Omics approaches for understanding heavy metal responses and tolerance in plants

Phosphorus homeostasis: acquisition, sensing, and long-distance signaling in plants

Lead is a very toxic and futile heavy metal for rice plants because of its injurious effects on plant growth and metabolic processes. Polyploidy or whole genome doubling increases the ability of plants to withstand biotic and abiotic stress. Considering the beneficial effects of nanoparticles and tetraploid rice, this research was conducted to examine the effectiveness of tetraploid and titanium dioxide nanoparticles (TiO2 NPs) in mitigating the toxic effects of lead. A diploid (E22-2x) and it's tetraploid (T-42) rice line were treated with Pb (200 μM) and TiO2 NPs (15 mg L−1). Lead toxicity dramatically reduced shoot length (16 % and 4 %) and root length (17 % and 9 %), biological yield (55 % and 36 %), and photosynthetic activity, as evidenced by lower levels of chlorophyll a and b (30 % and 9 %) in E-22 and T-42 rice cultivars compared to the control rice plants, respectively. Furthermore, lead toxicity amplified the levels of reactive oxygen species (ROS), such as malondialdehyde and H2O2, while decreasing activities of all antioxidant enzymes, such as superoxidase, peroxidase, and glutathione predominately in the diploid cultivar. Transmission electron microscopy and semi-thin section observations revealed that Pb-treated cells in E22-2x had more cell abnormalities than T-42, such as irregularly shaped mitochondria, cell wall, and reduced root cell size. Polyploidy and TiO2 reduced Pb uptake in rice cultivars and expression levels of metal transporter genes such as OsHMA9 and OsNRAMP5. According to the findings, genome doubling alleviates Pb toxicity by reducing Pb accumulation, ROS, and cell damage. Tetraploid rice can withstand the toxic effect of Pb better than diploid rice, and TiO2 NPs can alleviate the toxic impact of Pb. Our study findings act as a roadmap for future research endeavours, directing the focus toward risk management and assessing long-term impacts to balance environmental sustainability and agricultural growth.

Transcriptome analysis reveals candidate genes involved in multiple heavy metal tolerance in hyperaccumulator Sedum alfredii.

BACKGROUND: Trees of Bombax ceiba L. could produce a large number of viable seeds in the dry-hot valleys. However, the seedling regeneration of the species is difficult in these areas as mild drought often occur repeatedly which might be followed by heat stress. However, how the repeated drought affects the subsequent drought and heat tolerance of B. ceiba is not clear. In this study, chlorophyll fluorescence, soluble sugar content and lipid metabolism were measured for the drought-treated seedlings and heat-treated seedlings with or without drought hardening. RESULTS: Neither the first nor third dehydration treatments affected the photosynthetic activity and soluble sugar content of B. ceiba seedlings. However, they differentially affected the fluidity of the local membranes and the levels of diacylglycerol and phosphatidic acid. Heat shock severely decreased the photosynthetic efficiency but drought priming reduced the effects of heat shock. Moreover, heat shock with or without drought priming had differential effects on the metabolism of soluble sugars and some lipids. In addition, the unsaturation level of membrane glycerolipids increased following heat shock for non-drought-hardened seedlings which, however, maintained for drought-hardened seedlings. CONCLUSIONS: The results suggest that two cycles of dehydration/recovery can affect the metabolism of some lipids during the third drought stress and may enhance the heat tolerance of B. ceiba by adjusting lipid composition and membrane fluidity.

Effects of repeated drought stress on the physiological characteristics and lipid metabolism of Bombax ceiba L. during subsequent drought and heat stresses.

