Next-generation CRISPR/Cas9 transcriptional activation in Drosophila using flySAM
Yu Jia,Rong-Gang Xu,Xingjie Ren,Ben Ewen-Campen,Rajendhran Rajakumar,Jonathan Zirin,Donghui Yang-Zhou,Ruibao Zhu,Fang Wang,Decai Mao,Ping Peng,Huan-Huan Qiao,Xia Wang,Lu-Ping Liu,Lu-Ping Liu,Bowen Xu,Jun-Yuan Ji,Qingfei Liu,Jin Sun,Norbert Perrimon,Norbert Perrimon,Jian-Quan Ni +21 more
TL;DR: FlySAM greatly improves on existing in vivo CRISPRa techniques in terms of potency, scalability, and ease of use, and provides a simple and general method for conducting overexpression experiments and screens.

An efficient and multiple target transgenic RNAi technique with low toxicity in Drosophila.
Huan-Huan Qiao,Fang Wang,Rong-Gang Xu,Jin Sun,Ruibao Zhu,Decai Mao,Xingjie Ren,Xia Wang,Yu Jia,Ping Peng,Da Shen,Lu-Ping Liu,Zhijie Chang,Guirong Wang,Shao Li,Jun-Yuan Ji,Qingfei Liu,Jian-Quan Ni,Jian-Quan Ni +18 more
TL;DR: A next generation transgenic RNAi system with reduced leakiness of expression and simultaneous knockdown is developed and versatility is increased which can simultaneously knock down multiple genes in one step.
Varp interacts with Rab38 and functions as its potential effector.
Fang Wang,Haiwei Zhang,Xinjun Zhang,Yinyin Wang,Fangli Ren,Xiaoning Zhang,Yonggong Zhai,Zhijie Chang +7 more
TL;DR: This study demonstrates that Varp physically interacts with Rab38, and preferentially binds to the active GTP-bound form of Rab38 both in vitro and in vivo, and proposes Varp serves as both an effector and a GEF by interacting with different Rabs in mammalian cells.

Histone H1 defect in escort cells triggers germline tumor in Drosophila ovary
Zhihao Yang,Jin Sun,Yuzhao Hu,Fang Wang,Xia Wang,Huan-Huan Qiao,Jiang Xu,Decai Mao,Xingjie Ren,Li-Xia Pan,Rong-Gang Xu,Bowen Xu,Yifan Zhang,Haiyi Li,Wei Miao,Yanhui Hu,Zhijie Chang,Dong Wang,Haitao Li,Zai Chang,Lu-Ping Liu,Qingfei Liu,Jian-Quan Ni +22 more
TL;DR: It is found that histone H1 depletion in escort cells (ECs) resulted in an increase of spectrosome-containing cells (SCCs), an ovary tumor-like phenotype, and double knockdown of H1 and dpp in ECs can reduce the number of SCCs to the normal level, indicating that the additional Dpp secreted by ECs contributes to the germline tumor.

Perspectives on gene expression regulation techniques in Drosophila.
Rong-Gang Xu,Xia Wang,Da Shen,Jin Sun,Huan-Huan Qiao,Fang Wang,Lu-Ping Liu,Jian-Quan Ni,Jian-Quan Ni +8 more
TL;DR: The main purpose of this review is to provide the fly research community with an updated summary of newly developed gene expression regulation techniques and help the community to select appropriate methods and optimize the research strategy.
