Vibrio infections are common among marine fish and lead to serious problems in the aquaculture sector. This study reports a recent occurrence of Vibrio species (spp.) isolated from cultured groupers in Peninsular Malaysia using the gyrB and pyrH genes. A total of 147 Vibrio strains were successfully isolated from 77 (64%) groupers using culture method and subjected to gyrB and pyrH sequencing for species identification and confirmation. Results showed that 89% of Vibrio strains were identified and clustered to six groups of Vibrio spp., while 11% were not clustered to any Vibrio spp. using the gyrB sequences. Meanwhile, by analysis of the pyrH sequences all the 147 Vibrio strains (100%) were successfully identified and clustered into 11 groups of Vibrio spp., including the gyrB non‐identified strains. The pyrH gene provides a better resolution for identification of Vibrio spp. compared with the gyrB gene. Thus, the pyrH gene was more suitable for a rapid determination of Vibrio spp. distribution in Peninsular Malaysia. Using the pyrH gene, our study found higher prevalence of Vibrio vulnificus (33%), V. alginolyticus (24%) and V. parahaemolyticus (22%), followed by V. rotiferianus (5%), V. harveyi (3%), V. tubiashii (2%), V. campbellii (2%), V. ponticus (1%), V. diabolicus (1%), V. owensii (1%) and others Vibrio sp. (7%). Thus, the results of this study revealed that the occurrence of pathogenic vibrios among grouper fish is still high in Malaysian aquaculture. In addition, the pyrH gene was proved as a suitable marker for rapid identification of Vibrio species compared with the gyrB gene. Machine-induced vibrations represent, for several reasons, a crucial design issue for industrial buildings. At the early design stage, special attention is thus required for the static and dynamic performance assessment of the load-bearing members, given that they should optimally withstand ordinary design loads but also potentially severe machinery operations. The knowledge and reliable description of the input vibration source is a key step, similarly to a reliable description of the structural system, to verify. However, such a kind of detailing is often unavailable and results in a series of simplified calculation assumptions. In this paper, a case-study eyewear factory built in 2019 is investigated. Its layout takes the form of a two-story, two-span (2 × 14.6 m) precast concrete frame (poor customer/designer communication on the final equipment resulted in various non-isolated computer numerical control (CNC) vertical machines mounted on the inter-story floor, that started to suffer from pronounced resonance issues. Following past experience, this paper investigates the validity of a coupled experimental–numerical method that could be used for efficient assessment predictive studies. Based on on-site experiments with Micro Electro-Mechanical Systems (MEMS) accelerometers mounted on the floor and on the machine (spindle included), the most unfavorable machine-induced vibration sources and operational conditions are first characterized. The experimental outcomes are thus used to derive a synthetized signal that is integrated in efficient one-bay finite element (FE) numerical model of the floor, in which the machine–structure interaction can be taken into account. The predictability of marked resonance issues is thus emphasized, with a focus on potential and possible limits of FE methods characterized by an increasing level of detailing and computational cost.

Efficiency of Coupled Experimental–Numerical Predictive Analyses for Inter-Story Floors Under Non-Isolated Machine-Induced Vibrations

Engines and motors on board marine vessels are often mounted on flexible base supports. When the levels of vibration transmitted from such machines to the base structure are intolerable, vibration reduction techniques have to be used. In instances where machines are mounted via very stiff mounts to maintain shaft alignment, the transmission paths of the vibratory force are through the mounts. In such cases, active force cancellation at the mount locations using opposing controlled forces can be used. This paper discusses the use of frequency domain techniques to reduce the vibration with the H ∞ norm as a performance measure. The model of the system, identified using frequency response method is used in the design of the H ∞ controllers for active force cancellation. Two approaches were presented. Experimental results show a reduction of between 15 ~ 65% of the measured transmitted force at different mount locations when the machine is operated at a speed of about 1320 rpm. This frequency matches that of the machine-base system principal mode. Where the system response is not significant, reduction in transmitted force level is minimal. This limitation is the result of the control goal used in the design.
