Targeting STAT5 Expression Resulted in Molecular Response Improvement in Patients with Chronic Phase CML Treated with Imatinib
Philippe Rousselot,Lydia Roy,Gabriel Etienne,Laurence Legros,Aude Charbonnier,Valérie Coiteux,Pascale Cony-Makhoul,Francois-Xavier Mahon,Joelle Guilhot,Jean-Michel Cayuela,Francis Relouzat,Philippe Leboulche,Stéphane Prost +12 more
TL;DR: It is demonstrated that pioglitazone at pharmacological doses inhibited cell growth of the Bcr-Abl positive cell line K562 through the activation of the PPAR-g/STAT5 pathway and induced a clonogenic defect in CD34+ cells from CML patients.

Erosion Of The Chronic Myeloid Leukemia Stem Cell Pool By PPARγ Agonists
Francis Relouzat,Marc Spenchian,Gerald Massonnet,Jean-Paul Beressi,Els Verhoeyen,Benjamin Manéglier,Sylvie Castaigne,Christine Chomienne,Stany Chretien,Philippe Rousselot,Philippe Leboulch +10 more
TL;DR: It is shown that the residual CML LSC pool can be gradually purged by the glitazones, antidiabetic drugs that are agonists of peroxisome proliferator-activated receptor gamma (PPARg) that decreases expression of STAT5 and its downstream targets HIF2α and CITED2, which are key guardians of C ML LSC's quiescence and stemness.
