p38γ and p38δ reprogram liver metabolism by modulating neutrophil infiltration.
Bárbara González-Terán,Nuria Matesanz,Ivana Nikolic,M. Verdugo,M. Verdugo,Vinatha Sreeramkumar,Lourdes Hernández-Cosido,Alfonso Mora,Georgiana Crainiciuc,María Laura Saiz,Edgar Bernardo,Luis Leiva-Vega,Elena Rodríguez,Víctor Bondía,Jorge L Torres,Sonia Perez-Sieira,Luis Ortega,Ana Cuenda,Francisco Sánchez-Madrid,Rubén Nogueiras,Andrés Hidalgo,Miguel Marcos,Guadalupe Sabio +22 more
TL;DR: It is demonstrated that p38δ is elevated in livers of obese patients with NAFLD and that mice lacking p38γ/δ in myeloid cells are resistant to diet‐induced fatty liver, hepatic triglyceride accumulation and glucose intolerance, and neutrophil infiltration in wild‐type mice contributes to steatosis development by means of inflammation and liver metabolic changes.
MKK6 controls T3-mediated browning of white adipose tissue
Nuria Matesanz,Edgar Bernardo,Rebeca Acín-Pérez,Elisa Manieri,Elisa Manieri,Sonia Perez-Sieira,Lourdes Hernández-Cosido,Valle Montalvo-Romeral,Alfonso Mora,Elena Rodríguez,Luis Leiva-Vega,Ana Victoria Lechuga-Vieco,Jesús Ruiz-Cabello,Jesús Ruiz-Cabello,Jorge L Torres,Maria Crespo-Ruiz,Francisco Centeno,Clara V. Alvarez,Miguel Marcos,José Antonio Enríquez,Rubén Nogueiras,Guadalupe Sabio +21 more
TL;DR: The authors show that the stress activated kinase MKK6 acts as a repressor of UCP1 expression, suggesting that its inhibition promotes adipose tissue browning and increases organismal energy expenditure.

HNF1A recruits KDM6A to activate differentiated acinar cell programs that suppress pancreatic cancer
Mark Kalisz,Edgar Bernardo,Anthony Beucher,Miguel A. Maestro,Natalia del Pozo,Irene Millán,Lena Haeberle,Martin Schlensog,SA Safi,Wolfram T. Knoefel,Vanessa Grau,Matías de Vas,Karl B. Shpargel,Eva C. Vaquero,Terry Magnuson,Sagrario Ortega,Irene Esposito,Francisco X. Real,Jorge Ferrer +18 more
TL;DR: It is shown that pancreas‐specific Hnf1a null mutant transcriptomes phenocopy those of Kdm6a mutations, and both defects synergize with KrasG12D to cause PDAC with sarcomatoid features, providing direct genetic evidence that HNF1A deficiency promotes PDAC.

p38α blocks brown adipose tissue thermogenesis through p38δ inhibition.
Nuria Matesanz,Ivana Nikolic,Magdalena Leiva,Marta Pulgarín-Alfaro,Ayelén M Santamans,Edgar Bernardo,Alfonso Mora,Leticia Herrera-Melle,Elena Rodríguez,Daniel Beiroa,Ainoa Caballero,Elena Martín-García,Rebeca Acín-Pérez,Lourdes Hernández-Cosido,Luis Leiva-Vega,Jorge L Torres,Francisco Centeno,Angel R. Nebreda,José Antonio Enríquez,Rubén Nogueiras,Miguel Marcos,Guadalupe Sabio +21 more
TL;DR: A mechanism implicated in physiological BAT activation that has potential clinical implications for the treatment of obesity and related diseases such as diabetes is elucidated.
Pancreas agenesis mutations disrupt a lead enhancer controlling a developmental enhancer cluster
Irene Miguel-Escalada,Miguel A. Maestro,Diego Balboa,Anamaria Elek,Aina Bernal,Edgar Bernardo,Vanessa Grau,Javier García-Hurtado,Arnau Sebé-Pedrós,Jorge Ferrer +9 more
TL;DR: In this article , an epigenetic cascade that is required for duct and endocrine differentiation was shown to activate an entire PTF1A enhancer cluster in early pancreatic multipotent progenitors, which revealed an enhancer that acts as a regulatory master key and is thus vulnerable to pathogenic loss of function mutations.
