Role of the androgen receptor in breast cancer and preclinical analysis of enzalutamide.
Dawn R. Cochrane,Sebastian Bernales,Britta M. Jacobsen,Diana M. Cittelly,Erin N. Howe,Nicholas C. D'Amato,Nicole S. Spoelstra,Susan M. Edgerton,Annie Jean,Javier Sanchez Guerrero,Francisco Gómez,Satyanarayana Medicherla,Iván E. Alfaro,Emma McCullagh,Paul Jedlicka,Kathleen C. Torkko,Ann D. Thor,Anthony D. Elias,Andrew Asher Protter,Jennifer K. Richer +19 more
TL;DR: This preclinical study supports the initiation of clinical studies evaluating enzalutamide for treatment of AR+ tumors regardless of ER status, since it blocks both androgen- and estrogen- mediated tumor growth.
Endoplasmic reticulum stress-independent activation of unfolded protein response kinases by a small molecule ATP-mimic
Aaron S Mendez,Jennifer Alfaro,Marisol A Morales-Soto,Arvin C. Dar,Emma McCullagh,Katja Gotthardt,Han Li,Diego Acosta-Alvear,Carmela Sidrauski,Alexei Korennykh,Sebastian Bernales,Kevan M. Shokat,Peter Walter +12 more
TL;DR: It is proposed that IPA binding biases the PERK kinase towards its active conformation, which trans-activates apo-PERK molecules, which carries major implications for design and therapeutic application of kinase inhibitors.
Discovery of Pyrazolopyrimidine Derivatives as Novel Dual Inhibitors of BTK and PI3Kδ
Brahmam Pujala,Anil Agarwal,Sandip Middya,Monali Banerjee,Arjun Surya,Anjan Kumar Nayak,Ashu Gupta,Sweta Khare,Rambabu Guguloth,Nitin Atmaram Randive,Bharat Uttam Shinde,Anamika Thakur,Dhananjay I. Quinn,Roopa Rai,Son Minh Pham +26 more
TL;DR: A series of novel compounds that have low nanomolar potency against both BTK and PI3Kδ as well as acceptable PK properties that could be useful in the development of treatments against B-cell related diseases are reported.
Temozolomide analogs with improved brain/plasma ratios - Exploring the possibility of enhancing the therapeutic index of temozolomide.
Roopa Rai,Monali Banerjee,Darren H. Wong,Emma McCullagh,Ashu Gupta,Shailendra Tripathi,Eduardo Riquelme,Ramnivas Jangir,Shyamraj Yadav,Mohd. Raja,Pankaj Melkani,Vikas Dixit,Umesh K. Patil,Ritesh Shrivastava,Sandip Middya,Felipe Olivares,Javier Sanchez Guerrero,Arjun Surya,Son Minh Pham,Sebastian Bernales,Andrew A. Protter,David T. Hung,Sarvajit Chakravarty +22 more
TL;DR: These compounds are imidazotetrazine analogs, expected to act through the same mechanism as temozolomide, and have the potential to improve efficacy and therapeutic index in the treatment of glioblastoma.

Dual Inhibition of Bruton's Tyrosine Kinase and Phosphoinositide-3-Kinase p110δ as a Therapeutic Approach to Treat Non-Hodgkin's B Cell Malignancies.
Jennifer Alfaro,Felipe Pérez de Arce,Sebastian Belmar,Glenda Fuentealba,Patricio Avila,Gonzalo Ureta,Camila Flores,Claudia Acuña,Luz Delgado,Diana Gaete,Brahmam Pujala,Anup Barde,Anjan Kumar Nayak,T. V. R. Upendra,Dhananjay I. Patel,Shailender Chauhan,Vijay Kumar Sharma,Stacy Kanno,Ramona Almirez,David T. Hung,Sarvajit Chakravarty,Roopa Rai,Sebastian Bernales,Kevin P. Quinn,Son Minh Pham,Emma McCullagh +25 more
TL;DR: It is found that this orally bioavailable dual inhibitor reduced tumor growth in a B cell lymphoma xenograft model more effectively than either ibrutinib or idelalisib, suggesting that dual inhibition of these two key pathways by a single molecule could be a viable approach for treatment of NHL patients.
