Yeast Mph1 helicase dissociates Rad51-made D-loops: implications for crossover control in mitotic recombination
Rohit Prakash,Dominik Satory,Eloise Dray,Almas Papusha,Jürgen Scheller,Jürgen Scheller,Wilfried Kramer,Lumir Krejci,Hannah L. Klein,James E. Haber,Patrick Sung,Grzegorz Ira +11 more
TL;DR: It is shown that budding yeast Mph1, an ortholog of human FancM helicase, utilizes its helicase activity to suppress spontaneous unequal sister chromatid exchanges and DNA double-strand break-induced chromosome crossovers.
Enhancement of RAD51 recombinase activity by the tumor suppressor PALB2
Eloise Dray,Julia Etchin,Claudia Wiese,Dorina Saro,Gareth J. Williams,Michal Hammel,Xiong Yu,Vitold E. Galkin,Dongqing Liu,Miaw-Sheue Tsai,Shirley M.-H. Sy,Shirley M.-H. Sy,David Schild,Edward H. Egelman,Junjie Chen,Patrick Sung +15 more
TL;DR: This work documents a new function of PALB2: to enhance RAD51's ability to form the D loop, and demonstrates the multifaceted role of PALb2 in chromosome damage repair.
Targeting DNA Damage Response and Replication Stress in Pancreatic Cancer

TL;DR: Replication stress and DDR deficiency are independent of each other, creating opportunities for therapy in DDR-proficient PC and after platinum therapy.

Chromosome arm aneuploidies shape tumour evolution and drug response
Ankit Shukla,Thu H.M. Nguyen,Sarat B. Moka,Jonathan Ellis,John P. Grady,Harald Oey,Alexandre S. Cristino,Kum Kum Khanna,Dirk P. Kroese,Lutz Krause,Eloise Dray,J. Lynn Fink,Pascal H.G. Duijf,Pascal H.G. Duijf +13 more
TL;DR: Analysis of CAA profiles of 23,427 tumours identifies aspects of tumour evolution including probable orders in which CAAs occur and CAAs predicting tissue-specific metastasis and in predicting drug response, CAAs substantially outperform mutations and focal deletions/amplifications combined.

Sweasy,Patrick Sung,Sara Rockwell,Robert N. Nishimura,Robert N. Nishimura,Richard H. Weisbart,Peter M. Glazer +20 more
TL;DR: In this paper, a cell-penetrating lupus auto-antibody, 3E10, has been shown to preferentially bind DNA single-strand tails, inhibit key steps in DNA singlestrand and doublestrand break repair, and sensitiz cultured tumor cells and human tumor xenografts to DNA-damaging therapy, including doxorubicin and radiation.
