Programmable mutually exclusive alternative splicing for generating RNA and protein diversity
Melina Mathur,Cameron M. Kim,Sarah A. Munro,Shireen S. Rudina,Eric M. Sawyer,Eric M. Sawyer,Christina D. Smolke +6 more
TL;DR: A modular and extensible platform for regulating four programmable exons that undergo a mutually exclusive alternative splicing event to generate multiple functionally-distinct proteins and can ultimately be applied to program modular proteins and increase coding capacity of synthetic biological systems.
Tunable Transcriptional Interference at the Endogenous Alcohol Dehydrogenase Gene Locus in Drosophila melanogaster.
Victoria Jorgensen,Jingxun Chen,Helen Vander Wende,Devon E Harris,Alicia McCarthy,Shane Breznak,Siu Wah Wong-Deyrup,Yuzhang Chen,Prashanth Rangan,Gloria A. Brar,Eric M. Sawyer,Leon Y. Chan,Elçin Ünal +12 more
TL;DR: The endogenous Adh locus is developmentally regulated by transcriptional interference in a tunable manner and upregulation of the distal Adh transcript led to the enrichment of histone 3 lysine 36 trimethylation over the Adh proximal promoter.

Bacterial Logic Devices Reveal Unexpected Behavior of Frameshift Suppressor tRNAs
Todd T. Eckdahl,Eric M. Sawyer,Cody Barta,Romina Clemente,Michel Conn,Clif Davis,Catherine Doyle,Mary Gearing,Olivia Ho-Shing,Alyndria Mooney,Jerrad Morton,Shamita Punjabi,Ashley Schnoor,Siya Sun,Shashank Suresh,Bryce Szczepanik,D. Leland Taylor,Annie Temmink,William Vernon,A. Malcolm Campbell,Laurie J. Heyer,Jeffrey L. Poet +21 more
TL;DR: This study investigates frameshift suppressor tRNAs previously reported to use five-base anticodon-codon interactions and deduced a four-base binding hypothesis for the frameshIFT suppressionor tRNA interactions with mRNA as an alternative to the published five- base binding model.

Tunable Transcriptional Interference at the Endogenous Alcohol Dehydrogenase Gene Locus in Drosophila melanogaster
Victoria Jorgensen,Jingxun Chen,Helen Vander Wende,Devon E Harris,Siu Wah Wong-Deyrup,Yuzhang Chen,Prashanth Rangan,Gloria A. Brar,Eric M. Sawyer,Leon Y. Chan,Elçin Ünal +10 more
TL;DR: The CRISPR/Cas9 system is employed to edit the endogenous Adh locus and it is found that removal of the distal promoter does also result in the untimely expression of the proximal promoter-driven mRNA isoform in adults, albeit at lower levels than previously reported.

Developmental Regulation of an Organelle Tether Coordinates Mitochondrial Remodeling in Meiosis
TL;DR: This study defines a key mechanism that coordinates mitochondrial morphogenesis with the landmark events of meiosis and demonstrates that cells can developmentally regulate tethering to induce organelle remodeling.
