Recently a novel DNA virus (Torque Teno Virus (TTV) has been identified in Japan and shown to be associated with elevated aminotransferase s levels after transfusion. However the exact role of TTV in pathogenesis of liver disease is yet to be established. The purpose of this study was to determine the prevalence of TTV in thalassemic patients and its relationship with elevated alanine-aminotransfrase (ALT) and aspartateaminotransfrase (AST). This cross-sectional analysis study was conducted on 452 thallasemic patients. Serums were collected from all of the patients, first ALT and AST levels were determined. Then, after DNA extraction, TTV DNA was amplified and detected using semi-nested PCR, followed by gel electrophoresis. Demographic characteristics and clinical data were collected from each participant for statistical analysis. The findings showed that 160 of 452 (35.4%) samples had TTV DNA detected by PCR. From 160 TTV DNA positive, 98 (61.20%) were female and 62 (38.80%) of them were male (P=0.549). The mean ALT and AST values in TTV positive group were higher than in TTV negative group, and the difference was statistically significant (p<0.0001). The result showed that the prevalence of TTV in thalassemic patients in Jahrom is less than other studies in Iran and the mean ALT and AST values in TTV positive individuals were about 2 times more than in TTV negative individuals.

Influence of negative energy balance on cyclicity and fertility in the high producing dairy cow.

Regulation of ovarian activity is an integrated process encompassing both extraovarian signals and intrafollicular factors. Initiation of primordial follicle growth and the early stages of folliculogenesis can occur without gonadotropins, but FSH may affect the rate of preantral follicle growth. Antral follicle development from 1 to 4 mm in sheep and cattle is completely gonadotropin dependent. These recruited follicles express a range of mRNA encoding steroidogenic enzymes, gonadotropin receptors, and local regulatory factors and their receptors. As follicles continue to mature, there is a transfer of dependency from FSH to LH, which may be part of the mechanism involved in selection of follicles for continued growth. Locally produced growth factors, such as the IGF and members of the transforming growth factor-beta superfamily, work in concert with gonadotropins throughout the follicular growth continuum and can have significant effects on follicle selection. Environmental influences, such as changes in nutrition, also have an effect on ovarian activity. This can occur without significant variation in circulating gonadotropin concentrations and can be correlated with changes in circulating concentrations of metabolic hormones, including insulin, IGF-I, GH, and leptin. Nutrition can also affect the expression of mRNA encoding components of the ovarian IGF system to regulate the sensitivity/response of follicles toward gonadotropins. Hence, the roles of growth factors in follicular development and survival depend on gonadotropin status and differentiation state of the follicle. In conclusion, it is the integration of these extraovarian signals and intrafollicular factors that determine whether a follicle will continue to develop or be diverted into atretic pathways.

Invited review : New Perspectives on the Roles of Nutrition and Metabolic Priorities in the Subfertility of High-Producing Dairy Cows

This review addresses the suggestion that the decline in dairy reproductive performance, as increasingly observed these days, may be due to a hampered process of metabolic adaptation in early lactating cows. In our opinion, adaptation to the negative energy balance is a gradual process. Because almost all cows do adapt in the long run, it is not possible to classify animals as adapted or non-adapted. The use of risk factors is more appropriate in this case and is discussed in this review. Among them are the body condition score and its derivatives, feed intake, the calculated negative energy balance, and metabolic parameters like the plasma concentration of insulin or the triacylglycerol content in the liver. Moreover, factors that play a role in the link between declined reproductive performance and the metabolic situation of the cow during the early lactating period are discussed. Among these are insulin, insulin-like growth factors, leptin, neuropeptide Y, non-esterified fatty acids, thyroid hormones, urea, and ammonia.

In vitro blastocyst production was determined for oocytes recovered postmortem from 48 beef x dairy heifers offered low (Low NH3) or high (High NH3) plasma ammonia-generating diets during the period of late antral follicle development. Following the establishment of a reference estrus (d 0), the experimental diets were offered for an 18-d period starting on d 3 and during which a second estrus was induced (d 16) 4 d before the animals were slaughtered. Blood samples collected at varying intervals were analyzed for ammonia, urea, progesterone, and LH. Ovarian folliculogenesis was monitored daily by transrectal ultrasonography. Ovaries were collected at slaughter and cumulus-oocyte complexes were aspirated from small (1 to 4 mm) and medium-sized (> 4 to 8 mm) sized follicles. In vitro-matured and -fertilized putative d-1 zygotes were cultured for a further 7 d in vitro and embryo development and metabolism were assessed. Relative to the low-NH3-generating diet, the high-NH3-generating diet increased peak postprandial levels of plasma ammonia (326.1 +/- 43.3 vs 52.1 +/- 7.4 micromol/L; P < .001), mean levels of plasma urea (7.0 vs 5.7 mmol/L; SED = .2; P < .001), peak levels of plasma progesterone prior to induced luteolysis (8.9 +/- .4 vs 6.8 +/- .3 microg/L; P < .001), and follicular fluid levels of ammonia (267 +/- 18 vs 205 +/- 20 nmol/mL; P < .05) and progesterone (351 +/- 69 vs 199 +/- 26 ng/mL; P < .05). The timing and level of the preovulatory LH surge was not affected by dietary treatment. Of oocytes cultured, cleavage (47.4 vs 62.4%; P = .02) and blastocyst production (10.9 vs 20.6%; P = .06) rates were reduced when the oocytes were derived from heifers offered the high- rather than the low-NH3-generating diets. There were interactions between dietary treatment and follicle size class, which indicated that fewer blastocysts were produced from cleaved oocytes derived from medium-sized follicles of heifers offered the high-NH3 treatment but that de novo protein synthesis was increased in such embryos. In conclusion, exposure to high levels of ammonia and(or) urea in vivo can significantly compromise the subsequent capacity of oocytes to develop to blastocysts in vitro, and oocytes recovered from medium-sized follicles are particularly sensitive to this effect.
