International Zinc Nutrition Consultative Group (IZiNCG) technical document #1. Assessment of the risk of zinc deficiency in populations and options for its control.

The Biomarkers of Nutrition for Development (BOND) project is designed to provide evidence-based advice to anyone with an interest in the role of nutrition in health. Specifically, the BOND program provides state-of-the-art information and service with regard to selection, use, and interpretation of biomarkers of nutrient exposure, status, function, and effect. To accomplish this objective, expert panels are recruited to evaluate the literature and to draft comprehensive reports on the current state of the art with regard to specific nutrient biology and available biomarkers for assessing nutrients in body tissues at the individual and population level. Phase I of the BOND project includes the evaluation of biomarkers for 6 nutrients: iodine, iron, zinc, folate, vitamin A, and vitamin B-12. This review represents the second in the series of reviews and covers all relevant aspects of folate biology and biomarkers. The article is organized to provide the reader with a full appreciation of folate's history as a public health issue, its biology, and an overview of available biomarkers (serum folate, RBC folate, and plasma homocysteine concentrations) and their interpretation across a range of clinical and population-based uses. The article also includes a list of priority research needs for advancing the area of folate biomarkers related to nutritional health status and development.

Assessment of the risk of deficiency in populations and options for its control

A molecular understanding of high-density lipoprotein (HDL) will allow a more complete grasp of its interactions with key plasma remodelling factors and with cell-surface proteins that mediate HDL assembly and clearance. However, these particles are notoriously heterogeneous in terms of almost every physical, chemical and biological property. Furthermore, HDL particles have not lent themselves to high-resolution structural study through mainstream techniques like nuclear magnetic resonance and X-ray crystallography; investigators have therefore had to use a series of lower resolution methods to derive a general structural understanding of these enigmatic particles. This chapter reviews current knowledge of the composition, structure and heterogeneity of human plasma HDL. The multifaceted composition of the HDL proteome, the multiple major protein isoforms involving translational and posttranslational modifications, the rapidly expanding knowledge of the HDL lipidome, the highly complex world of HDL subclasses and putative models of HDL particle structure are extensively discussed. A brief history of structural studies of both plasma-derived and recombinant forms of HDL is presented with a focus on detailed structural models that have been derived from a range of techniques spanning mass spectrometry to molecular dynamics.

Despite the noteworthy genotoxic potency of many industrial wastewaters, the genotoxic hazard posed to the downstream ecosystem and its associated biota will be determined by genotoxic loading. Municipal wastewaters, although ranking low in potency, can achieve loading values that are several orders of magnitude greater than those of most industries. Although these wastewaters are generally mixtures of wastes from several different sources, the volumetric proportion of the daily discharge that is of industrial origin rarely exceeds 30%. Genotoxicity calculations for the Montreal Urban Community (MUC) municipal wastewater treatment facility indicate that over 90% of the genotoxic loading (31.1 kg benzo(a)pyrene equivalents per day) is nonindustrial in origin. Moreover, a mass balance of surface water genotoxicity for St. Lawrence river at Montreal indicates that over 85% of the total contributions from the Montreal region are nonindustrial in origin. Additional calculations for the Great Lakes, and other rivers throughout the world, provide further support of a strong relationship between surface water genotoxicity and population. Despite some information about physical/chemical properties, the identity of the putative genotoxins in municipal wastewaters and surface waters remains a mystery. Likely candidates include potent genotoxins, such as N-nitroso compounds and aromatic amines, known to be present in human sanitary wastes, as well as genotoxic PAHs known to be present in many municipal wastewaters. Calculations based on literature data indicate that human sanitary wastes may be able to account for a substantial fraction (4–70%) of the nonindustrial loading from municipal wastewaters. The genotoxic hazards of domestic wastes in surface waters.

Numerous studies have demonstrated that several diseases and stress conditions are associated with changes in the levels of zinc in the blood plasma and cellular elements. In this research the association between serum zinc concentrations and other hematic parameters of diagnostic interest has been evaluated. Quantitative determinations of zinc, total plasmatic proteins, albumin, hemoglobin and calculation of mean corpuscular volume were performed on blood samples from 58 males aged 20-61 years. Concentrations measured in our sample are comparable with reference values. Statistically significant correlation coefficients were found between age and albumin (r = -0.562, P < 0.001), serum zinc and albumin (r = 0.328, P < 0.05), serum zinc and hemoglobin (r = 0.291, P < 0.05), and total plasmatic proteins and albumin (r = 0.463, P < 0.001). These correlation coefficients were significant even after adjustment for age effect. The determination of serum zinc concentration may be useful in the assessment of clinical scenarios. Particularly, it may provide additional information for the diagnosis of specific pathologies, such as hepatic malfunctions. It could also be useful in the identification of different stages of anemia.

Zinc content of normal human serum and its correlation with some hematic parameters.

With the present study, we intend to verify the utility of hair as diagnostic tool for trace element analysis, to substitute, perhaps, conventional materials, such as blood serum. Blood and hair were collected from male individuals (n=107) aged 20–59 y. Determinations of Cu and Zn concentrations were performed with atomic absorption spectrometry. An influence of age on Zn in hair has been found, with significantly different values before and after age 30 y. There is no correlation between Cu concentrations in hair and in serum, and a positive one (r=0.3554,p<0.05) between Zn levels in hair and in serum. No association between Zn and Cu levels in hair has been demonstrated; on the contrary, these elements have a moderate positive correlation in serum (r=0.3586,p<0.01). The data indicate that hair may represent an additional analytical material for Cu and Zn to complement blood serum.

Electrophoretic analysis of non-human primates hair keratin

Keratins extracted from the hair of several mammals were characterized by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and isoelectric focusing (IEF). By SDS-PAGE, most differences between species were detected in the position and intensity of polypeptide bands with MW in the range 30-10 KDa (high sulphur keratins, hair matrix proteins, HMP). By IEF, differences were found in the intensity of polypeptide bands in the acidic pH range (< 5.1). Electrophoretic techniques could therefore have a taxonomic usefulness.