Phosphorus transporter (PHT) genes encode H2PO4−/H+ co-transporters that absorb and transport inorganic nutrient elements required for plant development and growth and protect plants from heavy metal stress. However, little is known about the roles of PHTs in Brassica compared to Arabidopsis thaliana. In this study, we identified and extensively analyzed 336 PHTs from three diploid (B. rapa, B. oleracea, and B. nigra) and two allotetraploid (B. juncea and B. napus) Brassica species. We categorized the PHTs into five phylogenetic clusters (PHT1–PHT5), including 201 PHT1 homologs, 15 PHT2 homologs, 40 PHT3 homologs, 54 PHT4 homologs, and 26 PHT5 homologs, which are unevenly distributed on the corresponding chromosomes of the five Brassica species. All PHT family genes from Brassica are more closely related to Arabidopsis PHTs in the same vs. other clusters, suggesting they are highly conserved and have similar functions. Duplication and synteny analysis revealed that segmental and tandem duplications led to the expansion of the PHT gene family during the process of polyploidization and that members of this family have undergone purifying selection during evolution based on Ka/Ks values. Finally, we explored the expression profiles of BnaPHT family genes in specific tissues, at various developmental stages, and under heavy metal stress via RNA-seq analysis and qRT-PCR. BnaPHTs that were induced by heavy metal treatment might mediate the response of rapeseed to this important stress. This study represents the first genome-wide analysis of PHT family genes in Brassica species. Our findings improve our understanding of PHT family genes and provide a basis for further studies of BnaPHTs in plant tolerance to heavy metal stress.

Genome-Wide Analysis of Phosphorus Transporter Genes in Brassica and Their Roles in Heavy Metal Stress Tolerance.

Diacylglycerol kinases (DGKs) are signaling enzymes that play pivotal roles in response to abiotic and biotic stresses by phosphorylating diacylglycerol (DAG) to form phosphatidic acid (PA). However, no comprehensive analysis of the DGK gene family had previously been reported in B. napus and its diploid progenitors (B. rapa and B. oleracea). In present study, we identified 21, 10, and 11 DGK genes from B. napus, B. rapa, and B. oleracea, respectively, which all contained conserved catalytic domain and were further divided into three clusters. Molecular evolutionary analysis showed that speciation and whole-genome triplication (WGT) was critical for the divergence of duplicated DGK genes. RNA-seq transcriptome data revealed that, with the exception of BnaDGK4 and BnaDGK6, BnaDGK genes have divergent expression patterns in most tissues. Furthermore, some DGK genes were upregulated or downregulated in response to hormone treatment and metal ion (arsenic and cadmium) stress. Quantitative real-time PCR analysis revealed that different BnaDGK genes contribute to seed oil content. Together, our results indicate that DGK genes have diverse roles in plant growth and development, hormone response, and metal ion stress, and in determining seed oil content, and lay a foundation for further elucidating the roles of DGKs in Brassica species.

Genome-wide identification and comparative analysis of diacylglycerol kinase (DGK) gene family and their expression profiling in Brassica napus under abiotic stress

The AINTEGUMENTA-like (AIL) proteins, which belong to the AP2 family, play important roles in regulating the growth and development of plant organs. The AIL family has not yet been comprehensively studied in rapeseed (Brassica napus), an allotetraploid and model organism for the study of polyploid evolution. In the present study, 99 AIL family genes were identified and characterized from B. rapa, B. oleracea, B. napus, B. juncea, and B. nigra using a comprehensive genome-wide study, including analyses of phylogeny, gene structure, chromosomal localization, and expression pattern. Using a phylogenetic analysis, the AIL genes were divided into eight groups, which were closely related to the eight AtAIL genes, and which shared highly conserved structural features within the same subfamily. The non-synonymous/synonymous substitution ratios of the paralogs and orthologs were less than 1, suggesting that the AIL genes mainly experienced purifying selection during evolution. In addition, the RNA sequencing data and qRT-PCR analysis revealed that the B. napus AIL genes exhibited organ- and developmental stage-specific expression patterns. Certain genes were highly expressed in the developing seeds (BnaAIL1, BnaAIL2, BnaAIL5, and BnaAIL6), the roots (BnaANT, BnaAIL5, and BnaAIL6), and the stem (BnaAIL7B). Our results provide valuable information for further functional analysis of the AIL family in B. napus and related Brassica species.
