In addition to the often-cited advantages of using Arabidopsis thaliana as a model system in plant biological research (1), Arabidopsis has many additional characteristics that make it an attractive experimental organism for studying lea d (Pb) accumulation and tolerance in plants. These include its fortuitous familial relationship to many known metal hyperaccumulators (Brassicaceae), as well as similar Pbaccumulation patterns to most other plants. Using nutrient-agar plates, hydroponic culture, and Pb-contaminated soils as growth media, we found significant variation in Arabidopsis thaliana ecotypes in accumulation and tolerance of Pb. In addition, we have found that Pb accumulation is not obligatorily linked with Pb tolerance, suggesti ng that different genetic factors control these two processes. We also screened ethyl methanesulfonate-mutagenized M2 populations and identified several Pb-accumulating mutants. Current characterization of these mutants indicates that their phenotypes are likely due to alteration of general metal ion uptake or translocation processes since these mutants also accumulate many other metals in shoots. We expect that further characterization of the ecotypes and mutants will shed light on the basic genetic and physiological underpinnings of plant-based Pb remediation. 7. Bacteria and plants might be used to enhance remediation of soil pollutants in situ. This study assessed the potential of bacteria (12 isolates), plants (16 forage grasses), and plant-bacteria associations (selected pairings) to remediate 2-chlorobenzoic acid (2CBA)-contaminated soil. Initially, grass viability was assessed in 2CBA-contaminated soil. Soil was contaminated wi th 2CBA, forage grasses were grown under growth chamber conditions for 42 or 60 d, and the 2CBA concentration in soil was determined by gas chromatography. Only five of 16 forage grasses grew in 2CBA-treated (816 mg/kg) soil. Growth of Bromus inermis had no effect on 2CBA concentration, whereas Agropyron intermedium, B. biebersteinii, A. riparum, and Elymus dauricus decreased 2CBA relative to nonplanted control soil by 32 to 42%. This study addresses the often-competing goals of organic fertility and weed control by evaluating alternative orchard floor management strategies for their impact on N cycling, tree performance, and soil biological activity in a newly established apple (Malus domestica Borkh.) orchard. The standard tillage weed control practice resulted in satisfactory tree growth with desirable levels of leaf N and most other nutrients; however, soil biological activity did not improve. Maintenance of a living cover understory increased soil N concentration and availability and improved soil biological activity; however, tree growth was less than in other treatments likely in response to competition with the living cover understory for space and water. Application of wood chip mulch resulted in exceptional tree growth which may have resulted from greater water availability, but available soil N was lower, and consequently, tree leaf N concentration was low; in addition, soil biological activity was not improved. Clove oil organic herbicide provided poor weed control resulting in lower leaf N and tree growth and did not improve soil biological activity. Brassicaceae seed meal applications enhanced N availability and soil nematode abundance, but leaf N and many other nutrients were below desirable levels, and additional research is needed to optimize this treatment. We conclude that meeting the multiple objectives of weed control, optimal tree health, and increased soil biological activity may require employment of different orchard floor management strategies at different times during the life of the orchard.

Orchard floor management effects on nitrogen fertility and soil biological activity in a newly established organic apple orchard

Several mineral nutrients can influence fruit quality and disorders of apple. Among these, nitrogen (N), potassium (K), phosphorous (P), calcium (Ca), and boron (B) are most often correlated to apple fruit quality and disorders. Leaf mineral analysis is a useful tool to diagnosis apple tree deficiencies but often is poorly related to fruit quality. Using fruit analysis alone or in combination with leaf analysis often permits more precise prediction of fruit quality. Over the last several years, we have developed several models for predicting apple fruit quality. In addition, we have examined the effects of various orchard factors and cultural practices, such as irrigation, rootstocks, and fertigation and foliar application of nutritional sprays, on apple fruit mineral composition and quality. A ranking of major minerals has been developed that predicts fruit quality within a year and between years. Increasing fruit N is inversely related to fruit yellow or red colour and positively associated with fruit respiration and ethylene. Fruit Ca tends to be imprecisely related to bitter pit and fruit firmness. Potassium fertigation in four apple cultivars increased fruit size, yield, acidity, and colour, but decreased firmness at harvest. Multiple sprays of soluble Ca often reduce bitter pit and usually but not always increase Ca concentrations in subdermal cortical tissue. Early-season Ca sprays often are more effective than later sprays at reducing bitter pit; however, later applications of Ca have a greater influence on fruit Ca concentration. The B concentration of apple fruit is much more strongly affected by early season B sprays that are B in leaves. Fruit from B-sprayed trees may exhibit quality loss due to B excess even though leaf B appears normal. Water stress reduced leaf and fruit K but increased leaf Mg. An overview of several orchard factors on mineral nutrition and fruit quality and disorders will be presented.

Response of Soil and Irrigated Fruit Trees to Fertigation or Broadcast Application of Nitrogen, Phosphorus, and Potassium

The severity of leaf chlorosis in iron (Fe)‐deficient fruit trees is often characterized using a semi‐quantitative visual rating index that is subject to evaluator bias. Analytical instruments are now available that provide a quantitative measure of leaf green color that could substitute for visual ratings. We injected limbs of mature chlorotic pear trees (Pyrus communis L. cv. Bartlett) with distilled water or a solution of 0.1% Fe (w/v) as FeSO4‐7H2O on 17 July 1995. Treatments were replicated eight‐fold. On 18 August 1995, a Minolta SPAD‐502 chlorophyll meter was used to measure the green color of 30 randomly sampled leaves located above the point of injection on each injected limb. Average leaf green color was higher in the Fe‐injected tree than in the water‐injected tree of each experimental block.
