plants from nematodes that had been allowed to feed on ArMV-infected or the virus-free control acquisition plants gave negative results. No symptoms appeared on bait plants used for ArMV transmission or the control bait plants. Xiphinema rivesi from Slovania Transmit Tobacco ringspot virus and Tomato ringspot virus to Cucumber Bait Plants

Root-knot nematodes (Meloidogyne spp.) are common pathogens that parasitize vegetables and other crops and cause significant yield reductions worldwide. In the early spring of 2006, severe plant stunting, chlorosis, and extensive root galling were observed on cucumber plants grown in a greenhouse on Zeta plain, Zetska ravnica, Montenegro. In the summer and autumn of 2006, infected roots of different crops were collected from greenhouses and vegetable production fields of Zeta plain, which represents the largest area of Montenegro's vegetable production. Several vegetable crops were found to be infected with root-knot nematodes, including tomato (Lycopersicon esculentum Mill.), squash (Cucurbita pepo L.), cucumber (Cucumis sativus L.), pepper (Capsicum annuum L.), and lettuce (Lactuca sativa L.).
